Navržené testování účinku serotonergní neurotransmise v animálním modelu schizofrenie na modelované positivní a negativní symptomy a kognitivní deficit přispějě k objasnění mechanismu Vlev serotoninu v účinku atypických antipsychotik v animálním modelu schizofrenie se bude řešit pomocí antagonisty 5-HT2A a agonisty 5-HT1A receptoru jednotlivě podaných nebo v kombinaci s D2 antagonistou ( haloperidolem). Vliv serotonergní modulace v účincích antipsychotik v animálním modelu schizofrenie bude řešen i hodnocením výskytu proteinu FOS ve vybraných oblastech mozku a výsledky budou porovnány s vybranými atypickými a typickými antipsychotiky. Vlev serotoninu v účincích APDs v animálním modelu schizofrenie bude řešen také pomocí stanovení změn hladin dopaminu ve vybraných oblastech mozku. Tyto změny budou porovnány s výsledky získanými po podání antipsychotik.; The study of the role of serotonin in APDs treatment will be based on a pharmacological animal model of schizophrenia. Explanation of the effect of serotonergic system in the treatment of schiz. - like behavior can clarify the function of atypical APD. The role of serotonin activity in atypical APDs will be tested by means of 5-HT2A antagonists and 5HT1A agonists administration alone or combined with D2 antagonist (haloperidol). The results will be compared with atypical APDs ( quetiapin risperid). Theeffect od serotonin modulation in atypical APDs will be evaluated by the testing of c-fos expression ( immunocytochemistry) in the prefrontal cortex and the striatum. The results will be compared with the effect of atypical APDs. The effect of serotoninmodulation on dopamine levels in the prefrontal cortex and striatum will be evaluated using the microdialysis and the chromatography ( HPLC) The results will be compared with the effect of atypical APDs ( quetiapin, risperidon).
