Live-imaging analysis is performed in many laboratories all over the world. Various tools have been developed to enable protein labeling either in plasmid or genomic context in live yeast cells. Here, we introduce a set of nine integrative modules for the C-terminal gene tagging that combines three fluorescent proteins (FPs)-ymTagBFP, mCherry and yTagRFP-T with three dominant selection markers: geneticin, nourseothricin and hygromycin. In addition, the construction of two episomal modules for Saccharomyces cerevisiae with photostable yTagRFP-T is also referred to. Our cassettes with orange, red and blue FPs can be combined with other fluorescent probes like green fluorescent protein to prepare double- or triple-labeled strains for multicolor live-cell imaging. Primers for PCR amplification of the cassettes were designed in such a way as to be fully compatible with the existing PCR toolbox representing over 50 various integrative modules and also with deletion cassettes either for single or repeated usage to enable a cost-effective and an easy exchange of tags. New modules can also be used for biochemical analysis since antibodies are available for all three fluorescent probes.

High transformation competency of Escherichia coli is one of the critical factors in the bacterial artificial chromosome (BAC)-based DNA library construction. Many electroporation protocols have been published until now, but the majority of them was optimized for transformation of small plasmids. Large plasmids with a size above 50 kbp display reduced transformation efficiency and thereby require specific conditions in the preparation and electroporation of electrocompetent cells. In the present work, we have optimized the parameters critical to the application of BAC DNA electrotransformation into E. coli. Systematic evaluation of electroporation variables has revealed several key factors like temperature of growth, media supplements, washing buffer, and cell concentration. Improvements made in the transformation protocol have led to electrocompetent cells with transformation efficiency up to 7 × 10(8) transformants per microgram of 120 kbp BAC plasmid DNA. We have successfully used in-house prepared competent cells, the quality of which is comparable with those produced by different companies, in the construction of metagenomic libraries from the soil. Our protocol can also be beneficial for other application with limited DNA source.

Streptococcus pyogenes, celosvětově rozšířený patogen, způsobuje celou řadu neinvazivních a invazivních onemocnění od akutní faryngitidy a impetiga po nekrotizující fascitidu a streptokokový syndrom toxického šoku. Cílem práce bylo stanovení toxigenních profilů metodou polymerázové řetězové reakce v reálném čase (RT-PCR) a emm typizace S. pyogenes pro epidemiologické účely a statistické vyhodnocení vztahu emm typu a genů toxigenního profilu k invazivitě onemocnění. V Národní referenční laboratoři pro streptokokové nákazy Státního zdravotního ústavu v Praze bylo v letech 2011–2012 vyšetřeno celkem 111 izolátů S. pyogenes pětadvaceti různých emm typů a 37 toxigenních profilů (streptokokové pyrogenní exotoxiny A-C, F-M, Z, streptokokový superantigen ssa a streptokokový mitogenní exotoxin Z smeZ). 55 izolátů bylo invazivních a 56 neinvazivních. V uvedeném období prevaloval typ emm1 (31,5 %) následovaný typem emm28 (11,7 %) a typem emm12 (9,9 %). U všech izolátů byl potvrzen streptokokový pyrogenní exotoxin B a F (speB a speF). SpeG a smeZ měla většina kmenů (97,3 % a 92,8 %), ssa byl zjištěn u 8,1 % kmenů a speL u 3,6 % kmenů. Nebyla potvrzena statisticky významná souvislost mezi emm typem a invazivitou izolátů ani mezi toxigenním profilem a invazivitou izolátů.
Streptococcus pyogenes, a globally spread pathogen, is the cause of a wide range of invasive and non-invasive diseases, from acute pharyngitis and impetigo to necrotizing fasciitis and streptococcal toxic shock syndrome. The study objectives were toxigenic profiling of S. pyogenes isolates by real-time polymerase chain reaction (RT-PCR) and emm typing for epidemiological purposes and analysis of the relationships between emm types, toxigenesis, and disease invasiveness. In 2011-2012, the National Reference Laboratory for Streptococcal Infections, National Institute of Public Health, analyzed 111 isolates of S. pyogenes assigned to 25 mm types and 37 toxigenic profiles (streptococcal pyrogenic exotoxins A-C, F-M, and Z, streptococcal superantigen ssa, and streptococcal mitogenic exotoxin Z smeZ). Fiftyfive isolates were invasive and 56 were non-invasive. The most prevalent emm type was emm1 (31.5%), followed by emm28 (11.7%) and emm12 (9.9%). All isolates carried genes for streptococcal pyrogenic exotoxin B and F (speB and speF), most isolates (97.3% and 92.8%) harboured speG and smeZ, respectively, 8.1% of isolates were ssa positive, and 3.6% encoded speL. No statistically significant association was found between emm type or toxigenic profile and invasiveness of isolates.

We aimed to establish an efficient RNA interference (RNAi) system in the industrially important filamentous fungus Trichoderma koningii using the DsRed protein as a reporter of the silencing process. To accomplish this, a DsRed expression cassette was transformed into T. koningii, and a recombinant strain that stably expressed DsRed was obtained. Next, a vector-directing expression of a DsRed hairpin RNA was constructed and transformed into the T. koningii recipient strain. Approximately 79 % of transformants displayed a decrease in DsRed fluorescence, and expression of DsRed in some transformants appeared to be fully suppressed. Characterization of randomly selected transformants by genomic DNA PCR analysis, real-time PCR quantification, and western blot confirmed downregulation of gene expression at different levels. The RNA silencing approach described here for T. koningii is effective, and the DsRed reporter gene provides a convenient tool for identification of silenced fungal transformants by their DsRed fluorescence compared to the control strain. The results of this study demonstrate the power of RNAi in T. koningii, which supports the use of this technology for strain development programs and functional genomics studies in industrial fungal strains.
