Obsahem článku je prezentace výsledků výzkumu, v jehož rámci byly zjišťovány souvislosti mezi negativními zkušenostmi v dětství a dospívání a následnými potížemi se společenským uplatněním. Stejná souvislost byla zjišťována ve vztahu k rozvoji antisociálních forem chování. Šetření byly podrobeny vybrané skupiny delikventní mládeže. Dosažené výsledky ukazují na statisticky významné souvislosti mezi traumatizací a následně sníženými adaptačnímu schopnostmi. Konstatujeme, že byly prokázány důsledky negativního vývoje v dětství a dospívání, které spočívají v celkově snížených schopnostech se později ve společnosti uplatňovat sociálně přijatelným způsobem. Tyto potíže dokonce mohou ústit až v typické příznaky specifických poruch osobnosti, a také v maladaptaci, kdy způsob seberealizace spočívá u šetřených osob v jiných typech strategií. Tyto strategie jsou pro normální společenské fungování nepotřebné. Při dosahování potřeb se bohužel jedná často o antisociální formy chování. Ze zjištění vyplývá, že traumatizace se může stát významnou příčinou nejen delikvence, ale také příčinou rozvoje specifické poruchy osobnosti. Zjištění a výsledky jsou využitelné pro forenzní psychologickou praxi. Jsou využitelné pro diagnostiku, a pro následnou terapeutickou práci s delikventní mládeží.
The article is an analysis of antisocial behaviour of delinquent youth.The relationships between negative events in childhood and adolescence, and subsequent problems in self-realization were studied. The relationships were analyzed in relation to the development of forms of antisocial behaviour. Psychological characteristics in the selected groups of juvenile delinquents and a control group were compared. Methods based on RSTI (Risk Sophistication Treatment Inventory), HARE PCT (Hare Psychopathy Checklist), and TSC 40 (Trauma Symptom Checklist) were used. Statistically significant differences were observed between trauma and the subsequently lower adaptation skills and development of typical symptoms of specific personality disorders. The findings suggest that negative events in childhood and adolescence may be a significant predictor of personality disorders in adulthood. The findings are in accordance with previous studies. Clinicians and researchers should be highly aware of stress and trauma in childhood as a risk factor for later psychopathological development.

Osamělost je nepříjemný subjektivní stav vznikající v důsledku nedostatečného zapojení do sociálních vztahů. Má různé formy: sociální a emoční. Sociální osamělost souvisí se samotou a je způsobena absencí vztahů především s přáteli a známými. Emoční osamělost vzniká v důsledku absence blízkého vztahu založeného na vzájemném porozumění. Jedná se o subjektivní prožitek, který se vyskytuje během celého života, problémem se stává, pokud je chronický. Základním nástrojem hodnocení osamělosti jsou škály. Nejčastěji používanou je UCLA Škála osamělosti, která má vysokou validitu a velmi dobrou reliabilitu. Existuje celá řada teorií osamělosti, nejrespektovanější současný model vychází z kognitivní teorie. Osamělost je rizikovým faktorem pro vznik somatických a duševních poruch, vede nejen k vyšší nemocnosti, ale i úmrtnosti. Je spojena především s depresí, nedostatečnou kvalitou spánku, úzkostí, poruchami paměti a schizofrenií, také narušuje kognitivní funkce. Zvyšuje riziko vysokého krevního tlaku a kardiovaskulárních onemocnění, zhoršuje funkci imunitního systému. U dětí může být nedostatek sociálního kontaktu prediktorem pozdější deprese, antisociálního chování, sebepoškozování či suicidálního chování. Byly prokázány genetické souvislosti. Na základě rozsáhlé metaanalýzy intervencí zaměřené na redukování osamělosti lze za nejúčinnější považovat intervence zaměřené na maladaptivní sociální kognice, tj. kognitivně-behaviorální terapii, i když zatím v této oblasti není široce rozšířena. Vhodné by proto byly další studie k prokázání efektivity.
Loneliness is an individually unpleasant state on both, social and emotional levels, coming up in consequence of insufficient social bonds. Social loneliness is interconnected with loneliness as such and is caused by the absence of relationships with friends and other people. Emotional loneliness is caused by the absence of a close relationship based on mutual understanding. Loneliness is a subjective, individual state occurring throughout the life; it tends to be considered a problem when it becomes chronic. One of the basic assessment methods of loneliness is scales. The UCLA Loneliness Scale is the most frequently used one due to its high validity and appropriate reliability. There are a number of theories on loneliness. The most respected one is based on cognitive theory. Loneliness is a risk factor in developing somatic and mental diseases; it contributes both to higher sickness and death rates. Loneliness tends to be accompanied by depression, low quality of sleep, anxiety, memory impairment, schizophrenia. It causes cognitive impairment, increases the risk of high blood pressure and cardiovascular diseases; it causes immune deficiency. In children, the lack of social contact may become a predictor of later depression, dissocial behaviour, self-injury, and suicidal behaviour. Genetic causal links were proved. The most effective, based on an extensive meta-analysis of intervention methods aimed at the reduction of loneliness, proved to be interventions aimed at changing maladaptive social cognitive patterns, i.e. cognitive-behavioural therapy even though it has not been widely used in this field. Thus, other studies to prove its efficiency are needed.

