Dihydromyricetin (DHM) is a natural flavonoid showing several health promoting effects such as protective activity during severe alcohol intoxication. The mechanism underlying the effects of DHM on alcohol metabolism is virtually unknown. The present paper is focused on clarifying the role of DHM in the liver alcohol elimination at its molecular level. First, impact of DHM on alcohol dehydrogenase (ADH) activity in vitro and the enzyme induction in vivo was examined. Neither the ADH activity nor the enzyme expression were influenced by DHM. Next, the effect of DHM during alcohol intoxication were studied on primary hepatocytes isolated from EtOH-premedicated and untreated rats. The viability of cells exposed to alcohol, estimated based on the released enzymes, alanine aminotransferase (ALT) and aspartate aminotransferase (AST), was slightly affected by DHM. Although the expected hepatoprotective effect of DHM was not fully achieved, DHM (in a concentration manner) proved to reduce the level of ROS/RNS in hepatocytes. However, no change in the rate of alcohol metabolism in vivo was found when rats were administered with a single or repeated dose of ethanol supplemented with DHM. In conclusion, the proposed positive effect of DHM during alcohol intoxication has not been proven. Moreover, there is no effect of DHM on the alcohol metabolism. The "hoped-for" DHM hepatoprotective activity can be attributed to the reduction of ROS/RNS levels in cells.

Methanol is the simplest alcohol. Compared to ethanol that is fully detoxified by metabolism. Methanol gets activated in toxic products by the enzymes, alcohol dehydrogenase and aldehyde dehydrogenase. Paradoxically, the same enzymes convert ethanol to harmless acetic acid. This review is focused on a discussion and overview of the literature devoted to methanol toxicology and antidotal therapy. Regarding the antidotal therapy, three main approaches are presented in the text: 1) ethanol as a competitive inhibitor in alcohol dehydrogenase; 2) use of drugs like fomepizole inhibiting alcohol dehydrogenase; 3) tetrahydrofolic acid and its analogues reacting with the formate as a final product of methanol metabolism. All the types of antidotal therapies are described and how they protect from toxic sequelae of methanol is explained.

Low oxygen conditions occur in grass sites due to high and frequent precipitation, poor soil quality, and over-irrigation followed by slow drainage. Three warm-season and one cool-season grass were analyzed at metabolic level during a time-course experiment performed in a controlled anoxic environment. Prolonged oxygen depletion proved detrimental by leading to premature death to all the species, with the exception of seashore paspalum. Moreover, the anoxia tolerance observed in these grasses has been associated with slow use of carbohydrates, rather than with their relative abundance, which was more important than their antioxidant capacity. Further physiological characterization of eight seashore paspalum genotypes to anoxia was also performed, by examining the variation in photosystem II (PSII) efficiency and gas exchange during post-anoxia recovery. Multivariate analysis highlighted the presence of three main clusters of seashore paspalum genotypes, characterized by different ability to restore the PSII photochemistry during recovery after one day of anoxia. Taken together, our data demonstrate that the analysis of post-anoxia recovery of fluorescence and gas exchange parameters can represent a fast and reliable indicator for selecting species and cultivars more able to acclimate their photosynthetic apparatus.

BACKGROUND: Alcohol variants such as ethanol and methanol are simple organic compounds widely used in foods, pharmaceuticals, chemical synthesis, etc. Both are becoming an emerging health problem; abuse of ethanol containing beverages can lead to disparate health problems and methanol is highly toxic and unfit for consumption. METHODS AND RESULTS: This review summarizes the basic knowledge about ethanol and methanol toxicity, the effect mechanism on the body, the current care of poisoned individuals and the implication of alcohols in the development of diseases. Alcohol related dementia, stroke, metabolic syndrome and hepatitis are discussed as well. Besides ethanol, methanol toxicity and its biodegradation pathways are addressed. CONCLUSIONS: The impact of ethanol and methanol on the body is shown as case reports, along with a discussion on the possible implication of alcohol in Alzheimer's disease and antidotal therapy for methanol poisoning. The role of ethanol in cancer and degenerative disorders seems to be underestimated given the current knowledge. Treatment in case of poisoning is another issue that remains unresolved even though effective protocols and drugs exist.

