PURPOSE: Fruit and vegetable intake is inversely correlated with cancer; thus, it is proposed that an extract of phytochemicals as present in whole fruits, vegetables, or grains may have anti-carcinogenic properties. Thus, the anti-tumour effects of fruit peel polyphenols (Flavin7) in the chemoprevention of N-methyl-N-nitrosourea-induced mammary carcinogenesis in female rats were evaluated. METHODS: Lyophilized substance of Flavin7 (F7) was administered at two concentrations of 0.3 and 3 % through diet. The experiment was terminated 14 weeks after carcinogen administration, and mammary tumours were removed and prepared for histopathological and immunohistochemical analysis. In addition, using an in vitro cytotoxicity assay, apoptosis and proliferation after F7 treatment in human breast adenocarcinoma (MCF-7) cells were performed. RESULTS: High-dose F7 suppressed tumour frequency by 58 % (P < 0.001), tumour incidence by 24 % (P < 0.05), and lengthened latency by 8 days (P > 0.05) in comparison with the control rats, whereas lower dose of F7 was less effective. Histopathological analysis of tumours showed significant decrease in the ratio of high-/low-grade carcinomas after high-dose F7 treatment. Immunohistochemical analysis of rat carcinoma cells in vivo found a significant increase in caspase-3 expression and significant decrease in Bcl-2, Ki67, and VEGFR-2 expression in the high-dose group. Both doses demonstrated significant positive effects on plasma lipid metabolism in rats. F7 significantly decreased survival of MCF-7 cells in vitro in MTT assay by dose- and time-dependent manner compared to control. F7 prevented cell cycle progression by significant enrichment in G1 cell populations. Incubation with F7 showed significant increase in the percentage of annexin V-/PI-positive MCF-7 cells and DNA fragmentation. CONCLUSIONS: Our results reveal a substantial tumour-suppressive effect of F7 in the breast cancer model. We propose that the effects of phytochemicals present in this fruit extract are responsible for observed potent anti-cancer activities.

Úvod a ciele: Epidermálny rastový hormón (EGF) podporuje bunkovú proliferáciu a je implementovaný v procese embryogenézy, angiogenézy a bunkovej diferenciácie. EGF-EGFR dráha má pravdepodobne kľúčovú úlohu v rozvoji kastračne rezistentného karcinómu prostaty. Funkčný genetický variant v EGF géne bol spojený s náchylnosťou na vznik nádorových ochorení. Cieľom predkladanej štúdie bolo testovanie potenciálnej prognostickej a prediktívnej úlohy EGF funkčného genetického variantu A61G u pacientov s karcinómom prostaty v slovenskej populácii. Materiál a metódy: U 172 pacientov s karcinómom prostaty a 193 zdravých jedincov sme genotypovali jednonukleotidový polymorfizmus (SNP) EGF s využitím polymerázovej reťazovej reakcie s následnou reštrikčnou analýzou (PCR-RFLP). Výsledky: Nezistili sme signifikantnú asociáciu medzi frekvenciami EGF genotypov a rizikom vzniku karcinómu prostaty. Nepotvrdila sa ani korelácia medzi EGF genotypmi a Gleansonovým skóre. Záver: Výsledky našej štúdie nepotvrdili vzťah medzi EGF polymorfizmom a rozvojom karcinómu prostaty v slovenskej populácii. V budúcnosti by boli potrebné ďalšie štúdie sledujúce vzťah medzi EGF genotypmi, environmentálnymi faktormi, ako i liečebným prístupom a rizikom vzniku karcinómu prostaty.
Introduction and Aims: Epidermal growth hormone (EGF) promotes cell proliferation and is implemented in the process of embryogenesis, angiogenesis, and cell differentiation. EGF-EGF receptor pathway probably has a key role in hormone-resistant prostate cancer stage. Functional genetic variants in the EGF gene were associated with susceptibility to cancer formation. The aim of our pilot study was to investigate the potential prognostic and predictive role of EGF A61G functional genetic variant in patients with prostate cancer in the Slovak population. Material and Methods: We genotyped single nucleotide polymorphism (SNPs) in EGF among 172 prostate cancer patients and 193 healthy controls by using PCR-RFLP (polymerase chain reaction – restriction fragment length polymorphism analysis). Results: We observed no significant association between EGF genotype frequencies and prostate cancer risk. There is no correlation between EGF genotypes and Gleanson score. Conclusions: The results of our pilot demonstrate that EGF polymorphism is not associated with the risk of developing prostate cancer in the Slovak population. Future studies concerning the association of EGF genotypes and environmental factors as well as the therapeutic approach will be important to clarify if the EGF polymorphism may contribute to susceptibility to prostate cancer.
