Bee health is influenced by multiple factors, including nutrition, immunity, and parasitic pressures. Since the spread of Varroa destructor, overwintering survival has significantly declined, making it one of the most serious threats to honey bee (Apis mellifera L.) populations worldwide. Natural acaricides, such as oxalic acid (OA), are widely employed for managing Varroa mites; however, their pharmacodynamics, particularly their impacts on honey bee physiology and immunity, remain insufficiently understood. We studied effects of oxalic acid on honey bee workers. The study compared three treatments: flumethrin, OA-glycerine strips (OA-G), and OA trickling (OA-T). Twelve colonies were divided into four groups, with samples collected at five time points (0, 24, 48, 72, and 192 h). Physiological changes were assessed through markers of oxidative stress, longevity, and immune parameters. Exposure to oxalic acid via glycerine strips induced a humoral immune response in adult bees. The antimicrobial activity of hemolymph and levels of antimicrobial peptides (abaecin, apidaecin, defensin, and hymenoptaecin) were elevated between 48 and 192 h after OA-G treatment compared to the control group. In contrast, these parameters were not influenced by OA-T or flumethrin treatment. These findings suggest that OA-G strips activate the honey bee's immune system, providing insights into broader implications of OA use in beekeeping. It is crucial to determine whether the activation of humoral immune systems has positive or negative effects, as well as to develop standardized and reliable treatment protocols that ensure both - health of colonies and their effectiveness in controlling Varroa mite infestations.

Understanding the lethal and sublethal impacts of pesticides on biocontrol agents is crucial for the successful implementation of integrated pest management (IPM) programs. This study investigated the sublethal effects of fenpyroximate, a broad-spectrum acaricide/insecticide, on the fitness of Scolothrips longicornis Priesner (Thysanoptera: Thripidae), a key predator of the two-spotted spider mite, in controlled laboratory environments. Adult predators were exposed to pesticide residues on leaf discs to estimate parameters of concentration-mortality response models for females and males and calculate median lethal concentration (LC50) as well as sublethal concentrations (LC10, LC20 and LC30) used in subsequent bioassays. The estimated LC50 values for female and male predators were 18.32 and 15.49 µg a.i./mL, respectively. Results of sublethal concentrations experiments did not reveal any significant impact on the development of each stage or the survival rate of S. longicornis juveniles compared to those in the control group. However, the longevity of adult males and females was significantly lower at all sublethal concentrations than in the control. Moreover, the fecundity decreased significantly at all sublethal concentration treatments. With one exception (LC10), both the adult preoviposition period and total preoviposition period increased with increasing sublethal concentrations compared to those of the control. The shortest oviposition period (9.30 days) was observed at the LC30. For the life table parameters of S. longicornis females treated with sublethal concentrations, a significant decrease was found in the intrinsic rate of increase, net reproductive rate and finite rate of increase, whereas the mean generation time in the LC10 treatment was lower than that in the other treatments. This underscores the imperative need to consider sublethal concentration effects of fenpyroximate in the strategic design and implementation of IPM systems.

Lyme disease, a tick-borne illness caused by Borrelia spirochetes, poses a significant threat to public health. While acaricides effectively control ticks on pets and livestock, their impact on pathogen transmission is often unclear. This study investigated the acaricidal efficacy of fipronil against Ixodes ricinus ticks and its potential to block Borrelia afzelii transmission. Initially, we employed the ex vivo membrane blood-feeding system to assess the dose–response acaricidal activity of ivermectin, fipronil and its metabolite fipronil sulfone, when supplemented in the blood meal throughout tick feeding. To obtain the temporal resolution of their acaricidal activity, ticks were allowed to initiate blood feeding on an artificial membrane before being exposed to a 1-time topical application of these acaricides. Fipronil demonstrated superior speed of acaricidal activity, with onset of tick moribundity within a few hours, prompting its selection for further in vivo testing with Borrelia-infected ticks. The I. ricinus nymphs infected with B. afzelii were topically treated with fipronil shortly after attachment to mice. Four weeks post-feeding, the skin and internal organs were examined for the presence of Borrelia. No spirochetes were detected in any organ of mice exposed to fipronil-treated ticks, while 9 out of 10 control mice, exposed to non-treated infectious ticks, displayed Borrelia infection. The in vitro co-culture experiments confirmed that fipronil had no direct effect on Borrelia viability, indicating a tick-directed effect. Overall, these results underline the potential of fipronil as a valuable tool for tick control strategies and suggest a concept for acaricide-mediated Borrelia-transmission blockers.

