OBJECTIVES: To evaluate the effect of short-term inhalational exposure to nanoparticles released during dental composite grinding on oxidative stress and antioxidant capacity markers. MATERIALS AND METHODS: Twenty-four healthy volunteers were examined before and after exposure in dental workshop. They spent 76.8 ± 0.7 min in the testing room during grinding of dental nanocomposites. The individual exposure to aerosol particles in each participant´s breathing zones was monitored using a personal nanoparticle sampler (PENS). Exhaled breath condensate (EBC), blood, and urine samples were collected pre- and post-exposure to measure one oxidative stress marker, i.e., thiobarbituric acid reactive substances (TBARS), and two biomarkers of antioxidant capacity, i.e., ferric-reducing antioxidant power (FRAP) and reduced glutathione (GSH) by spectrophotometry. Spirometry and fractional exhaled nitric oxide (FeNO) were used to evaluate the effect of acute inhalational exposure. RESULTS: Mean mass of dental nanocomposite ground away was 0.88 ± 0.32 g. Average individual doses of respirable particles and nanoparticles measured by PENS were 380 ± 150 and 3.3 ± 1.3 μg, respectively. No significant increase of the post-exposure oxidative stress marker TBARS in EBC and plasma was seen. No decrease in antioxidant capacity biomarkers FRAP and GSH in EBC post-exposure was seen, either. Post-exposure, conjunctival hyperemia was seen in 62.5% volunteers; however, no impairment in spirometry or FeNO results was observed. No correlation of any biomarker measured with individual exposure was found, however, several correlations with interfering factors (age, body mass index, hypertension, dyslipidemia, and environmental pollution parameters) were seen. CONCLUSIONS: This study, using oxidative stress biomarker and antioxidant capacity biomarkers in biological fluids of volunteers during the grinding of dental nanocomposites did not prove a negative effect of this intense short-term exposure. However, further studies are needed to evaluate oxidative stress in long-term exposure of both stomatologists and patients and diverse populations with varying health statuses.

The present study aimed to evaluate the biochemical and antioxidant responses of common carp after exposure to the antiepileptic and analgesic drug gabapentin at concentrations of 0.1, 1, 10, and 100 μg/l for 4 weeks. The exposure to the highest two concentrations resulted in significant changes in plasma indices such as glucose (only group 100 μg/l), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), lipase, creatine kinase (CK), amylase as well as butyrylcholinesterase (BChE). Similar trends were found in both groups exposed to the environmentally relevant concentrations (i.e., 0.1 and 1 μg/l). In addition, a significant increase in the ferric-reducing power of the plasma was noted in all treated groups. Numerous changes in antioxidant enzymes, superoxide dismutase, catalase, glutathione-S-transferase, as well as lipid peroxidation, were observed especially in the caudal kidney in the group exposed to 100 μg/l. Significant findings were also confirmed in the group exposed to an environmentally relevant concentration (1 μg/l), with a decrease in superoxide dismutase in the gill and an increased lipoperoxidation in the caudal kidney. Our research shows that subchronic exposure to gabapentin may pose a significant risk to non-target aquatic organisms, such as disruption of metabolic pathways or induction of oxidative stress, even at environmentally relevant concentrations.

The utilization of plant-derived chemicals with anti-diabetic properties is widely promoted for its advantageous tactics in managing diabetes, as they are cost-effective and have minimal or no adverse effects. Therefore, this work investigates the medicinal plant Jasminum officinale L. leaves by extraction and bio-guided fractionation. The ethyl acetate fraction showed a higher yield of 36.4 %. A phytochemical test on Jasminum officinale confirmed flavonoids, saponins, phenols, and tannins. The highest total phenol and flavonoid contents in the ethyl acetate fraction of J. officinale are 103.01 ± 1.1 mg GAE/g and 80.29 ± 1.03 mg QUE/ value found in methanol crude extract. Furthermore, HPTLC analysis of the ethyl acetate fraction detected the existence of flavonoids (kaempferol) and phenols (gallic acid, quercetin, and rutin). The compounds detected at the greatest concentrations in the LC-M/MS analysis of the ethyl acetate fraction were cirsiliol, kaempferol, and 2-tridecanone. Additionally, J. officinale (IC50 33.845 ± 1.09 μg/mL) demonstrated the highest DPPH scavenging activity in EAF like that of ascorbic acid (IC50 22.27 ± 0.96 μg/mL). Also, in the FRAP assay, the IC50 of this fraction is 15.14 ± 0.25 μM Fe equivalents. In the range of alpha-amylase deactivating action, from 13.25 % to 74.51 %, and IC50 value (47.40 ± 0.29 μg/mL) was significantly higher in the ethyl acetate fraction of J. officinale leaf extract. Moreover, J. officinale leaf extract had a substantially higher retention of glucose level (23.92 ± 0.85 % to 87.21 ± 0.6 %), significantly higher anti-inflammatory activity with the lowest IC50 value (66.00 ± 1.84), and lipid peroxidation (IC50 value 34.67 ± 1.69) by utilizing egg yolk as a substrate for lipids. Overall, the study revealed that J. officinale has considerable anti-diabetic characteristics. However, further comprehensive research is necessary to ascertain the medicinal purposes of J. officinale and its chemical components, pharmacological effects, and clinical uses.

