Di-(2-ethylhexyl)-phthalate (DEHP) is a compound widely used as a plasticizer, which can leach from plastics into the environment and thus influence human health. The aim of this study was to analyze whether exposure to an environmentally relevant dose of DEHP during mice fetal development or puberty can cause long-lasting changes detectable month/s after the last exposure. We used a DEHP concentration relevant to a daily human intake of 2.4-3 μg/kg of body weight/day. CD1 outbred mice were treated either in utero or postnatally during puberty and analyzed in adulthood. Analyzing fertility parameters using morphometric, histologic, genomic and proteomic methods we showed that DEHP exposure leads to decreased sperm concentration and quality, in both experimental groups. Moreover, the changes in anogenital distance, seminal vesicle weight, and testicular gene expression suggest a disturbance of androgen signaling in exposed animals. In conclusion, we hereby present, that the prenatal and pubertal exposure to a low dose of DEHP negatively influenced reproductive endpoints in male mice, and some of the effects were persistent until adulthood.

Spermiation and changes in androgen (testosterone, T and 11-ketotestosterone, 11-KT) levels were studied in sterlet (Acipenser ruthenus) treated with GnRH agonist implants (DAla(6)-Pro(9)-LHRHa) at 25 and 75 μg kg(-1) b.w. and compared with those males treated with 4 mg kg(-1) b.w. of carp pituitary extract (CPE) and 3 pellets of Ovopel kg(-1) b.w., which contains DAla(6)-Pro(9)NEt-mGnRH and metoclopramide. Sperm quality (sperm mass, spermatozoa concentration and sperm motility and velocity) was evaluated 24, 48 and 72 h after hormonal treatments. Males did not release sperm in the control group injected with physiological solution, while sperm could not be collected 7 days after treatments in all hormonally treated groups. Spermiation rates were 100 % in the CPE and Ovopel groups and 25-50 % in the GnRHa-treated groups. Sperm production was significantly lower in the GnRHa-treated groups than in the CPE and Ovopel groups and decreased 72 h after hormonal treatment. Sperm motility and velocity were higher in the Ovopel and GnRHa (75 μg) groups compared to the CPE and GnRHa (25 μg) groups and decreased 72 h after hormonal treatment. Androgens were only affected in spermiating males and changed in the Ovopel and GnRHa (75 μg) after hormonal treatment. Significant correlations were observed between sperm production, sperm motility and sperm velocity, but not androgens. The present study suggests involvement of dopamine in sturgeon spawning. Additionally, better sperm quality observed in the Ovopel group and particularly sperm motility in the GnRHa (75 μg) suggests enhancement of sperm quality in sturgeon treated with GnRHa. Therefore, further study is needed to induce fully spermiation using GnRHa implants in combination with a dopamine inhibitor.

Objective. To compare the effects on ovarian function of low chlorinated polychlorinated biphenyls (PCBs) mixture generally distributed at a faster rate and metabolised to a greater extent (Delor 103) with high chlorinated PCBs mixture which are accumulated in large amounts in biological samples (Delor 106). Methods. Theca interna and granulosa cells were isolated from prepubertal porcine follicles and initially cultured in M199/CS without test compounds for 24 hrs to allow for cell attachment to the wells. After 24 h, the medium was discarded and 0.3 ml of fresh M199 alone was added to the control culture, while Delor 103 or Delor 106 at concentrations of 0.02, 0.2 or 2 ng/ml medium were added to the cultures. For analysis of steroid levels, the media were collected after 24 hrs (single exposure) or 72 hrs (repeated exposure) and frozen at – 20 0C. Additionally, after 72 hrs, LDL test was used to show the action of both mixtures on the cell viability and cells were used for the measurement of caspase-3 activity. Results. An increase in testosterone secretion after the single exposure and strong stimulatory action on estradiol secretion was noted after permanent treatment with Delor 103. In Delor 106 treated cultures, antiestrogenic action after the single exposure and strong estrogenic action in permanently treated cells was observed. Both mixtures had no effect on cell viability and apoptosis. Conclusions. By stimulation of estradiol secretion from ovarian follicles in prepubertal animals, PCB mixture could be responsible for premature development of follicles and in consequence, premature sexual maturation.
