A fast method for the determination of tropane alkaloids, using a portable CE instrument with a capacitively coupled contactless conductivity detector (CE-C4D) was developed and validated for determination of atropine and scopolamine in seeds from Solanaceae family plants. Separation was obtained within 5 min, using an optimized background electrolyte consisting of 0.5 M acetic acid with 0.25% (w/v) β-CD. The limit of detection and quantification was 0.5 µg/mL and 1.5 µg/mL, respectively, for both atropine and scopolamine. The developed method was validated with the following parameters-precision (CV): 1.07-2.08%, accuracy of the assay (recovery, RE): 101.0-102.7% and matrix effect (ME): 92.99-94.23%. Moreover, the optimized CE-C4D method was applied to the analysis of plant extracts and pharmaceuticals, proving its applicability and accuracy.
In the search for new treatments for complex disorders such as Alzheimer's disease the Multi-Target-Directed Ligands represent a very promising approach. The aim of the present study was to identify multifunctional compounds among several series of non-imidazole histamine H3 receptor ligands, derivatives of 1-[2-thiazol-5-yl-(2-aminoethyl)]-4-n-propylpiperazine, 1-[2-thiazol-4-yl-(2-aminoethyl)]-4-n-propylpiperazine and 1-phenoxyalkyl-4-(amino)alkylopiperazine using in vitro and in vivo pharmacological evaluation and computational studies. Performed in vitro assays showed moderate potency of tested compounds against acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). Molecular modeling studies have revealed possible interactions between the active compounds and both AChE and BuChE as well as the human H3 histamine receptor. Computational studies showed the high drug-likeness of selected compounds with very good physicochemical profiles. The parallel artificial membrane permeation assay proved outstanding blood-brain barrier penetration in test conditions. The most promising compound, A12, chemically methyl(4-phenylbutyl){2-[2-(4-propylpiperazin-1-yl)-1,3-thiazol-5-yl]ethyl}amine, possesses good balanced multifunctional profile with potency toward studied targets - H3 antagonist activity (pA2 = 8.27), inhibitory activity against both AChE (IC50 = 13.96 μM), and BuChE (IC50 = 14.62 μM). The in vivo pharmacological studies revealed the anti-amnestic properties of compound A12 in the passive avoidance test on mice.

Na základě poznatků získaných z krystalografie acetylcholinesterasy (AChE) byly koncipovány nové homo- i heterodimerní molekuly, které jsou schopné vázat se do obou anionických míst enzymu a tím nejen zvýšit svůj inhibiční potenciál, ale i současně ovlivňovat další patologické mechanismy AD. Průkopnickou molekulou se v tomto ohledu stal bis(7)-takrin, který vznikl zdvojením takrinu přes alifatický můstek o délce sedmi uhlíkových atomů. Bis(7)-takrin je zajímavou multipotentní sloučeninou, která je schopna ovlivňovat řadu receptorových i enzymových systémů za podmínek in vitro i in vivo. Stal se rovněž předlohovou strukturou pro celou řadu nových sloučenin s dalšími pozitivními vlastnostmi, které jsou relevantní pro terapii AD.
The acetylcholinesterase (AChE) structure elucidation has significantly contributed to the development of both novel homo- and heterodimer molecules that are capable of simultaneous binding to both anionic sites of the enzyme. This mostly resulted into improved AChE inhibition potency concurrently affecting other pathological hallmarks of the disease. In this regard, bis(7)-tacrine can be considered as the pioneering molecule. It consists of two tacrine moieties connected via alkylene tether of seven carbon atoms. In in vitro and in vivo, bis(7)-tacrine revealed interesting multipotent profile capable to affect several enzymes and receptors which are implicated in the pathogenesis of AD. It also became a key structural template in the development of other more potent compounds intended for the therapy of neurodegenerative disorders.

RATIONALE: Development of new drugs for treatment of Alzheimer's disease (AD) requires valid paradigms for testing their efficacy and sensitive tests validated in translational research. OBJECTIVES: We present validation of a place-navigation task, a Hidden Goal Task (HGT) based on the Morris water maze (MWM), in comparable animal and human protocols. METHODS: We used scopolamine to model cognitive dysfunction similar to that seen in AD and donepezil, a symptomatic medication for AD, to assess its potential reversible effect on this scopolamine-induced cognitive dysfunction. We tested the effects of scopolamine and the combination of scopolamine and donepezil on place navigation and compared their effects in human and rat versions of the HGT. Place navigation testing consisted of 4 sessions of HGT performed at baseline, 2, 4, and 8 h after dosing in humans or 1, 2.5, and 5 h in rats. RESULTS: Scopolamine worsened performance in both animals and humans. In the animal experiment, co-administration of donepezil alleviated the negative effect of scopolamine. In the human experiment, subjects co-administered with scopolamine and donepezil performed similarly to subjects on placebo and scopolamine, indicating a partial ameliorative effect of donepezil. CONCLUSIONS: In the task based on the MWM, scopolamine impaired place navigation, while co-administration of donepezil alleviated this effect in comparable animal and human protocols. Using scopolamine and donepezil to challenge place navigation testing can be studied concurrently in animals and humans and may be a valid and reliable model for translational research, as well as for preclinical and clinical phases of drug trials.

Plant tropane alkaloids atropine and scopolamine are very strong anticholinergic drugs used in medicine and also found their place in military medicine, in particular atropine as an antidote in case of poisonings by nerve gases. Atropine and scopolamine both easily penetrate the blood-brain barrier and may cause central nervous system toxicity. Its symptoms are delirium, restlessness, confusion, and impairment of mental capacities. Scopolamine is much more likely to produce sedation and amnesia than atropine. Synthetic atropine-like compounds are also strong anticholinergics, and found use as a modern type of chemical weapons that incapacitate enemy for some time, but do not kill him. They are so-called non-lethal weapons, which are able to temporarily disable personnel from combat action and to protect the environment without limiting desired negative consequences to the enemy’s national economy. Such non-lethal weapons is Agent BZ.

