The influence of repeated i.v. administration of dimethoxybenfluron (NO-1-B) (12 or 24 mg base/kg once weekly, 10 weeks) on biochemical and haematological parameters were studied in rabbits in vivo. No significant changes were mostly found in the serum ion levels between the dimethoxybenfluron and the control groups, as well as in most of other biochemical parameters (including total protein and albumin levels). Nevertheless, the lower dose of dimethoxybenfluron caused an increase in the glucose level. Furthermore, no significant changes were mostly present also in haematological parameters in the dimethoxybenfluron groups of rabbits (a mild decrease in thrombocytes and leucocytes). The results of our study support an assumption of good tolerance of dimethoxybenfluron from the viewpoint of its influence on biochemical and haematological parameters in rabbits and may be considered of importance for a possible therapeutic use of the derivatives.

BACKGROUND: Selenitetriglycerides are biologically active, organic forms of selenium formed as a result of the modification of selenic acid and sunflower oil. Studies in rats have shown that they are well absorbed and of low toxicity. There are no published studies on selenitetriglycerides supplementation in calves. RESULTS: In this study, selenitetriglycerides were administered once orally on the 2nd day of life at a dose of 0.5 or 1 mg Se/kg body weight to each of six Holstein-Friesian calves while six control calves were not supplemented. Blood for determination of selenium concentration, glutathione peroxidase activity, haematological parameters, aspartate aminotransferase, creatine kinase, and lactate dehydrogenase activities and glucose, total protein, albumin, triglycerides, cholesterol, urea, and creatinine concentration was collected before supplementation (day 0) and 1, 2, 5, 10 and 14 days after supplementation. Selenitetriglycerides administration increased (P < 0.01) serum selenium concentration in supplemented calves as early as day1, from a mean of 63.4 to 184.22 µg/l in calves receiving selenium at a dose of 0.5 mg/kg BW, and from 63.17 to 200.33 µg/l in calves receiving 1 mg/kg. Serum selenium concentrations remained significantly higher compared to the control group throughout the experiment. Glutathione peroxidase activity was higher in supplemented than control calves, significantly so in animals receiving the 1 mg/kg dose of Se on the 10th and 14th days (P < 0.05). There were no significant differences in the haematological and biochemical parameters between the groups. CONCLUSIONS: This experiment showed that supplementation with selenitetriglycerides could significantly improve blood selenium status in calves without adverse effects on haematological or biochemical parameters. These findings are essential prerequisites for future studies on selenitetriglycerides supplementation to manage clinical selenium deficiency in calves.

OBJECTIVES: Deoxynivalenol (DON) - trichothecene mycotoxin, is frequently detected in high concentrations in cereals in the temperate region of Europe. The aim of this study was to determine the effect of DON in feed on haematological and biochemical parameters and on oxidative stress in piglets. METHODS: Two concentrations of DON in feedstuff for pigs were chosen: 0.6 mg/kg (group C) and 2.0 mg/kg (group M). Twelve weaned pigs were used in each group. Pigs were fed with naturally contaminated feed for 4 weeks. On days 14, 21 and at the end of the experiment (day 28) samples of blood were taken to determine haematological parameters, plasma biochemical parameters, ceruloplasmin activity and FRAP (ferric reducing ability of plasma). RESULTS: The haematological variables did not show changes in response to contaminated diet with exception of the mean corpuscular volume, which was significantly decreased at the end of the experiment in the group M. A significant increase of alkaline phosphatase activity (140%, p<0.01) was found in the group M compared to the group C at the end of the experiment. A significant decrease was found on the day 21 in FRAP (85%, p<0.001) and on the day 28 in ceruloplasmin (75%, p<0.01) in the group M compared to the group C. CONCLUSIONS: The decrease of FRAP and ceruloplasmin indicate a lowered ability of organism to scavenge reactive oxygen species. The higher concentration of DON in feedstuffs had a negative influence on the antioxidant ability of piglet's plasma.