Úvod a ciele: Enzým N-acetyltransferáza 2 (NAT2) sa podieľa na detoxikácii širokého spektra prirodzene sa vyskytujúcich xenobiotík, zahŕňajúc karcinogény a lieky. Variácie v NAT2 géne spájajúce sa s acetylačnou kapacitou „pomalí“ alebo „rýchli“ metabolizéri modifikujú náchylnosť na vznik nádorových ochorení a škodlivé reakcie liekov. Cieľom našej pilotnej štúdie bolo sledovať asociáciu medzi NAT2 genotypmi/fenotypmi a zvýšeným rizikom vzniku karcinómu prostaty v slovenskej populácii, ako aj štúdium klinickopatologických korelácií a prognostického významu NAT2. Materiál a metódy: U 179 pacientov s karcinómom prostaty a 269 zdravých jedincov sme genotypovali 4 jednonukleotidové polymorfizmy (SNPs) NAT2 s využitím polymerázovej reťazovej reakcie s následnou reštrikčnou analýzou (PCR-RFLP). Chi- kvadrátový test sme použili na odhad relatívneho rizika karcinómu prostaty s NAT2 genotypmi/NAT2 acetylačnými fenotypmi. Výsledky: Nezistili sme signifikantnú asociáciu medzi NAT2 genotypmi/fenotypmi a rizikom vzniku karcinómu prostaty. U rýchlych acetylátorov sme zistili signifikantne vyššie hladiny sérového PSA v porovnaní s pomalými acetylátormi, predpokladajúc zvýšenú tvorbu karcinogénov a modifikáciu tumorigenézy. Záver: Výsledky našej pilotnej štúdie nepotvrdili vzťah medzi NAT2 polymorfizmom a rozvojom karcinómu prostaty v slovenskej populácii. V budúcnosti by preto boli potrebné ďalšie štúdie sledujúce reálny význam a vzťah medzi NAT2 genotypmi a environmentálnymi faktormi/životným štýlom a rizikom vzniku karcinómu prostaty.
Introduction and Aims: The N-acetyltransferase 2 (NAT2) enzyme detoxifies a wide spectrum of naturally occurring xenobiotics including carcinogens and drugs. Variation at the NAT2 gene has been linked to the human acetylation capacity, either “slow” or “fast”, which modifies susceptibility to cancer and adverse drug reactions. The aim of our pilot study was to investigate whether NAT2 genotypes/phenotypes are associated with increased risk factor for prostate cancer in the Slovak population and to study the clinico-pathological correlations and the prognostic significance of NAT2. Material and Methods: We genotyped four common single nucleotide polymorphisms (SNPs) in NAT2 among 179 prostate cancer patients and 269 healthy controls by using PCR-RFLP (polymerase chain reaction restriction fragment length polymorphism analysis). Relative risk of prostate cancer for NAT2 genotypes/NAT2 acetylation phenotypes was estimated using chi squared method. Results: We observed no significant association between NAT2 genotypes/phenotypes and prostate cancer risk. We have found that rapid acetylators have significantly higher serum PSA levels than slow acetylators (p < 0.05), indicating increase generation of carcinogens and enhance tumourogenesis. Conclusions: Data of our pilot study demonstrate that the NAT2 polymorphism is not associated with the risk of developing prostate cancer in the Slovak population. Future studies concerning the association of NAT2 genotypes and environmental/lifestyle factors will be important to elucidate the real meaning of NAT2 polymorphisms in the susceptibility to prostate cancer.
Prostatická žľaza je bohatá na peptidové rastové faktory, ktoré hrajú dôležitú úlohu v intracelulárnych procesoch, ako je napr. bunkový rast, diferenciácia, angiogenéza a apoptóza. Významnou mierou prispievajú v procese karcinogenézy k malígnej transformácii. Medzi najzávažnejšie ochorenia prostaty patrí karcinóm prostaty (CaP), ktorý postihuje mužov najmä v staršom veku. Predkladaný prehľadový článok zhŕňa údaje a zovšeobecňuje poznatky vyplývajúce z úlohy peptidového rastového faktora VEGF a jeho receptorov vo vývoji karcinómu prostaty, ako i sledovanie potenciálnej prognostickej a prediktívnej úlohy polymorfizmov v géne VEGF u pacientov s karcinómom prostaty.
The prostate gland is abundant in peptide growth factors which play an important role in intracellular processes such as cell growth, differentiation, angiogenesis and apoptosis. They also contribute significantly to the malignant transformation during carcinogenesis process. The most common prostate disease is prostate cancer (CaP) that mainly affects men in old age. The aim of this review is to summarize the current knowledge about the role of VEGF and its receptors in the development of prostate cancer as well as monitoring of potential prognostic and predictive role of gene polymorphisms of VEGF in patients with prostate cancer.