Lidské chování je velmi složité, zahrnuje širokou škálu projevů od jednoduché reflexní aktivity přes smyslovou percepci, motorickou koordinaci až po kognitivní procesy. Podílí se na něm celá řada vnějších a vnitřních faktorů. Z vnitřních faktorů jsou to především neurotransmitery a hormony, ze zevních faktorů hlavně sociální vlivy a prostředí. Také chemické prvky, např. brom, fluor a lithium, ovlivňují naše chování. Chování je pod vlivem prakticky všech hormonů a neurotransmiterů – hormonů štítné žlázy nebo kůry nadledvin – a sexuální chování je ovlivňováno estrogeny, progesteronem, testosteronem nebo oxytocinem. Léčiva zasahují do našeho chování ovlivněním různých neurotransmiterů. Také běžně užívané přípravky proti hypertenzi ovlivňují sníže-ním tonu sympatiku naše chování. Po aplikaci některých intravenózních anestetik se setkáváme s výskytem tzv. amorózního chování, kdy zejména u žen dochází desinhibicí podkoří k chování se sexuálním obsahem. Již od pradávna jsou používána četná afrodiziaka sloužící k překonání sexuálních zábran žen. Jsou to především alkohol, některé drogy, např. yohimbin nebo španělské mušky nebo Pantokrin. Date rape drugs je pojmenování pro farmaka, která mají usnadnit sexuální násilí. Patří mezi ně především benzodiazepiny, γ hydroxybutyrát, ketamin a v Latinské Americe také scopolamin. V posledních letech vstupuje do hry také neuropeptid oxytocin, který má vedle periferních účinků na prsní žlázu a dělohu také centrální účinky projevující se anxiolýzou, zvýše-ním empatie a redukcí sexuálního studu. Tyto jeho farmakologické účinky mohou být u některých žen zneužity jako tzv. date rape drugs.
Human behavior is a complex phenomenon, comprising a wide range of manifestations from simple reflex activity, sensory perception, motor coordination to cognitive processes. It is influenced by many external and internal factors. The most important internal factors are neurotransmitters and hormones, the crucial external factors being the influence of society and environment. Chemical elements such as bromine, fluorine, and lithium, also influence how we behave. Our behavior is modified by practically all hormones and neurotransmitters, especially thyroid or adrenal hormones; sexual behavior is influenced by estrogens, progesterone, testosterone or oxytocin. Drugs modify our behavior through various neurotransmitters. Common antihypertensives lower the sympathetic tone, which can also be reflected in behavioral changes. Some intravenous anesthetics induce so called amorous behavior, especially in women – in these cases, disinhibition of subcortical structures leads to behavior with sexual content. For ages, many aphrodisiacs have been used to overcome sexual scruples in women. These are especially alcohol, some drugs such as yohimbine, Spanish flies or Pantocrine. Date rape drugs are medications making sexual violence easier: benzodiazepines, γ hydroxybutyrate, ketamin, and – in Latin America – scopolamine. Lately, neuropeptide oxytocin also “joined the play”, exhibiting not only peripheral effects on the breast and womb but also central effects: anxiolysis, increased empathy, and reduction of sexual shame. Owing to these pharmacological characteristics, these substances can be abused in certain women as so called date rape drugs.

Place navigation is essential for an animal's survival and this behavior has attracted the attention of scientists focused on the neural and neuropharmacological bases of learning and memory. Many navigational tests are employed today, such as the Morris water maze (MWM) which demands a precise representation of an unmarked place. Another spatial paradigm is the active place avoidance task. It requires mainly cognitive coordination in contrast to the MWM. Various rat strains are used in the research of animal models of cognitive impairments. The aim of this study was to compare sensitivity to the administration of higher doses (1.5mg/kg and 3mg/kg) of the central cholinergic blocker, scopolamine in outbred Long-Evans and Wistar rats. The results showed that while Wistar rats were more strongly affected by cholinergic blockade in the active place avoidance than Long-Evans rats, no differences were seen in the MWM. Long-Evans rats also showed better baseline performance in the active place avoidance and visible platform versions of the MWM (the latter suggesting better vision). This study demonstrated task-specific inter-strain differences in sensitivity to central cholinergic blockade in an active place avoidance task requiring cognitive coordination.

Antifungal activity of hyoscyamine (Hcy) and scopolamine (Sco) were determined by TLC-bioautography against fungi associated with H. muticus grown in Egypt, and those isolated from other plants grown in Japan. All 40 fungal strains were tolerant to Sco and sensitive to Hcy, exhibiting a growth inhibition zone around the Hcy spot on the bioautography plate. The strains were grouped into three types based on the appearance of the inhibition zone: (i) 17 strains exhibiting a clear inhibition zone, which remained clear at 8 d after incubation (type I); (ii) 22 strains exhibiting the inhibition zone with a brown circle surrounding the zone and regrowth within the inhibition zone (type II); (iii) 1 strain exhibiting the inhibition zone with no brown circle and regrowth within the inhibition zone (type III). In the type II and III strains, Hcy disappeared, and other alkaloids were found in the inhibition zones in its place. Hcy feeding experiments using Penicillium purpurogenum (type II) and Cunninghamella elegans (type III) revealed that these fungi may convert Hcy to a new alkaloid compound.
