The effects were investigated of a hormonal preparation of serum gonadotrophin (SC) on neurosecretion, PAS-positive mucopolysaccharides and on histological changes in the third cerebral ventricle; at the same time variations were determined of catecholamine concentrations of ependyma surface in the hypothalamic regions that control directly reproductive functions in ewes of the Merino and Wallachian breeds in the oestrus period. Ewe oestrus was synchronized by insertion of Ageline (Spofa) sponges into the vagina. A dose of 2000 IU of serum gonadotrophin (Bioveta, Ivanovice in Haná) was administered to stimulate the ewes (n = 30). Twelve ewes of the Wallachian breed, six control and six test ones, were used for radioenzymatic detection of catecholamines (Catechola test, Prague). The brains were segmented immediately after bleeding and samples were taken from the eminentia medialis, area preoptica and corpus mamillare pursuant to the stereotactic configuration of the sheep hypothalamus (Welento et al., 1968). The brains of ten ewes of the Merino breed, four control and six test ones, and of eight ewes of the Wallachian breed (four control and four test ones) were used for histological treatment. For the purposes of a histological study, the brains were treated with current histological methods. The amount of neurosecretory material was assessed by light microscopy (Nakahara, 1963). Samples for the study of ependyma of the lower part of the third cerebral ventricle were examined in a scanning electron microscope. The obtained results demonstrate a statistically significant decrease by 36.4% (P less than 0.05) in epinephrin concentrations in the area preoptica of hypothalamus after SG administration (Fig. 3), in comparison with the control group. A decrease in norepinephrin concentration in the corpus mamillare (Fig. 4) was statistically significant (P less than 0.001). The dose of 2000 IU SG influenced most markedly catecholamine concentrations in the eminentia medialis (Fig. 5), where a statistically significant decrease in epinephrin and norepinephrin concentration was observed (P less than 0.01). Dopamine concentrations also decreased significantly (P less than 0.05) in the eminentia medialis (Fig. 5). Following the hormonal stimulation, a significant increase in dopamine and epinephrin concentrations was observed in the hypophysis of sheep (Fig. 6; P less than 0.05) and in the epiphysis there was a significant decrease in epinephrin and dopamine concentration (P less than 0.01).(ABSTRACT TRUNCATED AT 400 WORDS)

Experiments were carried out with six sheep of the Slovak Merino breed, weighing 22-28 kg. For 28 days the animals were given 4 mg inorganic Hg2+ in the feed per animal/day. In contrast with the controls, the following residual mercury concentrations were determined in the single organs and tissues: liver 1.580 +/- 0.326 mg.kg-1 Hg2+ and 0.091 +/- 0.014 mg.kg-1 Hg2+, respectively muscle 0.064 +/- 0.009 mg.kg-1 Hg2+, and 0.026 +/- 0.006 mg.kg-1 Hg2+, resp. spleen 0.142 +/- 0.025 mg.kg-1 Hg2+, and 0.022 +/- 0.010 mg.kg-1 Hg2+, resp. kidney 9.054 +/- 3.794 mg.kg-1 Hg2+, and 0.128 +/- 0.080 mg.kg-1 Hg2+, resp. (Fig. 1), abomasal contents 0.309 +/- 0.069 mg.kg-1 Hg2+, and 0.021 +/- 0.007 mg.kg-1 Hg2+, resp. large intestinal contents 0.267 +/- 0.058 mg.kg-1 Hg2+, and 0.043 +/- 0.004 mg.kg-1 Hg2+, resp. The results suggest that the long-term ingestion of mercury with feed leads to a pronounced Hg accumulation in the kidneys and liver. Much lower levels were observed in the muscle tissue and spleen. The affinity of mercury to the kidney and liver is probably related to the preferential bonds of organic mercury compounds to the SH- groups of the plasma proteins in these organs. It is the bond to the sulphydryl groups of proteins that results in the inhibition of proteosynthesis and thus enzyme and antibody inhibition. Under the conditions of continuing chemical contamination of the environment, a permanent supply of low concentrations of heavy metals the animal organism is observed.(ABSTRACT TRUNCATED AT 250 WORDS)

