The effect of venlafaxine, a pharmaceutical commonly found in aquatic environment, was analyzed on non-target organism, Danio rerio (Hamilton, 1822). D. rerio embryos were treated by two different concentrations of venlafaxine: either concentration relevant in aquatic environment (0.3 μg/L) or concentration that was two orders of magnitude higher (30 μg/L) for the evaluation of dose-dependent effect. Time-dependent effect was rated at 24, 96, and 144 h post-fertilization (hpf). For gene expression, genes representing one of the phases of xenobiotic biotransformation (0 to III) were selected. The results of this study showed that the effect of venlafaxine on the zebrafish embryos is the most evident at hatching (96 hpf). At this time, the results showed a downregulation of gene expression in each phase of biotransformation and in both tested concentrations. In contrast, an upregulation of most of the genes was observed 144 hpf for both tested venlafaxine concentrations. The study shows that venlafaxine can affect the gene expression of biotransformation enzymes in D. rerio embryos even in the environmentally relevant concentration and thus disrupt the process of biotransformation. Moreover, the pxr regulation of genes seems to be disrupted after venlafaxine exposure in dose- and time-dependent manner.

Toxicity of the neonicotinoid pesticide thiacloprid to embryos and larvae of common carp Cyprinus carpio was assessed under laboratory conditions. Carp embryos and larvae were exposed to thiacloprid at four concentrations: 4.5 (environmental concentration), 45, 225, and 450 μg/L for 35 days and compared to a non-treated control group. The effects on lipid peroxidation, antioxidant enzyme levels (superoxide dismutase, glutathione reductase, catalase, glutathione S-transferase, and reduced glutathione), mortality, growth, development, and histopathogenesis were assessed. Thiacloprid at tested concentrations were not associated with negative effects on hatching, behaviour, embryo viability, or Fulton's condition factor. Carp exposed to 45 μg/L thiacloprid showed lower weight and length compared to controls. Thiacloprid at 225 and 450 μg/L delayed development and led to lower weight and length of carp. Significantly lower superoxide dismutase and glutathione reductase activity was observed at all tested concentrations of thiacloprid compared with the control group. Chronic thiacloprid exposure of early-life stages of carp affected ontogeny and growth rate, and inhibited antioxidant capacity.

The sensitivity of early life stages of common carp (Cyprinus carpio L.) to chronic exposure to single and combined environmental concentrations of the triazine metabolites terbuthylazine 2-hydroxy, terbuthylazine-desethyl and atrazine 2-hydroxy was evaluated under laboratory conditions. Their effects were assessed on lipid peroxidation, antioxidant enzymes (total superoxide dismutase, glutathione reductase, catalase, glutathione S-transferase, reduced glutathione), mortality, growth, development and histology. Single metabolites (terbuthylazine 2-hydroxy-0.73 μg/L; terbuthylazine-desethyl-1.80 μg/L; atrazine 2-hydroxy-0.66 μg/L) and combinations were not associated with negative effects on hatching, behaviour, embryo viability, growth or early ontogeny. Carp exposed to terbuthylazine-desethyl at 1.80 μg/L showed significantly lower total superoxide dismutase and glutathione reductase activity compared with the control group. Liver histology revealed diffused steatosis associated with the presence of lipid inclusions in hepatic cells in groups exposed to terbuthylazine-desethyl, atrazine 2-hydroxy and the tested combination of metabolites.
