UV záření významně přispívá k urychlení procesu stárnutí kůže. Po podrobném studování mikrobiomu lidského těla se pozornost ubírá ke zkoumání jeho konkrétních zástupců a jejich podílu na procesu stárnutí kůže a fotoprotekci. Jedním z nejnovějších trendů v boji proti stárnutí pleti je probiotická péče. Přestože výzkum v této oblasti je teprve v počátečních fázích, na trh se rychle dostávají nové produkty. Cílem tohoto článku je zaměřit pozornost na aktuální poznatky o vlivu mikrobiomu a možnosti uplatnění probiotik v prevenci stárnutí kůže a ochraně před UV zářením.
UV radiation significantly contributes to the acceleration of the skin aging process. After a detailed study of the microbiome of the human body, attention is now turning to the investigation of its specific representatives and their contribution to the skin ageing process and photoprotection. One of the latest trends in the fight against skin aging is probiotic skin care. Although research in this area is only in its early stages, new products are rapidly entering the market. The aim of this article is to focus attention on current knowledge about the influence of the microbiome and the possibility of using probiotics in the prevention of skin aging and protection against UV radiation.

The anoxygenic phototrophic bacteria (APB) are an active component of aquatic microbial communities. While DNA-based studies have delivered a detailed picture of APB diversity, they cannot provide any information on the activity of individual species. Therefore, we focused on the expression of a photosynthetic gene by APB communities in two freshwater lakes (Cep lake and the Římov Reservoir) in the Czech Republic. First, we analyzed expression levels of pufM during the diel cycle using RT-qPCR. The transcription underwent a strong diel cycle and was inhibited during the day in both lakes. Then, we compared DNA- (total) and RNA-based (active) community composition by sequencing pufM amplicon libraries. We observed large differences in expression activity among different APB phylogroups. While the total APB community in the Římov Reservoir was dominated by Betaproteobacteria, Alphaproteobacteria prevailed in the active library. A different situation was encountered in the oligotrophic lake Cep where Betaproteobacteria (order Burkholderiales) dominated both the DNA and RNA libraries. Interestingly, in Cep lake we found smaller amounts of highly active uncultured phototrophic Chloroflexi, as well as phototrophic Gemmatimonadetes. Despite the large diversity of APB communities, light repression of pufM expression seems to be a common feature of all aerobic APB present in the studied lakes.

Shark Bay, Western Australia is a World Heritage area with extensive microbial mats and stromatolites. Microbial communities that comprise these mats have developed a range of mitigation strategies against changing levels of photosynthetically active and ultraviolet radiation, including the ability to biosynthesise the UV-absorbing natural products scytonemin and mycosporine-like amino acids (MAAs). To this end, the distribution of photoprotective pigments within Shark Bay microbial mats was delineated in the present study. This involved amplicon sequencing of bacterial 16S rDNA from communities at the surface and subsurface in three distinct mat types (smooth, pustular and tufted), and correlating this data with the chemical and molecular distribution of scytonemin and MAAs. Employing UV spectroscopy and MS/MS fragmentation, mycosporine-glycine, asterina and an unknown MAA were identified based on typical fragmentation patterns. Marker genes for scytonemin and MAA production (scyC and mysC) were amplified from microbial mat DNA and placed into phylogenetic context against a broad screen throughout 363 cyanobacterial genomes. Results indicate that occurrence of UV screening compounds is associated with the upper layer of Shark Bay microbial mats, and the occurrence of scytonemin is closely dependent on the abundance of cyanobacteria.
