In low concentration, fluoride is considered a necessary compound for human health. Exposure to high concentrations of fluoride is the reason for a serious disease called fluorosis. Fluorosis is categorized as Skeletal and Dental fluorosis. Several Asian countries, such as India, face contamination of water resources with fluoride. In this study, a comprehensive overview on fluoride contamination in Asian water resources has been presented. Since water contamination with fluoride in India is higher than other Asian countries, a separate section was dedicated to review published articles on fluoride contamination in this country. The status of health effects in Asian countries was another topic that was reviewed in this study. The effects of fluoride on human organs/systems such as urinary, renal, endocrine, gastrointestinal, cardiovascular, brain, and reproductive systems were another topic that was reviewed in this study. Different methods to remove fluoride from water such as reverse osmosis, electrocoagulation, nanofiltration, adsorption, ion-exchange and precipitation/coagulation were introduced in this study. Although several studies have been carried out on contamination of water resources with fluoride, the situation of water contamination with fluoride and newly developed technology to remove fluoride from water in Asian countries has not been reviewed. Therefore, this review is focused on these issues: 1) The status of fluoride contamination in Asian countries, 2) health effects of fluoride contamination in drinking water in Asia, and 3) the existing current technologies for defluoridation in Asia.

Medzi človekom a viacerými mikroorganizmami sa počas fylogenetického vývoja rozvinuli rôzne typy synbiózy. Najvyšším typom je mutuálny typ synbiózy, ktorý bol základom pre vývin špecializovaných systémov človeka, ako sú napr. GALT a MALT. Táto užitočná mikroflóra je tvorená najmä probiotikami, ktoré vytvárajú špecifické osadenstvá a z nich je najvýznamnejší mikrobióm tráviaceho traktu. Ľudský plod sa pôvodne považoval za sterilný, avšak výsledky prác poslednej dekády naznačujú, že už gravidita je obdobie, kedy dochádza k prvému kontaktu plodu s baktériami. Vznik mikroflóry novorodenca sa považuje za kritický krok určujúci prirodzený metabolický a imunologický vývin tráviaceho traktu a z neho vyplývajúci krátkodobý i dlhodobý vplyv na zdravotný stav jedinca. Interakcia mikrobiómu s makroorganizmom je veľmi intenzívna a významná pre človeka a to nielen v imunologickej, ale napríklad aj v metabolickej či psychickej oblasti. V práci sú uvedené definície probiotík, prebiotík a postbiotík, ktoré sú novo definovanou skupinou účinných látok, ktoré vznikajú metabolickou činnosťou probiotík vplyvom na prebiotiká. Tieto postbiotiká hrajú významnú úlohu v ovplyvňovaní ostatnej mikroflóry, majú antimikrobiálne, imunomodulačné i antioxidačné vlastnosti. Uvádzame význam materského mlieka a oligosacharidov pri kreovaní črevného mikrobiómu a jeho možné ovplyvnenie stravou a inými faktormi, čo môže mať význam pre zdravotný stav nielen samotného dieťaťa, ale aj v jeho ďalšom živote. Podávame prehľad niektorých účinkov prebiotík a postbiotík aj na podklade EBM i experimentálnych prác, ktorých výsledky je možné využívať v klinickej praxi. Modulácia ekosystému tráviaceho traktu s využitím probiotických mikroorganizmov, prebiotík a bioaktívnych látok by mohla zefektívniť prevenciu a terapiu mnohých chorobných procesov.
They have developed different types of synbiosis between different microorganisms during phyloge- netic development. Thehighest type of synbiosis is the mutual type, which was the basis for the development of specialized human systems, GALT andMALT. This useful microflora consists mainly of probiotics that create spe- cific habits and which are the most important in digestivetract. The human fetus was originally thought to be sterile, but the scientific results, in the last decade, indicate that pregnancyis a period when the first phase of contact with bacteria occurs. The emergence of a newborn microflora is considered to be acritical stage determining the natu- ral metabolic and immunological development of the gastrointestinal tract and resultingshort- and long-term effects on the individual‘s state of health. Interaction of the microbe with the macro organism is very intenseand important for humans and not only in the immunological, but also in the metabolic or psychological area.This paper includes the definitions of probiotics, prebiotics and postbiotics, which are newly defined by a group of active substances,which are created by the metabolic activity of probiotics on prebiotics. These postbiot- ic substances play an important role ininfluencing other microflora, have antimicrobial, immunomodulatory and an- tioxidant properties. We describe the importance ofbreast milk and oligosaccharides in the formation of intestinal micro biome and its potential influence on diet and other factors,which may be important for the health of the child, but also in his/her next life. We reviewed the effects of oligosacharides andpostbiotics on the basis of EBM and experimental works, the results of which can be used in clinical practice.Modulation of the digestive tract ecosystem with the use of probiotic micro-organisms, prebiotics and bioactive substances couldeffectively prevent the prevention and therapy of many disease processes.