In search of new sustainable biopesticides, we determined the phytochemical profiles, acaricidal and insecticidal properties of EOs distilled from the aerial parts of three Mexican Bursera species. Results were obtained by GC-MS analysis and three different bioassays, indicating that the EO of Bursera glabrifolia exhibited high relative abundancies of α-pinene, β-myrcene, and α-phellandrene, as well as promising pesticidal activity against Spodoptera littoralis larvae (LD50,90 = 32.4, 107.2 µg/larva), and Musca domestica (LD50,90 = 23.2, 103.2, and 13.5, 77.4 µg/female or male adult, respectively) and Tetranychus urticae adults (LD50,90 = 7.4, 30.3 µg/cm2). The Bursera lancifolia and Bursera linanoe samples contained mainly D-limonene or linalyl acetate and linalool, respectively, and showed generally less potent pesticidal properties (S. littoralis larva, LD50,90 = 45.4, 154.4 and 52.2, 158.7 µg/larva, respectively; female M. domestica adult, LD50,90 = 69.2, 210.9 and 45.1, 243.8 µg/female adult, respectively; T. urticae adults, LD50,90 = 20.7, 90.5 and 17.5, 71.4 µg/cm2, respectively). However, the EO of B. linanoe exhibited an especially pronounced activity against male M. domestica adults (LD50,90 = 10.6, 77.2 µg/male adult). Our findings prove the pesticidal potential of Mexican Bursera species in the context of integrated pest management (IPM) and highlight the importance of conducting further research to elucidate both the active principles and possibly existing synergistic effects.

The Varroa mite is one of the most important pests of honey bee colonies. This study compares the efficiency of thymol-based acaricides with the tau-fluvalinate-based commercial treatment to control this mite species. Two experiments were conducted: (1) in the Fall, thymol-based gel was compared with a widely used acaricide, fluvalinate-impregnated strips (Apistan®), and (2) in the Spring, gels containing thymol-only, thymol + menthol + eucalyptus essential oil, and thymol + thyme essential oil were compared with fluvalinate-impregnated strips. In the first experiment, both treatments reduced Varroa mite infestation levels significantly after 70 days of their application compared to the control treatment. The results of the second experiment showed that thymol + thyme essential oil gel, thymol + menthol + eucalyptus essential oil gel, and Apistan strips had always higher efficiency on decreasing mite infestation levels of the selected colonies compared to the control treatment and thymol-only gel during their 7-week application. The results of this study demonstrate that plant-based acaricides for controlling Varroa mites in honey bee colonies have similar efficiency compared to the chemical control methods. Therefore, it is better to use these selected acaricides to reduce mite resistance to chemical control treatments in honey bee colonies.

Ticks attaching to ear canals of humans and animals are the cause of otoacariasis, common in rural areas of Nepal. The plant Clerodendrum viscosum is used in multiple indigenous systems of medicine by ethnic communities in the Indo-Nepali-Malaysian region. Visiting the Chitwan National Park, we learned that in indigenous medicine, flower extract of C. viscosum is utilized to treat digestive disorders and extracts from leaves as tick repellent to prevent ticks from invading or to remove them from the ear canal. The objective of our study was to provide support to indigenous medicine by characterizing the in vivo effect of leave extracts on ticks under laboratory conditions and its phytochemical composition. We collected plant parts of C. viscosum (leaves and flowers) and mango (Mangifera indica) leaves at the Chitwan National Park, previously associated with repellent activity to characterize their effect on Ixodes ricinus ticks by in vivo bioassays. A Q-ToF high-resolution analysis (HPLC-ESI-QToF) was conducted to elucidate phenolic compounds with potential repellent activity. Clerodendrum viscosum and M. indica leaf extracts had the highest tick repellent efficacy (%E = 80-100%) with significant differences when compared to C. viscosum flowers extracts (%E = 20-60%) and phosphate-buffered saline. Phytochemicals with tick repellent function as caffeic acid, fumaric acid and p-coumaric acid glucoside were identified in C. viscosum leaf extracts by HPLC-ESI-QToF, but not in non-repellent flower extracts. These results support the Nepali indigenous medicine application of C. viscosum leaf extracts to repel ticks. Additional research is needed for the development of natural and green repellent formulations to reduce the risks associated with ticks resistant to acaricides.