Haematological parameters, plasma iron concentration, and bodyweight were monitored in Melanoma-bearing Libechov Minipigs (MeLiM) from 5 to 18 weeks old. Animals with melanoma progression (P group) and spontaneous regression (SR group) were compared. The P group showed the lowest median values of red blood cell counts (RBC), haematocrit (HCT), haemoglobin concentration (HGB), and bodyweight, whereas the control white (tumour-free) pigs (C group) revealed the highest mean values of these parameters. The mean values of pigs with SR fell between the P and C groups. In addition, a stable concentration of plasma iron was found in the C group, while iron deficiency that increases with age was observed in the MeLiM groups. These results indicate that MeLiM are affected by cancer-related microcytic hypochromic anaemia. The lowest values of HGB, RBC, and HCT, together with the highest number of platelets (PLT) in the P group correspond to melanoma progression. Higher values of these parameters and lower PLT in the MeLiM pigs with SR reflected health improvement due to the destruction of melanoma cells during spontaneous regression. Monitoring of these haematological parameters can help distinguish MeLiM piglets with progression and spontaneous regression of melanoma in the early stages of postnatal development. The findings of this study correspond to findings in human patients in which cancer-related anaemia, thrombocytosis, and iron deficiency are often diagnosed.

OBJECTIVES: To investigate whether a combination of hysterectomy and antibiotic treatment leads to an improvement of altered haematological and immunological parameters in bitches affected by pyometra. METHODS: Blood samples obtained from 13 bitches affected by pyometra were investigated before hysterectomy and seven days later for a total and differential leucocyte count, activity of neutrophils and lymphocytes, and quantification of total serum immunoglobulins, lysozyme and circulating immune complexes. RESULTS: The parameters most affected included changes in blood profile (leucocytosis because of neutrophilia or leucopenia) and inhibition of lymphocyte activity. Seven days after hysterectomy, all affected parameters returned to normal levels comparable to clinically healthy dogs. CLINICAL SIGNIFICANCE: Application of immunostimulating therapy is not necessary to manage changes in blood cell counts and suppression of lymphocyte activity in bitches with pyometra. Removal of the infected uterus, being the source of infection, leads to improvement.

Thirty-five juvenile (mean age 6.3 weeks) and 35 adult (mean age 2.0 years) healthy degus (Octodon degus) were studied to investigate selected haematological and plasma biochemistry parameters. Animals were anaesthetised with isoflurane, and blood was withdrawn from the cranial vena cava. Erythrocyte, haematocrit and neutrophil counts (including the percentage of neutrophils) were significantly higher in the adult degus than in the juveniles. In contrast, the reticulocyte count, mean corpuscular volume, mean corpuscular haemoglobin, number of platelets and percentage of lymphocytes were significantly lower in the adult animals. Total protein and globulin levels were significantly higher in the adult degus. The albumin:globulin ratio and plasma levels of urea nitrogen, cholesterol, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, potassium, total calcium and inorganic phosphorus were significantly lower in adults than in juveniles.

The development of selected haematological and biochemical parameters during the laying period was monitored in common pheasant hens housed in an enhanced cage system. The cages were enhanced by the addition of two perches and a shelter formed by strips of cloth hanging in the corner of the cage. The results showed significant changes in the haematological and biochemical parameters monitored during egg laying. At the time when laying capacity approached a maximum, a decrease was observed (P < 0.05) in haematocrit, erythrocytes, and haemoglobin values, whereas monocytes, eosinophils, the heterophil/lymphocyte ratio, phosphorus, and calcium exhibited an increase (P < 0.05). At the end of the laying period, an increase (P < 0.05) was recorded in the count of leukocytes, heterophils, lymphocytes and basophils, the heterophil to lymphocyte ratio, and the concentrations of aspartate aminotransferase, cholesterol, phosphorus, and calcium, whereas lower values (P < 0.05) were recorded for haematocrit and plasma total protein in comparison with the values of the indicators at the beginning of the laying period. The results provide new information about dynamic changes in selected haematological and biochemical parameters in clinically healthy common pheasant hens during the laying period.