Antioxidants in nutrition are a widely discussed topic. In this study, a synergistic effect was observed for 13 selected substances - antioxidants and potential synergists, whereby two substances were mixed in the same concentration ratio of 1:1. The antioxidant capacity (AC) of the mixtures was determined using the FRAP method. The AC measured was compared with a theoretical AC value (as only additive effect) to calculate the synergistic or antagonistic effect. Out of 78 possible combinations, a synergistic effect (SE) was detected in 72. For the 10 combinations, the SE was more than twice that of the pure substances. The largest synergistic effect was exhibited by vanillin and 4-hydroxybenzoic acid with increases even above 200% compared to the pure substances. Some of the phenolic substances that were subject to measurement can be used for the fortification of fruit juices.

Diabetic nephropathy, characterized by inflammation and oxidative stress, poses a management challenge. This study investigates the effect of Polygonum hyrcanicum extract on diabetic nephropathy in alloxan-induced diabetic mice. In this experimental animal study, the P. hyrcanicum extract was prepared using continuous macerations. Thirty male Albino mice, divided into five groups, were induced with alloxan-induced diabetes. They received intraperitoneal injections of the plant extract (100 and 200 mg/kg) and metformin (300 mg/kg) for four weeks. Kidney and blood samples were collected to assess protein carbonyl, glutathione, lipid peroxidation, TNF-α and IL-6 levels. The amount of total flavonoid and phenolic content in the hydroalcoholic extract of P. hyrcanicum were 7.5 ± 0.3 mg of quercetin and 88.2 ± 1.3 mg gallic acid per gram of extract respectively. The antioxidant activity level of the hydroalcoholic extract was determined to be 1.78 ± 0.51 mM equivalent per gram of extract. Alloxan administration resulted in a significant reduction in glutathione levels and a significant increase in protein carbonyl, lipid peroxidation, TNF-α, and IL-6 levels. Hydroalcoholic extract of P. hyrcanicum effectively reduced oxidative stress markers and inflammatory cytokines (TNF-α, IL-6), indicating its potential in mitigating diabetic nephropathy. However, no significant difference in efficacy was observed between the 100 mg/kg and 200 mg/kg doses in terms of reducing these toxicities.

Foods enriched with insects can potentially prevent several health disorders, including cardiovascular diseases, by reducing inflammation and improving antioxidant status. In this study, Tenebrio molitor and Gryllus assimilis were selected to determine the effect on the development of atherosclerosis in ApoE/LDLR-/- mice. Animals were fed AIN-93G-based diets (control) with 10% Tenebrio molitor (TM) and 10% Gryllus assimilis (GA) for 8 weeks. The nutritional value as well as antioxidant activity of selected insects were determined. The lipid profile, liver enzyme activity, and the fatty acid composition of liver and adipose tissue of model mice were evaluated. Quantitative analysis of atherosclerotic lesions in the entire aorta was performed using the en face method, and for aortic roots, the cross-section method was used. The antioxidant status of the GA cricket was significantly higher compared to the TM larvae. The results showed that the area of atherosclerosis (en face method) was not significantly different between groups. Dietary GA reduced plaque formation in the aortic root; additionally, significant differences were observed in sections at 200 and 300 µm compared to other groups. Furthermore, liver enzyme ALT activity was lower in insect-fed groups compared to the control group. The finding suggests that a diet containing edible insect GA potentially prevents atherosclerotic plaque development in the aortic root, due to its high antioxidant activity.

The imidazolinone group of herbicides generally work for controlling weeds by limiting the synthesis of the aceto-hydroxy-acid enzyme, which is linked to the biosynthesis of branched-chain amino acids in plant cells. The herbicide imazethapyr is from the class and the active ingredient of this herbicide is the same as other herbicides Contour, Hammer, Overtop, Passport, Pivot, Pursuit, Pursuit Plus, and Resolve. It is commonly used for controlling weeds in soybeans, alfalfa hay, corn, rice, peanuts, etc. Generally, the herbicide imazethapyr is safe and non-toxic for target crops and environmentally friendly when it is used at low concentration levels. Even though crops are extremely susceptible to herbicide treatment at the seedling stage, there have been no observations of its higher dose on lentils (Lens culinaris Medik.) at that stage. The current study reports the consequence of imazethapyr treatment on phenolic acid and flavonoid contents along with the antioxidant activity of the phenolic extract. Imazethapyr treatment significantly increased the activities of several antioxidant enzymes, including phenylalanine ammonia lyase (PAL), phenol oxidase (POD), glutathione reductase (GR), and glutathione-s-transferase (GST), in lentil seedlings at doses of 0 RFD, 0.5 RFD, 1 RFD, 1.25 RFD, 1.5 RFD, and 2 RFD. Application of imazethapyr resulted in the 3.2 to 26.31 and 4.57-27.85% increase in mean phenolic acid and flavonoid content, respectively, over control. However, the consequent fold increase in mean antioxidant activity under 2, 2- diphenylpicrylhdrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assay system was in the range of 1.17-1.85 and 1.47-2.03%. Mean PAL and POD activities increased by 1.63 to 3.66 and 1.71 to 3.35-fold, respectively, in agreement with the rise in phenolic compounds, indicating that these enzyme's activities were modulated in response to herbicide treatment. Following herbicide treatments, the mean thiol content also increased significantly in corroboration with the enhancement in GR activity in a dose-dependent approach. A similar increase in GST activity was also observed with increasing herbicide dose.