A three-month feeding trial was conducted with sheep of the Slovak Merino breed (13-14 months of age, males and females) as a part of the obligatory toxicological tests of the Czechoslovak developmental herbicide bentazone (Research Institute for Chemical Technology, Bratislava). Partial results are shown describing the action of this herbicide on the overall health of sheep with respect to weight gains, trias values and intrauterine foetus development. The sheep of the first experimental group were fed bentazone as an additive to molasses feed M at a daily rate of 195 mg/kg lwt. (1/10 from LD50), and the sheep of the second group were fed a rate of 97.5 mg/kg lwt. (1/20 from LD50). No symptoms of the toxic effects of bentazone were observed in the course of the experiment. There were no differences between the experimental and test animals in the dynamics of pulse value changes (Fig. 1), breathing value changes (Fig. 2) and body temperature value changes (Fig. 3) from the beginning till the end of the trial. A slight increase in the pulse and breathing values above the upper reference value observed in the experimental groups in the last two to three weeks of the trial cannot be associated with the bentazone action. This increase reflects the physiological condition of the animals in the given conditions because the identical changes were also found in the control group. We assume that the changes were evoked by the elevated temperature of the external environment due to the fact that the sheep were not sheared.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of a serum gonadotropin (SG) superovulation hormonal preparation were investigated on catecholamine levels (norepinephrine, dopamine and epinephrine) in the blood plasma of ewes with synchronized oestrus in the oestrus period. In this trial the blood plasma of eleven ewes of the Slovak Merino breed was analyzed to detect catecholamines in the oestrus period. Superovulation was induced by an i.m. administration of 1500 IU SG as soon as oestrus synchronization with Agelin vaginal tampons finished (20 mg chlorsuperlutin) which lasted for 10 days. Catecholamines were detected in the blood plasma before synchronization, on the day of Agelin vaginal tampons application, and in 48 and 72 hours after the hormone administration--on the days of the expected ovulation. Catecholamine concentrations in the blood plasma were determined by a radioenzymatic assay using a Catechola test (Praha). The results indicate that synchronization and hormonal stimulation influence plasma catecholamine levels. The norepinephrine (NE) concentration in the blood plasma of the control samples has the value of 8.31 +/- 0.732 pmol/ml. An insignificant increase in the NE levels (13.12 +/- 0.120 pmol/ml; Fig. 1) was recorded on day 1 of the trial, after the start of synchronization. On the day of the expected ovulation the NE concentrations rose to 17.12 +/- 1.289 pmol/ml (P less than 0.001) and they remained significantly increased (P less than 0.01) at the level of 12.89 +/- 1.020 pmol/ml on the following day of the experiment. The effects of zinc administration at a rate of 3 mg/kg lw. in the preparation Zindep inj. ad usum vet. (Biotika, Slovenská L'upca) were evaluated as exerted on zinc concentrations in the blood serum of 16 dairy cows in the middle of the 7th month of pregnancy. With respect to zinc injection, T-rosetted lymphocytes and beta-lysine activity were inestigated. Blood was collected from all dairy cows from v. jugularis before the preparation was administered, on days 2, 5, 8, 15, 30 and 60 after Zindep administration. Atom absorption spectrophotometry, applying a flame technique on a Perkin Elmer 1100 apparatus (Bíres, 1986), was used to determine Zn concentrations in the blood serum of all dairy cows. T-lymphocytes were determined by a rosette test after Paul et al. (1977), and beta-lysine was detected spectrophotometrically after Bucharin et al. (1987). Zinc dynamics in the blood serum of dairy cows is presented in Fig. 1. The starting values of zinc in the test cows were 9.68 +/- 2.30 mumol/l and in the control ones 10.15 +/- 1.27 mumol/l. Zincaemia of experimental dairy cows was significantly lower (P less than 0.01) on day 2 after Zindep administration, in comparison with the control group. A significant increase in zinc concentrations in the blood serum of experimental animals, in comparison with the control ones, was observed from day 8 to day 60 (P less than 0.01). The maximum zincaemia values were recorded in experimental dairy cows within days 15 and 30 (15.65 +/- 3.33, and/or 14.55 +/- 2.10 mumol/l).(ABSTRACT TRUNCATED AT 250 WORDS)