African grey parrots (Psittacus erithacus) are very popular pets, commonly seen in avian clinical practice. Haematological profiles are critical to the understanding of several disease processes, being particularly useful as diagnostic tools in clinical practice, since birds tend to hide clinical signs of disease. We have previously proposed new haematological reference intervals (RI) for captive African grey parrots, and in the present work the basic data obtained was studied in detail to investigate the influence of factors, such as age and sex, on the haematological profile of this bird species. During an 8-year period (March 2009 to July 2017), animals (n = 239) examined in first consultations or check-ups at the Zoològic Veterinaris (Barcelona) were submitted to blood collection at different time points, rendering a total of 459 blood samples. The haematological testing was performed according to the guidelines of the American Society of Veterinary Clinical Pathology to determine the packed cell volume (PCV), haemoglobin (Hb), mean haemoglobin concentration (MHC), mean corpuscular volume (MCV), mean corpuscular haemoglobin concentration (MCHC), total erythrocyte count (TRBC), total leukocyte count (TWBC), and differential leukogram with absolute and relative counts. All the haematological testing was performed in an in-house laboratory as previously described. Animals with 0 to 4 years of age showed higher values of PCV (P < 0.001), Hb (P = 0.023) and RBC (P = 0.018), and lower values of MCHC (P = 0.008), WBC (P = 0.012) and heterophils (P < 0.001) than older animals. There were significant differences exhibited in the monocytes (P = 0.035) between different age groups. Females presented higher PCV, Hb and RBC values (P < 0.001) compared to males. Our results suggest that the age and sex influence the haematological parameters in a significant manner in African grey parrots and should be accounted for when assessing the health status of individuals from this species.

OBJECTIVES: Therapeutic drug monitoring of thiopurine erythrocyte levels is not available in all centers and it usually requires quite a long time to obtain the results. The aims of this study were to build a model predicting low levels of 6-thioguanine and 6-methylmercaptopurine in pediatric inflammatory bowel disease (IBD) patients and to build a model to predict nonadherence in patients treated with azathioprine (AZA). METHODS: The study consisted of 332 observations in 88 pediatric IBD patients. Low AZA dosing was defined as 6-thioguanine levels <125 pmol/8 × 10 erythrocytes and 6-methylmercaptopurine levels <5700 pmol/8 × 10 erythrocytes. Nonadherence was defined as undetectable levels of 6-thioguanine and 6-methylmercaptopurine <240 pmol/8 × 10 erythrocytes. Data were divided into training and testing part. To construct the model predicting low 6-thioguanine levels, nonadherence, and the level of 6-thioguanine, the modification of random forest method with cross-validation and resampling was used. RESULTS: The final models predicting low 6-thioguanine levels and nonadherence had area under the curve, 0.87 and 0.94; sensitivity, 0.81 and 0.82; specificity, 0.80 and 86; and distance, 0.31 and 0.21, respectively, when applied on the testing part of the dataset. When the final model for prediction of 6-thioguanine values was applied on testing dataset, a root-mean-square error of 110 was obtained. CONCLUSIONS: Using easily obtained laboratory parameters, we constructed a model with sufficient accuracy to predict patients with low 6-thioguanine levels and a model for prediction of AZA treatment nonadherence (web applications: https://hradskyo.shinyapps.io/6TG_prediction/ and https://hradskyo.shinyapps.io/Non_adherence/).

OBJECTIVES: The study aimed at assessing the effects of a pyrethroid cypermethrin on common carp (Cyprinus carpio L.) on the basis of the results of acute toxicity tests and haematological, biochemical and histological examinations. METHODS: Experimental carp were exposed to the pesticide preparation Alimetrin 10 EM in the concentration of 29.1 microg/l(-1) (96hLC(50)) corresponding to 29.1 microg/l(-1) of cypermethrin in a 96 h acute toxicity test. The results of the test and of the examinations of exposed carp were compared with the results of carp from the control group. RESULTS: In haematological profile, the experimental group of carp showed a significant (P<0.01) increase in the number of erythrocytes, segmented neutrophile granulocytes, developmental forms of myeloid sequence and eosinophiles, and a significant (P<0.01) decrease in mean erythrocyte volume, mean erythrocyte haemoglobin and lymphocyte count. As far as biochemical indices are concerned, the exposure caused a significant (P<0.01) increase in glucose and lactate concentration and creatine kinase activity, and a significant (P<0.01) decrease in total proteins, albumins, total globulins, ammonia, lactate dehydrogenase and alkaline phosphatase in blood plasma of exposed carp compared to the control group. Histopathological tissue examinations proved random hyperaemia and perivascular lymphocyte infiltration in skin, respiratory epithelium hyperplasia and chloride cell activation in gills, and pancreas exocrine cell vacuolisation in common carp after the exposure. CONCLUSIONS: The exposure of common carp to 29.1 microg/l(-1) of Alimetrin 10 EM caused significant shifts in haematological and biochemical profile and histopathological changes in organ tissues. The cypermethrin-based pesticide Alimetrin 10 EM was classified among substances strongly toxic for fish.
