Zein is renewable plant protein with valuable film-forming properties that can be used as a packaging material. It is known that the addition of natural cross-linkers can enhance a film's tensile properties. In this study, we aimed to prepare antimicrobial zein-based films enriched with monolaurin, eugenol, oregano, and thyme essential oil. Films were prepared using the solvent casting technique from ethanol solution. Their physicochemical properties were investigated using structural, morphological, and thermal techniques. Polar and dispersive components were analyzed using two models to evaluate the effects on the surface free energy values. The antimicrobial activity was proven using a disk diffusion method and the suppression of bacterial growth was confirmed via a growth kinetics study with the Gompertz function. The films' morphological characteristics led to systems with uniform distribution of essential oils or eugenol droplets combined with a flat-plated structure of monolaurin. A unique combination of polyphenolic eugenol and amphiphilic monoglyceride provided highly stretchable films with enhanced barrier properties and efficiency against Gram-positive and Gram-negative bacteria, yeasts, and molds. The prepared zein-based films with tunable surface properties represent an alternative to non-renewable resources with a potential application as active packaging materials.

PURPOSE: To evaluate the short-term results of water vapor therapy (Rezūm) for BPH/LUTS in the first cohort of Czech patients. MATERIALS AND METHODS: Patients with BPH and moderate to severe LUTS (N = 76) who underwent Rezūm treatment from December 2019 to July 2020 were included in the prospective study. Prior to the procedure, they completed the IPSS and OABv8 questionnaires and underwent uroflowmetry, transrectal ultrasound of the prostate, and PSA sampling. The parameters before and 3 months after the procedure were compared and statistically evaluated. RESULTS: The study protocol was completed by 92% of patients (N = 70). We observed a significant increase in Qmax (median 17.7 vs. 8.8 mL/s, P < .001), Qave (9 vs. 4.5 mL/s, P = .001) and voided volume (241 vs. 171 mL, P < .001) and a significant reduction in post-void residual (average 17.5 vs. 67.7 mL), prostate volume (39.3 vs. 62.3 mL) and total PSA (median 1.9 vs. 2.5 ng/mL, resp. P values < .001). There was also a significant decrease in OABv8 score (average 7.6 vs. 16.6, P < .001) and IPSS QoL (1.6 vs. 4.0, P = .037). The improvement in the IPSS score was apparent, yet statistically insignificant (6.8 vs. 16, P = .079). CONCLUSION: Water vapor therapy is an effective and safe method of BPH/LUTS treatment in the short-term.

The purpose of this study was to compare the yield, chemical composition, antimicrobial and antioxidant properties of essential oils isolated from leaves of Laurus nobilis L. by two different distillation methods. The essential oils isolated by hydrodistillation (HD) and steam distillation (SD) were analyzed by gas chromatography coupled to mass spectrometry (GC-MS) and gas chromatography with flame ionization detector (GC-FID). Hydrodistillation produced a yield of 0.95 ± 0.06% which is slightly higher than yield obtained by steam distillation 0.79 ± 0.07%. Seventy three compounds in the bay leaves oil obtained by steam distillation were identified while in essential oil obtained by hydrodistillation were identified only 54 compounds. The antioxidant activity was evaluated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical method. Antimicrobial activity of obtained essential oils was evaluated by disc diffusion method in comparison with several chosen antimicrobials. The antimicrobial activity was tested on five microorganisms - Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa and Candida albicans. In general, oils produced by steam distillation had higher antimicrobial and antioxidant activities than hydrodistillation extracts. It seems that hydrodistillation is better for higher yield while steam distillation is better to use for more quality oils with stronger biological properties.
The present research was undertaken to develop a chitosan-collagen film for controlled delivery of combinations of local anesthetics. The film has been prepared by casting which is a versatile, rapid and low-cost approach distinguished by high reproducibility. The mechanical, morphological, and physicochemical properties of the films and the impact of the drug loading were evaluated. We showed that the formulations have a good combination of strength and flexibility with high water permeability. Surface morphology investigation indicates a variation in roughness depending on the loaded compound. Release studies were performed in controlled environments and the data processed by the Higuchi model to assess the dynamics of the release. The local anesthetics, lidocaine, tetracaine, and benzocaine, were uniformly distributed within the matrix and released in a rate and magnitude specific for the drug concentration and combination tunable in a range time from 6 h to 24 h. The films dissolve completely in the physiological environment within 24 h without leaving any toxic metabolites as both of the components are recognized as safe. In vitro cytotoxicity and cell proliferation tests performed on human dermal fibroblast demonstrate the biocompatibility and lack of cytotoxicity of the prepared formulations.

A study has been carried out on the volatile organic compounds (VOCs) in the exhaled breath of patients suffering from inflammatory bowel disease (IBD), comprising 136 with Crohn's disease (CD) and 51 with ulcerative colitis (UC), together with a cohort of 14 healthy persons as controls. Breath samples were collected by requesting the patients to inflate Nalophan bags, which were then quantitatively analysed using selected ion flow tube mass spectrometry (SIFT-MS). Initially, the focus was on n-pentane that had previously been quantified in single exhalations on-line to SIFT-MS for smaller cohorts of IBD patients. It was seen that the median concentration of pentane was elevated in the bag breath samples of the IBD patients compared to those of the healthy controls, in accordance with the previous study. However, the absolute median pentane concentrations in the bag samples were about a factor of two lower than those in the directly analysed single exhalations-a good illustration of the dilution of VOCs in the samples of breath collected into bags. Accounting for this dilution effect, the concentrations of the common breath VOCs, ethanol, propanol, acetone and isoprene, were largely as expected for healthy controls. The concentrations of the much less frequently measured hydrogen sulphide, acetic acid, propanoic acid and butanoic acid were seen to be more widely spread in the exhaled breath of the IBD patients compared to those for the healthy controls. The relative concentrations of pentane and these other VOCs weakly correlate with simple clinical activity indices. It is speculated that, potentially, hydrogen sulphide and these carboxylic acids could be exhaled breath biomarkers of intestinal bacterial overgrowth, which could assist therapeutic intervention and thus alleviate the symptoms of IBD.