Ticks can seriously affect human and animal health around the globe, causing significant economic losses each year. Chemical acaricides are widely used to control ticks, which negatively impact the environment and result in the emergence of acaricide-resistant tick populations. A vaccine is considered as one of the best alternative approaches to control ticks and tick-borne diseases, as it is less expensive and more effective than chemical controls. Many antigen-based vaccines have been developed as a result of current advances in transcriptomics, genomics, and proteomic techniques. A few of these (e.g., Gavac® and TickGARD®) are commercially available and are commonly used in different countries. Furthermore, a significant number of novel antigens are being investigated with the perspective of developing new anti-tick vaccines. However, more research is required to develop new and more efficient antigen-based vaccines, including on assessing the efficiency of various epitopes against different tick species to confirm their cross-reactivity and their high immunogenicity. In this review, we discuss the recent advancements in the development of antigen-based vaccines (traditional and RNA-based) and provide a brief overview of recent discoveries of novel antigens, along with their sources, characteristics, and the methods used to test their efficiency.

The aim of this study was to determine the presence of zoocide (insecticide and miticide) residues in hops collected in three hop-growing regions located in the Czech Republic, and to assess their zoocide profiles and residue levels in terms of variability in temperature and precipitation across the 2018-2020 seasons. Furthermore, the weather factors that influenced the occurrence of hop pests are described and discussed. During our 3-year survey, a total of 120 samples of whole-cone hops samples harvested in three hop-growing regions were analysed for the presence of 29 insecticides and miticides using the modified QuEChERS extraction method, followed by liquid chromatography-tandem mass spectrometry and gas chromatography-tandem mass spectrometry. A majority, 119 of 120 samples, contained a residue of at least one of the active substances surveyed in this study, and 34 analysed samples contained multiple residues with three to four zoocides presented. Concerning the most frequently detected zoocide residues, spirotetramat and/or its metabolites were found in 94.2% of the samples at levels ranging from 0.02 to 1.08 mg/kg. Of the other zoocides surveyed, residues of fenpyroximate, hexythiazox, bifenazate and lambda-cyhalothrin were routinely found in hop cone samples. Obtained data were then used for evaluating seasonal and geographical variations in the profile of zoocide residues among the hop-growing regions in the years 2018-2020, and the compliance with legal regulations concerning the use of zoocides on hops was ascertained. The results showed that (1) the profile and levels of zoocide residues found in the samples reflected seasonal prevalence of pest infestation on hop plants; (2) the strategy to control pests (especially aphids) used in most of hop yards was consistent across the seasons; and (3) a concentration of spirotetramat residues less than 1 mg/kg was typical for hops grown in the Czech Republic.

It has been demonstrated that impairing protein synthesis using drugs targeted against tRNA amino acid synthetases presents a promising strategy for the treatment of a wide variety of parasitic diseases, including malaria and toxoplasmosis. This is the first study evaluating tRNA synthetases as potential drug targets in ticks. RNAi knock-down of all tested tRNA synthetases had a strong deleterious phenotype on Ixodes ricinus feeding. Our data indicate that tRNA synthetases represent attractive, anti-tick targets warranting the design of selective inhibitors. Further, we tested whether these severely impaired ticks were capable of transmitting Borrelia afzelii spirochaetes. Interestingly, biologically handicapped I. ricinus nymphs transmitted B. afzelii in a manner quantitatively sufficient to develop a systemic infection in mice. These data suggest that initial blood-feeding, despite the incapability of ticks to fully feed and salivate, is sufficient for activating B. afzelii from a dormant to an infectious mode, enabling transmission and dissemination in host tissues.

1. The aim was to evaluate the acaricidal effects of pure active components of essential oils against poultry red mite (Dermanyssus gallinae) as an alternative to chemical acaricides (organophosphates and pyrethroids).2. The toxicities of five pure active components of essential oils (eugenol from clove bud, eucalyptol from rosemary, limonene from citrus fruits, linalool from lavender and cinnamaldehyde from cinnamon) were tested on D. gallinae females in an impregnated paper assay.3. The active substances were dissolved in water and Tween 20 and applied at concentrations ranging from 0.002 to 0.06 µl/cm2. Toxicity was expressed as a lethal dose (LD50 or LD90).4. The highest mortality was observed with eugenol. The LD90 was estimated to be 5.1 µg/cm2 for this substance, followed by cinnamaldehyde, the LD90 of which was estimated to be 11.0 µg/cm2. Limonene and eucalyptol were generally less effective in controlling D. gallinae.