The Kékfrankos is the most frequently cultivated wine grape in Hungary, with a significant national and regional impact, resulting in considerable amounts of byproducts (e.g. pomace, seeds). To the best of our knowledge no research has been conducted on the antioxidant and antibacterial properties of its seed extracts (GSE). A novel apporach of applying direct microwave treatment on grape seeds was implemented for the first time to enhance antioxidant and antimicrobial properties of GSE. Antioxidant properties were assayed using the DPPH (2,2-diphenyl-1-picrylhydrazyl), FRAP (Ferric Reducing Antioxidant Power) and TPC (Folin-Ciocâlteu's Total Polyphenol Content) methods. Profile and content of polyphenols was studied using high-performance liquid chromatography/tandem mass spectrometry and matrix-assisted laser desorption/ionization mass spectrometry. Antibacterial properties were evaluated using Gram-positive Staphylococcus aureus (SA), methicillin-resistant Staphylococcus aureus (ST239) (MRSA) and Gram-negative Escherichia coli (EC) bacteria strains. Results proved that the mild direct microwave treatment of grape seeds significantly increased total polyphenol, (+)-catechin, (-)-epicatechin as well as antioxidant capacity levels by 20-30 % compared to untreated samples and resulted the best antibacterial properties based on bacterial growth curves (SA and MRSA: 0.015625 mg/mL, EC: 0.25 mg/mL). Results justify the importance of further pharmacological investigations on Kékfrankos grape seed extracts and that the direct microwave treatment of grape seeds is an innovative approach for the fast and cost efficient improvement of the antibacterial properties of grape seed extracts.

Mosses (Bryophyta), particularly species of the genus Sphagnum, which have been used for centuries for the treatment of skin diseases and damage, are still not explored enough in terms of their use in cosmetics. The purpose of this study was to determine the antioxidant properties of water-ethanol extracts from four selected species of the genus Sphagnum (S. girgenshonii Russow, S. magellanicum Brid., S. palustre L., and S. squarrosum Crome) and their impact on the expression of genes encoding key enzymes for the functioning of the skin. In this study, the effects of Sphagnum extracts on the expression of genes encoding tyrosinase, collagenase, elastase, hyaluronidase and hyaluronic acid synthase in human dermal fibroblasts were determined for the first time in vitro. The extracts inhibited tyrosinase gene expression and showed antioxidant activity. The experiment showed an increase in the expression of some genes encoding collagenase (MMP1) or hyaluronidase (HYAL2, HYAL3 and HYAL4) and a decrease in the hyaluronan synthase (HAS1, HAS2 and HAS3) genes expression by the tested extracts. The obtained results suggest that using extracts from the tested Sphagnum species in anti-aging cosmetics does not seem beneficial. Further studies are needed to clarify their impact on the skin.

INTRODUCTION: Drought stress unfavorably influences the growth and physiological traits of plants in the arid and semi-arid regions of the world. This study aimed to determine the effects of arbuscular mycorrhiza fungi (AMF; Funneliformis mosseae) inoculation on the physiological and biochemical responses of summer savory (Satureja hortensis L.) under different irrigation regimes. METHODS: The first factor was different irrigation regimes, including no drought stress (100% field capacity; FC), moderate drought stress (60% FC), and severe drought stress (30% FC); the second factor included the plants without AMF (AMF0) and with AMF inoculation (AMF1). RESULTS: The results showed that better values, higher plant height, shoot mass (fresh and dry weight), relative water content (RWC), membrane stability index (MSI), photosynthesis pigments, Fv, Fm, Fv/Fm, and total soluble proteins were obtained in the plants inoculated with AMF. The highest values were obtained for plants with no drought stress, then the plants subjected to AMF1 under 60% FC, and the lowest ones for plants under 30% FC without AMF inoculation. Thus, these properties are reduced under moderate and severe drought stress. At the same time, the utmost activity of superoxide dismutase (SOD), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), and the highest malondialdehyde (MDA), H2O2, proline, and antioxidant activity (TAA) were achieved for 30% FC + AMF0. It was also found that AMF inoculation improved essential oil (EO) composition, also as EO obtained from plants under drought stress. Carvacrol (50.84-60.03%) was the dominant component in EO; γ-terpinene (19.03-27.33%), p-cymene, α-terpinene, and myrcene, were recognized as other important components in EO. The higher carvacrol and γ-terpinene contents were obtained from summer savory plants with AMF inoculation and the lowest for plants without AMF and under 30% FC. CONCLUSION: According to the present findings, using AMF inoculation could be a sustainable and eco-friendly approach to improve physiological and biochemical characteristics and the essential oil quality of summer savory plants under water shortage conditions.
