Nestin is a class VI intermediate filament protein expressed in the cytoplasm of stem and progenitor cells in the mammalian CNS during development. In adults, nestin is present only in a small subset of cells and tissues, including the subventricular zone of the adult mammalian brain, where neurogenesis occurs. Nestin expression has also been detected under such pathological conditions as ischemia, inflammation, and brain injury, as well as in various types of human solid tumors and their corresponding cell lines. Furthermore, nestin was recently found in the nuclei of glioblastoma, neuroblastoma, and angiosarcoma cells and it was proved to interact directly with the nuclear DNA in neuroblastoma cells. Here, we perform the first study of the intracellular distribution of nestin in cell lines derived from neurogenic tumors. Using immunodetection methods, we examined nestin expression in tumor-derived cell lines obtained from 11 patients with neuroblastoma, medulloblastoma, or glioblastoma multiforme. Besides its standard cytoplasmic localization, nestin was present in the nuclei of two neuroblastoma cell lines and one medulloblastoma cell line. Nestin was only present in the nuclei of cells with diffuse cytoplasmic staining for this protein, and the proportion of cells positive for nestin in nuclei, as well as the intensity of staining, varied. The presence of nestin in the nuclei was confirmed by both transmission electron microscopy and Western blotting. Our results indicate that the presence of nestin in the nuclei of tumor cells is not very rare, especially under in vitro conditions.

Studie analyzuje výsledky ultrastrukturálního vyšetření 89 nemocných s neuroblastickými nádory. Nálezy u všech nádorů, včetně 21 případů nediferencovaných neuroblastomů (NB), byly charakterizovány výběžky nervových buněk, které obsahují neurofilamenta, mitochondrie a neurosekreční granula. Granula byla obvykle chudě zastoupena a jejich množství a distribuce byly nezávislé na jiných rysech diferenciace nádorových buněk. Výrazným znakem byly degenerativní změny v tělech nádorových buněk a v jejich výběžcích. Přestože se vyskytovaly jak u nemocných, kteří byli operováni primárně, tak u nemocných, kteří byli předoperačně léčeni chemoterapii, nacházeli jsme tyto změny extenzivněji vyjádřeny u předoperačně léčených (28 z 80 nemocných). Degenerativní změny zahrnovaly zduření a dilatace neuronálních výběžků, akumulaci heterofagosomů, myelinových tělísek a cytoskeletálních filament. Některé gangliové buňky měly dilatované endoplazmatické retikulum a obsahovaly lipofuscin. U jednoho ganglioneuroblastomu jsme pozorovali denzní granulovaná tělíska podobtgící se nenromelaninu. Diagnózu nediferencovaného NB jsme mohli stanovit na základě ultrastrukturálních vyšetření u 13 z 21 nediferencovaných NB; u zb3h^ajících osmi případů elektronová mikroskopie potvrdila diagnózu spolu s imunohistochemickými reakcemi. Elektronová mikroskopie je stále cenným nástrojem V diagnostice neuroblastických nádorů, vedle novějších diagnostických přístupů, jakými jsou imunohistochemická a molekulární vyšetření.
The study presents ultrastructural findmgs in 89 neuroblastic tumors. All of them including 21 undifferentiated neuroblastomas (NB) were characterized by neural cell processes which contained neurofilaments, mitochondria and neurosecretory granules. The granules were generally sparse and their number and distribution was a feature independent on other signs of differentiation of the tumor cells. We found degenerative changes in the tumor cell bodies and their neuronal processes to be a prominent phenomenon in neuroblastic tumors. Although they occurred both in patients who were operated primarily and in those who received a preoperative chemotherapy they were most conspicuous in the latter group (28 of 80 patients). The degenerative changes included swelling and dilatations of the neuronal processes, accumulation of heterophagosomes, myelinated bodies and cytoskeletal filaments. Some ganglion cells had dilated endoplasmic reticulum, and contained lipofuscin. In one ganglioneuroblastoma we detected dense granulated bodies resembling neuromelanin. The diagnosis of undifferentiated NB was estabhshed on the basis of ultrastructural investigations in 13 out of 21 undifferentiated NB; in the results of immunohistochemical reactions. The electron microscope is still a valuable instrument for diagnosis of neuroblastic tumors in addition to newer diagnostic approaches such as immunohistochemical and molecular investigations.