It is becoming increasingly evident that a high degree of regulation is involved in the protein synthesis machinery entailing more interacting regulatory factors. A multitude of proteins have been identified recently which show regulatory function upon binding to the ribosome. Here, we identify tight association of a metabolic protein aldehyde-alcohol dehydrogenase E (AdhE) with the E. coli 70S ribosome isolated from cell extract under low salt wash conditions. Cryo-EM reconstruction of the ribosome sample allows us to localize its position on the head of the small subunit, near the mRNA entrance. Our study demonstrates substantial RNA unwinding activity of AdhE which can account for the ability of ribosome to translate through downstream of at least certain mRNA helices. Thus far, in E. coli, no ribosome-associated factor has been identified that shows downstream mRNA helicase activity. Additionally, the cryo-EM map reveals interaction of another extracellular protein, outer membrane protein C (OmpC), with the ribosome at the peripheral solvent side of the 50S subunit. Our result also provides important insight into plausible functional role of OmpC upon ribosome binding. Visualization of the ribosome purified directly from the cell lysate unveils for the first time interactions of additional regulatory proteins with the ribosome.

Je známou a medicínsky overenou skutočnosťou, že po vypití rovnakej dávky alkoholu pri rovnakej telesnej hmotnosti sa u žien prejavujú výraznejšie známky alkoholového ovplyvnenia ako u mužov. Najdôležitejšou determinantou tohto javu je habituálny dimorfizmus muža a ženy. Ide najmä o nižšie pomerné zastúpenie telesnej vody v ženskom tele voči tukovému tkanivu, ktorá poskytuje vhodné biofyzikálne prostredie pre distribúciu vstrebaného a fyzikálne rozpusteného alkoholu v organizme. Významnú úlohu zohráva aj nižšia aktivita gastrickej frakcie alkoholdehydrogenázy (ADH) u žien v porovnaní s mužmi, zodpovedná za dosiahnutie vyšších krvných koncentrácii alkoholu (BAC) u žien po vypití ekvivalentného množstva alkoholu oproti mužom. Tento enzým s rozhodujúcou úlohou pre biotransformáciu molekuly alkoholu má nižšiu afinitu k alkoholu z nápojov s 10–40 % vol., a preto je aj následný „first-pass“ metabolizmus nižší a do krvného obehu sa u žien tak dostáva relatívne väčšie množstvo chemicky nezmeneného alkoholu, ktorý sa potom podieľa na samotnom alkoholovom ovplyvnení jedinca. Aktivita tohto enzýmu sa ešte viac znižuje so stúpajúcim vekom subjektu, u mužov dokonca výraznejšie. U jedincov vyššieho veku preto dochádza k zotretiu intersexuálnych rozdielov v BAC, ba až k preklopeniu sa do vyšších hodnôt BAC u mužov v porovnaní so ženami pri požití ekvivalentných množstiev alkoholu. Prekvapivou skutočnosťou je, že alkohol sa pri prechode pečeňou vo „first-pass“ metabolizme u žien metabolizuje rýchlejšie ako u mužov. Podľa niektorých autorov je za to zodpovedný relatívne väčší objem pečeňového tkaniva žien vo vzťahu k čistej tkanivovej hmote. Ak sa však porovnáva metabolizmus muža a ženy s rovnakým pomerom objemu pečeňového tkaniva k čistej tkanivovej hmote tela, ukazuje sa, že metabolická rýchlosť spracovania alkoholu sa u oboch pohlaví vyrovnáva. Bolo preukázané, že alkohol a pohlavné hormóny sa recipročne ovplyvňujú. U žien s rýchlejšou elimináciou alkoholu sa pri testoch preukázali vyššie hladiny pohlavných hormónov ako u mužov. Ženský estrogén potencuje aktivitu hepatálnej frakcie ADH a súčasne mužský dihydrotestosterón prispieva k nižšiemu alkoholovému ovplyvneniu mužov inhibíciou gastrickej ADH. Zdá sa teda, že špecifická konštitúcia ženského organizmu ako aj jedinečná kombinácia pohlavných hormónov sú zodpovedné za rozdielne účinky alkoholu na ľudských jedincov opačného pohlavia.
It is a well-known fact that after drinking the same amount of alcohol, women show more signs of the effects of alcohol than men of the same weight. It seems that the main factors responsible for sex differences in alcohol metabolism and influence are the relatively lower amount of body water related to body fat in women than men and lower gastric ADH activity in women, both of which enable women to reach higher BAC after drinking equivalent amounts of alcohol with men. On the other hand, first-pass metabolism of alcohol during passing through the liver is more rapid in women, probably due to bigger liver mass in women than in men. It is proven that alcohol and sex hormones have bilateral influence on each other. Women with more rapid alcohol elimination ability show higher levels of sex hormones in blood than the rest. It seems that the specific body constitution of the female organism as well as the unique combination of their sex hormones is responsible for the gender differences in alcohol influence.

