Background: The design and initial experience with the e-learning course “Basis of Harvest and Preservation of Tissues” used as a support of an elective subject is presented. The aim of the e-learning course was to enable the students to learn the theoretical principles of the subject individually and to present the gained knowledge at the final seminar. Methods: All functions of the course were operated in Moodle, local application of the Charles University in Prague, Faculty of Medicine in Hradec Králové. The course was divided into 3 main topics corresponding with topics of lectures: 1. Principles of tissue and organ donation, 2. Low temperature preservation of cells, tissues and organs, 3. Quality and safety assurance in practice of tissue and procurement establishments. A test consisting of 5 questions selected randomly from the bank of questions followed each topic. If the student answers correctly at least 3 questions he is allowed to pass to the next topic. The fourth topic “Basic processes in the tissue establishment and principles of their validation” was added into the electronic version as a tool for repeating and improving of knowledge. The fifth topic was represented by a database for uploading theses presented by students at the final seminar. The final test consisted of 15 questions (5 ones from each basic topic). It was necessary to answer correctly at least 10 questions to receive a certificate of completing the course. Results: The course was put into operation during the summer term of the academic year 2012/2013. To the date 15 of September the total of 23 students enrolled (17, i.e. all students of the elective subject in the Czech version, 2 students of this subject in the English version, 2 postgraduate students and 2 medical doctors). All enrolled students used the course for on-line learning, downloading, or printing course study materials. All undergraduate students were obliged to use it for preparation, consultation and presentation of seminar theses, i.e. 10 minute Power Point presentations followed by 5 minute discussion. Verification of the course was planed during the summer term of the academic year 2013/2014. Conclusions: The first experience showed that the presented e-learning course could serve as a useful support of the elective subject “Basis of Harvest and Preservation of Tissues”. It substantially enlarged the choice of study materials that can be regularly updated, so that the students receive the newest information. It can be also attractive for students that are not enrolled in the elective subject because of its limited capacity (not more than 20 persons) The efficiency of such form of education is being a subject of further verification.

Autori prezentujú metódu voskovej impregnácie, ktorú demonštrujú na preparátoch srdca. Predkladajú podrobný opis preparácie orgánu, jeho úpravy a fixácie. Spracovanie zahŕňa bežný postup dehydratácie tkaniva s následnou impregnáciou parafínom. Tento konzervovaný trvalý preparát umožňuje zachovanie celého orgánu a súčasne znázomenie jeho jednotlivých štruktúr.
The authors present a method of paraffin infiltration applied to heart specimens. They describe the dissection, trimming and fixation of the specimen which is followed by common tissue dehydration and infiltration by paraffin wax. Thus preserved permanent specimen allows both retaining the enture organ and visualizing its particular structures.
Cílem práce bylo nalézt optimální konzervační médium pro krátkodobé uchování žilních štěpů, které by mohly být následně použity k potažení kovových stentů. Zevní jugulární žíla psa (n = 15) byla chirurgicky odebrána, rozdělena na díly a vložena do konzervačních médií. Khypotermní konzervaci (+ 4 °C) byly použity roztoky Optisol (Chiron, USA), University of Wisconsin (Baxter, USA), EurocoUins (Fresenius, SRN) a fyziologický roztok (Bieffe Medital, Itálie). Pro normotermní konzervaci (+ 37 °C) v atmosféře s 5% CO2 bylo použito Dulbeccovo médium pro tkáňové kultury (Sigma, USA). Při hypotermní konzervaci byly vzorky uchovávány po dobu 24 hodin, 3 dny a 7 dnů, při normotermní konzervaci v Dulbeccově médiu rovněž 24 hodin, 3 dny a 7 dnů. Vzorky byly hodnoceny světelnou mikroskopií a rastrovací elektronovou mikroskopií. Výsledky naší práce ukázaly, že k minimálním změnám na endoteliích žilních štěpů došlo při normotermní konzervaci v Dulbeccově médiu, kde ani po 7 dnech, nedošlo k odlučování endotelu či změnám vitality buněk. Pro hypotermní konzervaci se jako nejvhodnější ukázal roztok University of Wisconsin. Přidáním 5% lidského albuminu k tomuto roztoku se navíc podařilo s: sm'žit pyknózu buněčných jader endotelií. Vzorky uchovávané ve fyziologickém roztoku vykazovaly jaderné deformace, edém a ztrátu endotelií včetně počínajícího edému buněčné stěny již po 24 hodinách hypotermm' konzervace, použití tohoto roztoku ke konzervaci žil považujeme za nevhodné.
The objective of the work was to find an optimal preservation medium for short-term preservation of venous grafts which could be subsequently used to line metal stents. The external jugular vein of dogs (n = 15) was removed surgically, divided into portions and immersed into preservation media. For hypothermic preservation (+ 4 °C) solutions of Optisol (Chiron, USA), University of Wisconsin (Baxter,USA), EurocoUins (Fresenius, GFR) and saline (Bieffe Medital, Italy) were used. For normothermic preservation (+ 37 °C) in an atmosphere with 5 % CO2 Dulbecc's medium for tissue cultures (Sigma,USA) was used. During hypothermic preservation the specimens were kept for 24 hours, 3 and 7 days, during normothermic preservation in Dulbecc s medium also for 24 hours, 3 and 7 days. The specimens were evaluated by light microscopy and raster electron microscopy. The results revealed that mir nimal changes on the endotiielia of venous grafts occurred during normothermic preservation in Dulbecc's medium where after 7 days the endothelium did not become detached and the vitality of cells did not change. During hypothermic preservation the solution of Wisconsin University proved most suitable. By addition of 5 % human albumin to this solution it proved possible moreover to reduce pyknosis of the endothelial cell nuclei. The specimens kept in saline displayed deformities of the nuclei, oedema and loss of endothelia incl. incipient oedema of the cellular wall already after 24 hours of hypothermic preservation. The authors consider the use of this solution unsuitable for preservation.
Clinical success of small bowel transplantation depends on quality of the preservation small bowel graft which is notoriously sensitive to ischemia. There is still no general agreement as to which segment of the small bowel is preferred (jejunum or ileum) for clinical use. In our study, using a light microscopy and concentrations of tissue serotonin-positive cells, we tried to identify a part of the human intestine, which is more resistant to preservation injury sustained by HTK preservation solution with 1-24 hr of cold ischemia. Statistical analysis of both parameters did not reveal any significant differences between the jejunum and ileum. According to our data, there is no difference between jejunal and ileal grafts in susceptibility to ischemic injury due to cold ischemia within 24 hours when using HTK preservation solution. A significant difference was observed in histological pictures only after 12-hour of cold ischemia in both groups (jejunum and ileum) (Fig. 2, Ref. 11). Full Text (Free, PDF) www.bmj.sk.
