The aim of the study is to investigate serum tissue transglutaminase antibodies (tTG) IgG and IgA in patient with liver injury of various reasons including hepatocelullar carcinoma. Approximately five hundred patients will be included into the study. Statistical analysis will be performed using non-parametric Mann-Whitney test. Analysis of endomysial antibodies will be performed in patients with positive results to tTG. Analysis of antibodies specificity against tTG could an important marker in auto reactivity of patients with liver injury and celiac disease. The other aim of our study is characterization of antigen epitopes of tTG recognized by antibodies of IgA isotype in patients with liver disease using a PEPSCAN method. Our study will include thepatients after liver transplantation as well. In patients with confirmed celiac disease a liver biopsy and duodenal mucosal biopsy with imunohistochemical evaluation will be performed.

Cílem projektu je stanovit souvislost mezi jaterními onemocnění a celiakií u jinak asymptomatických jedinců jako projev extraintestinální manifestace celiakie v souboru pacientů IKEM jak před tak i po transplantaci jater včetně sledování efektu bezlepkové diety. Zaměříme se na analýzu detailní specificity autoprotilátek proti tkáňové transglutamináze k odhalení možných příčin autoreaktivity pacientů s jaterními onemocněním a floridní celiakií. Součástí bude i imunohistochemické vyšetření bioptických vzorků duodena a jater u indikovaných jedinců. Dalším cílem je charakterizace antigenních epitopů tkáňové transglutaminázy rozpoznávaných sérovými IgA protilátkami pacientů s aktivní celiakií a séropozitivních pacientů s hepatopatiemi.
