The mechanisms leading to autoimmune and inflammatory diseases in the CNS have not been elucidated. The environmental triggers of the aberrant presence of CD4+ T cells in the CNS are not known. In this article, we report that abnormal β-catenin expression in T cells drives a fatal neuroinflammatory disease in mice that is characterized by CNS infiltration of T cells, glial activation, and progressive loss of motor function. We show that enhanced β-catenin expression in T cells leads to aberrant and Th1-biased T cell activation, enhanced expression of integrin α4β1, and infiltration of activated T cells into the spinal cord, without affecting regulatory T cell function. Importantly, expression of β-catenin in mature naive T cells was sufficient to drive integrin α4β1 expression and CNS migration, whereas pharmacologic inhibition of integrin α4β1 reduced the abnormal T cell presence in the CNS of β-catenin-expressing mice. Together, these results implicate deregulation of the Wnt/β-catenin pathway in CNS inflammation and suggest novel therapeutic strategies for neuroinflammatory disorders.

BACKGROUND: The WNT/planar-cell-polarity (PCP) pathway is a key regulator of cell polarity and directional cell movements. Core PCP proteins such as Van Gogh-like2 (VANGL2) are evolutionarily highly conserved; however, the mammalian PCP machinery is still poorly understood mainly due to lack of suitable models and quantitative methodology. WNT/PCP has been implicated in many human diseases with the most distinguished positive role in the metastatic process, which accounts for more than 90% of cancer related deaths, and presents therefore an attractive target for pharmacological interventions. However, cellular assays for the assessment of PCP signaling, which would allow a more detailed mechanistic analysis of PCP function and possibly also high throughput screening for chemical compounds targeting mammalian PCP signaling, are still missing. RESULTS: Here we describe a mammalian cell culture model, which correlates B lymphocyte migration of patient-derived MEC1 cells and asymmetric localization of fluorescently-tagged VANGL2. We show by live cell imaging that PCP proteins are polarized in MEC1 cells and that VANGL2 polarization is controlled by the same mechanism as in tissues i.e. it is dependent on casein kinase 1 activity. In addition, destruction of the actin cytoskeleton leads to migratory arrest and cell rounding while VANGL2-EGFP remains polarized suggesting that active PCP signaling visualized by polarized distribution of VANGL2 is a cause for and not a consequence of the asymmetric shape of a migrating cell. CONCLUSIONS: The presented imaging-based methodology allows overcoming limitations of earlier approaches to study the mammalian WNT/PCP pathway, which required in vivo models and analysis of complex tissues. Our system investigating PCP-like signaling on a single-cell level thus opens new possibilities for screening of compounds, which control asymmetric distribution of proteins in the PCP pathway.

Mnohočetný myelom je hematoonkologické onemocnění charakterizované maligní proliferací plazmatických buněk. Tyto buňky se hromadí v kostní dřeni, kde potlačují fyziologickou krvetvorbu a zároveň interagují s celou škálou cytokinů, růstových faktorů a adhezivních molekul. Je zřejmé, že právě mikroprostředí kostní dřeně hraje velkou roli v patogenezi onemocnění, ale i v rezistenci k léčbě.
Multiple myeloma is a hematooncological disease characterized by malignant proliferation of plasma cells. These cells accumulate in the bone marrow where they suppress physiological hematopoiesis; at the same time, these cells interact with a wide variety of cytokines, growth factors and adhesion molecules. It is obvious that the bone marrow microenvironment plays an important role in disease pathogenesis as well as treatment resistance.

Agresívna fibromatóza, známa tiež pod pojmom desmoid, desmoidný tumor, je špecifické a pomerne zriedkavo sa vyskytujúce ochorenie. Zaraďuje sa medzi heterogénnu skupinu mezenchýmových nádorov. V pravom slova zmysle sa jedná o monoklonálnu proliferáciu fibroblastov vychádzajúcu z fibro-aponeurotického tkaniva s typicky lokálne invazívnym rastom bez metastatického potenciálu. Poznanie procesov na molekulárnej úrovni vďaka podrobnému štúdiu génu APC (adenomatous polyposis coli) pri FAP zohráva významnú úlohu v odhaľovaní biologickej povahy desmoidov. Vo všeobecnosti sa dá skonštatovať, že β-katenínová dysregulácia je základ pri vzniku desmoidov asociovaných s FAP. Wingless/Wnt dráha zohráva dôležitú úlohu v patogenéze hlbokých fibromatóz, pričom zatiaľ nie je jednoznačne preukázané, že spúšťacím mechanizmom sú mutácie génu APC či β-katenínu. Z doterajších poznatkov môžeme predpokladať, že objavenie nových geneticko-biologických korelácií prispeje k využitiu ďalších liečebných možností.
Aggressive fibromatosis, also known as desmoid tumor, is specific and relatively rarely occuring disease. It belongs to heterogenous group of soft tissue tumors. Originally, it arises from fibroblasts with monoclonal proliferation derived from fibro-aponeurotic tissue with typical local invasive spreading without metastatic tendency. Increased amount of knowledge about the role of the APC gene and its protein product in FAP play an important role in revealing the molecular nature of desmoid tumors. In general, we can conclude that the β-catenin dysregulation is the key player of the FAP associated desmoid tumor onset. The Wingless/Wnt cascade plays a crucial role in the pathogenesis of aggressive fibromatosis. However, it has not been definitely proven that the mutations of APC or β-catenin genes are the trigger mechanisms. The research outcome can pave the way for using target biological therapy in routine practice in patients with aggressive fibromatosis in the future.