Sixty-one sheep were administered experimentally a VX chemical (organophosphate) at different rates. In the process of dissection, samples were taken for a histopathological examination. These were samples of brain, liver, kidneys, rumen wall, small intestine, muscles, myocardium, lungs and spleen. For another histochemical examination, samples of tongue, m. longissimus dorsi, jejunum, rumen, liver, kidneys and m. interconstalis were also taken. The activities of alkaline and acid phosphatase, nonspecific esterase, acetylcholinesterase and dehydrogenase of lactic acid were investigated. The most significant changes were found out in the lungs - in form of oedemas and acute catarrhal bronchopneumonias in those animals which died within three days after chemical administration. Sporadic haemorrhages or haemorrhages in form of vast spots were found out under the epicardium. Their range did not relate to the amount of the chemical administered. Rather dilated vessels were observed in the brain and also in the meninges. The histochemical examination showed different activities of enzymes in particular organs of sheep.

Developmental dynamics was investigated in the activity of glutamate dehydrogenase (GDH, E.C. 1.4.1.2.-4) and glutamine synthetase (GS, E.C. 6.3.1.2) in different parts of the digestive tract of lambs, in dependence on the age from 10 to 90 days; the goal of these investigations was to elucidate in greater detail the role of the above enzymes in nitrogen metabolism. The activity of GDH, and of the coenzymes NADH and NADPH, was followed in the digesta because simple organisms (bacteria, fungi, plants) have two glutamate dehydrogenases: they differ from each other by coenzyme specificity, unlike GDH from animal sources which can utilize both NADH coenzyme and NADPH coenzyme (Fahien et al., 1965; Frieden, 1964). The following activities of GDH and GS were found out in trials with lambs at the age of 10, 20, 30, 40 and 90 days, as to the different parts of digestive tract: in the tissues of rumen, omasum, reticulum, spleen, duodenum, jejunum, ileum, int. caecum and colon the activity of GDH (NADH) varied from 0.031 to 0.305 nkat/mg dry matter, in the digesta from 0 to 2.92 nkat/mg dry matter. An investigation of GDH (NADH, NADPH) dynamics in the digesta of lambs showed the relatively high activity of GDH (NADH) in the digesta of colon at the age of 10 days and that of GDH (NADPH) in the digesta of int. caecum. The activity of GDH (NADH) was also found to be high in the digesta of int. caecum at the age of 20 days. In that period the activity of GDH (NADH, NADPH) in the digesta of rumen, omasum and reticulum was zero.(ABSTRACT TRUNCATED AT 250 WORDS)

Serine proteases help to regulate the ovarian cycle at different levels and they are subjected to the control of gonadotropic hormones and protease inhibitors. Superovulation stimulations influence the activities of trypsin inhibitors (model serine protease) in blood plasma (BP) and in follicular fluid (FF), and also in dependence on the breed. Trypsin inhibiting activities were determined from the reduced rate of trypsin hydrolysis of a chromogenic substrate (TAPA) and they were determined in percent. A change in absorbancy at 405 nm = 1.0 after 10-minute incubation at 25 degrees C and pH = 8.05 was taken as 100%. The incubation mixture as a sample contained 100 microliters blood plasma or 10 microliters follicular fluid, diluted with gammaglobulin at 1:10. The differences in the trypsin inhibiting activities (TIA) of BP in ewes of the Merino, Tsigai and Wallachian breeds were insignificant, but Agelin synchronization (20 mg chlorsuperlutin per vaginal swab) induced statistically significant differences. The lowest TIA BP was recorded in the Tsigai breed (T), P less than 0.001 in comparison with the Wallachian (W) and Merino (M) breeds. Following the administration of 1,500 IU PMSG, the TIA BP within 120 hours decreased in W (P less than 0.001), it increased in T (P less than 0.1) and in M the changes in the TIA BP were insignificant. The average numbers of ovulations increased from 2.25 +/- 2.5 to 3.0 +/- 1.2 in W; from 0.25 +/- 0.43 to 2.5 +/- 1.6 in T and from 0.00 +/- 0.0 to 2.5 +/- 2.3 in M. Following the single administration of 2,000 IU PMSG after Agelin synchronization, the changes in M ewes were also insignificant, and there were no different responses in pregnant (1st to 2nd month) and nonpregnant ewes. In pregnant T ewes the TIA BP increased after Agelin synchronization and stimulation (P less than 0.01), in nonpregnant ewes these changes were not significant. In W lambing ewes the TIA BP increased (P less than 0.001), the effects of Agelin were greater than those of PMSG. The TIA of follicular fluid (FF) of antral follicles were on average tenfold if compared with BP. After hormonal treatment of ovaries, the TIA FF mostly increased at different levels of statistical significance. The TIA FF of follicles less than 10 mm were lower than in follicles greater than 10 mm (P less than 0.001 for M and W, P less than 0.1 for T).(ABSTRACT TRUNCATED AT 400 WORDS)