BACKGROUND: Hematophagous mosquitos and ticks avoid host hemostatic system through expression of enzyme inhibitors targeting proteolytic reactions of the coagulation and complement cascades. While most inhibitors characterized to date were found in the salivary glands, relatively few others have been identified in the midgut. Among those, Boophilin is a 2-Kunitz multifunctional inhibitor targeting thrombin, elastase, and kallikrein. However, the kinetics of Boophilin interaction with these enzymes, how it modulates platelet function, and whether it inhibits thrombosis in vivo have not been determined. METHODOLOGY/PRINCIPAL FINDINGS: Boophilin was expressed in HEK293 cells and purified to homogeneity. Using amidolytic assays and surface plasmon resonance experiments, we have demonstrated that Boophilin behaves as a classical, non-competitive inhibitor of thrombin with respect to small chromogenic substrates by a mechanism dependent on both exosite-1 and catalytic site. Inhibition is accompanied by blockade of platelet aggregation, fibrin formation, and clot-bound thrombin in vitro. Notably, we also identified Boophilin as a non-competitive inhibitor of FXIa, preventing FIX activation. In addition, Boophilin inhibits kallikrein activity and the reciprocal activation, indicating that it targets the contact pathway. Furthermore, Boophilin abrogates cathepsin G- and plasmin-induced platelet aggregation and partially affects elastase-mediated cleavage of Tissue Factor Pathway Inhibitor (TFPI). Finally, Boophilin inhibits carotid artery occlusion in vivo triggered by FeCl3, and promotes bleeding according to the mice tail transection method. CONCLUSION/SIGNIFICANCE: Through inhibition of several enzymes involved in proteolytic cascades and cell activation, Boophilin plays a major role in keeping the midgut microenvironment at low hemostatic and inflammatory tonus. This response allows ticks to successfully digest a blood meal which is critical for metabolism and egg development. Boophilin is the first tick midgut FXIa anticoagulant also found to inhibit thrombosis.

The saprophagous larvae of bibionid flies harbor bacteria in their alkaline intestinal tracts, but little is known about the contribution of the gut microbiota to the digestion of their recalcitrant diet. In this study, we measured oxygen and hydrogen partial pressure, redox potential and pH in the midgut, gastric caeca and hindgut of larvae of the bibionid fly Penthetria holosericea with Clark-type O2 and H2 microsensors, platinum redox microelectrodes, and LIX-type pH microelectrodes. The center of the midgut lumen was anoxic, whereas gastric caeca and hindgut were hypoxic. However, redox potential profiles indicated oxidizing conditions throughout the gut, with lowest values in the midgut (+20 to +60mV). Hydrogen production was not detected. The midgut was extremely alkaline (pH around 11), whereas hindgut and gastric caeca were neutral to slightly alkaline. While HPLC analysis showed high concentrations of glucose in the midgut (15mM) and gastric caeca (27mM), the concentrations of microbial fermentation products such as lactate (2-4mM), acetate (<1mM) and succinate (<0.5mM) were low in all gut regions, suggesting that the contribution of microorganisms to the digestive process, particularly in the alkaline midgut, is only of minor importance. We conclude that the digestive strategy of the saprophytic larva of P. holosericea, which feeds selectively on decomposed leaves and its own microbe-rich faeces, differs fundamentally from those of detritivorous and humivorous insects, which host a highly active, fermentative microbiota in their alkaline midgut or hindgut compartments.

V posledních letech se lidé stále více zajímají o potraviny, které nabízejí vyváženou stravu a podporují lidské zdraví. To je důvod, proč poptávka po funkčních potravinách neustále roste. Mnoho studií se zaměřuje na stadium příznivého vlivu probiotik, které jsou jednou z hlavních součástí funkčních potravin, jakož i na zlepšení schopnosti probiotik přežít během průchodu trávicím traktem poskytování v životaschopné podobě až do tlustého střeva. Proto koncepce efektivního, nízkonákladového systému pro transport probiotických bakterií přitahuje stále větší pozornost odborníků. V tomto článku je testována mikroenkapsulace do alginátu sodného s cílem zlepšit životaschopnost Lb plantarum G1 v in vitro simulovaném žaludečním a střevním systému. Inkubace probíhala v přítomnosti tří nápojů, které jsou hojně konzumované v alžírské populaci a na celém světě – černá káva, zelený čaj a pomerančový džus. Cílem bylo přispět doporu- čením pro řádnou konzumaci probiotik. Výsledky ukázaly, že požití probiotika současně s jedním ze studovaných nápojů vedlo ke snížení životaschopnosti probiotických bakterií. Mikroenkapsulace do alginátu ovšem zlepšila jejich životaschopnost, dokonce i na konci gastrointestinálního systému. Viabilita bakterií v černé kávě, zeleném čaji a pomerančovém džusu byla 9.47, 9.07 a 10.39 log CFU/mL, resp.
In recent years, people are increasingly interested in foods which offer a balanced diet and promote human health. This is why the demand on functional foods is growing steadily. Many recent studies are focusing on the beneficial effect of probiotics, which are one of the main components of functional foods as well as improving their capacity to survive during passage through digestive tract to arrive viable and in sufficient amounts to colon. Therefore, the conception of an efficient, low-cost system for the delivery of probiotic bacteria is highly attracting food science researchers. In the current paper, microencapsulation within sodium alginate is tested for its capacity to improve Lb. plantarum G1 viability in an in vitro simulated gastrointestinal system designed in the laboratory. Incubation was realized in presence of three beverages that are commonly and highly consumed by Algerian population and worldwide they include black coffee, green tea and orange juice, the objective was to contribute in developing recommendations to properly consume probiotics, The results indicated that introducing probiotics simultaneously with one of the studied beverages resulted in a decrease in probiotic viability, however, microencapsulation within alginate beads improved its viability even at the end of the gastrointestinal simulated transit, suggesting that probiotics, when encapsulated, can reach the lower intestine. The cell viable counts for black coffee, green tea and orange juice were 9.47, 9.07 and 10.39 log CFU/mL, respectively.