Závislost na alkoholu je multifaktoriální chronické recidivující onemocnění, u kterého nenacházíme jasné, dominantní etiologické faktory, spíše velké množství faktorů genetických i psychosociálních, které zvyšují riziko závislosti. Molekulární genetika závislosti na alkoholu je tedy velmi komplexní a suma znalostí se stále dynamicky rozvíjí. Závislost na alkoholu je spojena se změnami genů pro glutamátergní, GABAergní, dopaminové, serotoninové, cholinergní, adrenergní, opioidní, kanabinoidní mechanismy, genů proteinů regulujících stresovou odpověď organismu, genů enzymů metabolizujících alkohol. Tyto změny se na klinické úrovni manifestují poruchami systému odměny, motivačních, hédonických a emočních systémů, inhibičních a excitačních procesů. Ukazuje se též, že specifická genetická náchylnost je v pozadí i odvykacího stavu a jeho závažných komplikací.
Alcohol dependence is a multifactorial chronic relapsing disorder with no clear dominant etiological factors; rather, there is a large number of both genetic and psychosocial issues that increase the risk of dependence. Molecular genetics of alcohol dependence is, therefore, a very complex and the body of evidence is still dynamically evolving. It is linked with changes of glutamatergic, GABA-ergic, dopamine, serotonine, cholinergic, adrenergic, opioid, cannabinoid genes, genes that regulate stress response, and alcohol metabolism. These changes are manifested as abnormalities of reward system, motivational, hedonic and emotional processes, and inhibition and excitation mechanisms. Specific genetic susceptibility exists for withdrawal syndrome and its complications.

Tolerance k alkoholu znamená nárůst snášenlivosti alkoholu při jeho opakovaném požívání. Toleranci k alkoholu dělíme na funkční, akutní, naučenou a metabolickou. V toleranci k alkoholu existují etnické i individuální rozdíly. Nižší toleranci k alkoholu zaviněnou nízkou hladinou alkoholdehydrogenázy nebo sníženou aktivitou aldehyddehydrogenázy mají původní obyvatelé Ameriky a Austrálie a část Japonců, Číňanů či Korejců. Děti z rodin alkoholiků a osoby s určitými povahovými vlastnostmi mají zvýšené riziko stát se alkoholiky. Alkohol v mozku zvyšuje účinek kyseliny γ-aminomáselné projevující se sedativně a stimuluje sekreci dopaminu a serotoninu. Souhra mezi genetickými vlohami a vlivem životního prostředí jsou rozhodující pro vývoj tolerance k alkoholu i pití a závislosti na něm.