Porucha pozornosti s hyperaktivitou – ADHD (Attention Deficit Hyperactivity Disorder) představuje široce rozšířenou neurobiologickou poruchu v dětském věku s poměrně vysokou mírou genetické podmíněnosti. Heritabilita symptomů ADHD je uváděna až na úrovni 75 %. Mechanizmus genetického přenosu však stále není zcela objasněn. Největší pozornost je v současné době zaměřena na geny dopaminergního a serotoninergního systému, ale i řadu genů dalších. Článek přináší souhrnné informace o nejvýznamnějších genetických asociacích u ADHD.
Attention deficit hyperactivity disorder (ADHD) is a common, probably highly genetically conditioned, neurobiological disorder. Heritability of ADHD symptoms is up to about 75%. However, the mechanism is still not fully understood. Attention is focused mainly on genes of the dopaminergic and serotonergic system as well as many other genes. The paper presents a summary of the most significant genetic associations with ADHD.

Attention deficit hyperactivity disorder (ADHD) is one of the most commonly diagnosed childhood psychiatric disorders. It is manifested in every part of an affected child's behavior, with multiple symptomatology and heterogenous etiology. Published studies report that ADHD children may show changes in growth and development. Most of the studies on ADHD have been focused on connections between medication and growth changes and describe growth delays associated with medication. However, recent research results point to the low significance of the changes accompanying pharmacological treatment. Changes in growth may not only be a secondary effect of the treatment, but may also be specific characteristics of ADHD.

Understanding the control of gene expression is critical for our understanding of the relationship between genotype and phenotype. The need for reliable assessment of transcript abundance in biological samples has driven scientists to develop novel technologies such as DNA microarray and RNA-Seq to meet this demand. This review focuses on comparing the two most useful methods for whole transcriptome gene expression profiling. Microarrays are reliable and more cost effective than RNA-Seq for gene expression profiling in model organisms. RNA-Seq will eventually be used more routinely than microarray, but right now the techniques can be complementary to each other. Microarrays will not become obsolete but might be relegated to only a few uses. RNA-Seq clearly has a bright future in bioinformatic data collection.
BACKGROUND: Attention deficit hyperactivity disorder (ADHD) is a neurological/behavioral disorder characterized by inattention or hyperactivity and impulsivity, or combined symptomatology. Children with ADHD are predisposed to irregular and/or impulsive eating patterns often leading to compromised physical condition. The goal of the present study was to statistically evaluate parental scoring of patterned eating behaviors and associated lifestyles within a cohort of 100 boys diagnosed with ADHD in comparison to age-matched male controls. MATERIAL AND METHODS: The study population consisted of 100 boys aged 6-10 years diagnosed with mixed type ADHD by DSM-IV criteria and 100 aged-matched healthy male control subjects. Patterns of eating behaviors and associated lifestyles were scored by structured parental interviews using a nominal rating scale. RESULTS: Interview scores indicated statistically significant differences in patterned eating behaviors in subjects with ADHD in comparison to healthy controls. Notably, subjects diagnosed with ADHD exhibited markedly diminished adherence to a traditional breakfast, lunch, and dinner schedule, which was linked to a significantly higher frequency (>5/day) of irregular eating times. In the ADHD cohort, disruptive patterns of eating behaviors were associated with diminished nutritional value of ingested food (expressed as lowered content of fruits and vegetables) and increased consumption of sweetened beverages. CONCLUSIONS: Disruptive patterns of eating behaviors, metabolically unfavorable nutritional status, and diminished physical activities of male children diagnosed with ADHD are linked to compromised growth and development and appearance of metabolic diseases in adulthood.

By operational criteria, burnout appears to be a multifaceted behavioral syndrome consisting of maladaptive individual responses subsequent to prolonged stressful situations. Given the intense physical and cognitive demands of providing high quality healthcare to a wide spectrum of patients, healthcare professionals are particularly susceptible to developing burnout syndrome, a notable phenomenon that has gleaned significant societal attention in recent years. Clearly, widespread manifestation of burnout by health care professionals represents a serious potential threat to the overall quality of patient care and to the realization of positive outcomes to multiple treatment strategies. It will most certainly engender a serious negative impact on the economic viability of the entire healthcare system. Presently, our brief review focuses on current research efforts to 1) provide precise behavioral and psychiatric diagnostic criteria for burnout syndrome in healthcare professionals, 2) identify potential etiological factors and ongoing stressors, and 3) outline an integrative approach for treatment and prevention.

A wealth of preclinical and clinical literature has established functional associations of CNS dopamine (DA) and its multiple G protein-coupled receptor (GPCR) types in the integration of key neurological processes linked to complex behavioral activities. Conversely, an equivalent vast literature supports the role of aberrant CNS DA expression and DA receptor signaling in the etiology and persistence of major psychiatric illnesses and has established selective targeting of DA-ergic systems as a cornerstone of pharmacotherapeutic intervention and current neuroleptic drug development. The present short review focuses on potential functional/behavioral alterations linked to polymorphisms in the primary DNA sequence of the DA receptor type 4 (DRD4) gene in reference to major psychiatric illnesses. The potential clinical relevance of major polymorphisms of the DRD4 gene are discussed within the context of practical aspects of typical and atypical neuroleptic drug usage within afflicted populations of psychiatric patients. It is anticipated that additional complementary molecular, biochemical, and behavioral studies of DRD4 gene polymorphisms will provide essential information for selective targeting of heterogeneous populations of CNS D4 receptors and advance drug discovery and therapeutic development efforts for highly efficacious treatment of psychiatric illnesses.