Digestibility of nutrients was determined in healthy calves and in those with diarrhoea at the age of two and three weeks. The total collection of feces and indigestible marker methods (with Cr2O3) were used. High equality of results and statistically non-significant differences between the method of a total collection of feces and indigestible marker one in the suckling calves (Tab. II) confirm the suitability of using these two methods to determine digestibility. Based on the composition and commercially recommended feeding guide of milk replacer, it is clear that it has lower digestible energy, Zn and Fe intake. Slightly higher digestible crude protein and Mg intake, optimum Ca and P intake, all these values were calculated for 1 kg of live weight (Tab. I). Nutrient contents is responsible for nutrient ratio which is unbalanced and for low contents of digestible energy (76.5 KJ per g of digestible crude protein) instead of recommended values of 100-110 KJ per of digestible crude protein in milk replacer in suckling calves. In six clinically healthy calves fed with milk replacer Laktavit twice a day, an apparent digestibility of organic matter was found to be 95.7 +/- 1.8%, crude protein 93.5 +/- 1.8%, fat 94.5 +/- 1.4%, carbohydrates 97.2 +/- 1.5%, Ca absorption was 86.5 +/- 2.8%, P 95.3 +/- 2.9%. There was a depressed absorption for Mg 43.4 +/- 6.8%, Fe 24.2 +/- 4.3% and Zn 36.8 +/- 3.5% (Tab. III).(ABSTRACT TRUNCATED AT 250 WORDS)

The synthesis and secretion of trypsin (trypsin model serine protease) inhibitors are regulated in ovarian follicles by gonadotropins. The superovulation stimulations with 400 IU FSH, 1000 IU PMSG, 1000 IU HCG, 750 IIU PMSG + 750 IU HCG influence in a different way the trypsin inhibiting activities (TIA) of blood plasma (BP) (Figs 1 and 2) and follicular fluid (fig. 3); this points to a possibility of local effects. An increase in the average values of TIA in BP was statistically significant during the whole experiment: P less than 0.05 to P less than 0.001 (following the administration of PMSG+HCG, or PMSG, and HCG); Antisergon administered in 68 hours after PMSG reduced this increase. The changes in the fraction of low-molecular TIA in BP (after BP treatment with perchloric acid) were of converse nature; a decrease in the average values ranged from P less than 0.02 to P less than 0.001 (following PMSG or other stimulations). Antisergon did not influence this decrease. The changes observed on particular days of the trial (Figs. 1 and 2) also indicate different effects of the preparations, mainly of the component LH, which resulted in the occurrence of large nonovulating follicles (greater than 10 mm--"cystic" ones). No such follicles were observed in nonstimulated ewes and after FSH stimulation. The administration of antisergon (goat's antiserum against PMSG) 68 hours after PMSG administration did not prevent their creation. The TIA of follicular fluid (FF) of antral follicles was on average tenfold in comparison with that of blood plasma; and the TIA FF of follicles greater than 10 mm was higher (up to P less than 0.001) than the TIA FF of follicles less than 10 mm. The administration of Antisergon in shorter intervals following PMSG administration (12, 24, 48 and 58 hours) influenced the average values of TIA BP in 120 hours (since PMSG administration) in dependence on time (Tab. I). The effects of Antisergon administered in 12 and 24 hours after PMSG administration on the TIA BP were insignificant if it was administered in 48 and 58 hours the TIA BP increased (P less than 0.02; P less than 0.001) in comparison with the interval of 12 hours. The TIA FF of follicles less than 5 mm, 5-10 mm and greater than 10 mm varied in dependence on the time intervals of Antisergon administration (Fig. 4).
