Natural sweeteners are in high demand as a part of a healthy lifestyle. Among them, sweeteners with decreased caloric value and suitability for diabetes patients are most requested. Extension in their consumption extends the need for their quality control. A fast gradient UHPLC coupled with charged aerosol detection enabling quantitation of stevioside, rebaudioside A-D, and steviolbioside in commercial sweeteners and Stevia rebaudiana plant extracts has been developed. The method was developed to achieve high efficiency, simplicity, versatility, and low solvent consumption. All steviol glycosides were baseline-separated in less than 4 min with a total run time of 7 min. Buffer-free eluents were used in the separations and only 2.45 mL solvent were needed per analysis. The Luna Omega Polar column featuring polar modification of the C18 stationary phase was employed with mobile phases composed of water and acetonitrile for the excellent separation of polar steviol glycosides. The flow rate of the mobile phase 0.35 mL/min, column temperature 50 °C and injection volume 2 µL were used. Critical pair of glycosides, stevioside and rebaudioside A, were baseline separated with a resolution of 2.41. The universal charged aerosol detector allowed quantitation of steviol glycosides with a limit of detection and quantitation 0.15 and 0.5 µg/mL, respectively. Method intra-day precision was less than 2% (RSD), and the recovery was 89.6-105.0% and 93.8-111.4% for plant material and sweetener tablets, respectively. The quantity of steviol glycosides in three out of four commercial sweeteners was 3.0-12.3% higher than declared. The content was about 12.4% less than declared in one sample. But the difference from the labeled content corresponded to trueness and precision of the developed method together with variability of sweeteners production. The most abundant glycoside detected in sweeteners was stevioside followed by rebaudioside A. A leaf-to-stem ratio describing the dominant accumulation of steviol glycosides in leaves affected the differences in the amount of steviol glycosides among plant samples.
Pollution by chemical substances is of concern for the maintenance of healthy and sustainable aquatic environments. While the occurrence and fate of numerous emerging contaminants, especially pharmaceuticals, is well documented in freshwater, their occurrence and behavior in coastal and marine waters is much less studied and understood. This study investigates the occurrence of 58 chemicals in the open surface water of the Western Mediterranean Sea for the first time. 70 samples in total were collected in 10 different sampling areas. 3 pesticides, 11 pharmaceuticals and personal care products and 2 artificial sweeteners were detected at sub-ng to ng/L levels. Among them, the herbicide terbuthylazine, the pharmaceuticals caffeine, carbamazepine, naproxen and paracetamol, the antibiotic sulfamethoxazole, the antibacterial triclocarban and the two artificial sweeteners acesulfame and saccharin were detected in all samples. The compound detected at the highest concentration was saccharin (up to 5.23 ng/L). Generally small spatial differences among individual sampling areas point to a diffuse character of sources which are likely dominated by WWTP effluents and runoffs from agricultural areas or even, at least for pharmaceuticals and artificial food additives, from offshore sources such as ferries and cruising ships. The implications of the ubiquitous presence in the open sea of chemicals that are bio-active or toxic at low doses on photosynthetic organisms and/or bacteria (i.e., terbuthylazine, sulfamethoxazole or triclocarban) deserve scientific attention, especially concerning possible subtle impacts from chronic exposure of pelagic microorganisms.

Energetické nápoje jsou nealkoholické nápoje, které stimulují organismus a odstraňují únavu. Tyto nápoje obvykle nemají dodávat energii díky kalorické hodnotě živin, které obsahují, ale díky kombinovanému účinku kofeinu, taurinu, karnitinu, vitaminů a dalších rostlinných složek. Ústřední složkou ve většině energetických nápojů je kofein, stejný stimulant, jaký nacházíme v kávě nebo čaji. Většina z těchto nápojů obsahuje 70 až 200 mg kofeinu, který stimuluje CNS a dává tělu pocit čilosti. To může zvýšit srdeční frekvenci a krevní tlak za současné dehydratace organismu. Většina lidí pozoruje nežádoucí účinky kofeinu u dávek vyšších než 200 mg, mezi které patří nespavost, bušení srdce a bolesti hlavy. Dalšími přísadami jsou např. taurin, L-karnitin, inozitol, glukuronolakton, vitaminy a různé formy přírodních antioxidantů ve formě bylinných extraktů, jako jsou guarana, ženšen nebo ginkgo biloba. Taurin je aminokyselina, která ovlivňuje přenos nervových vzruchů a má schopnost pronikat z krevního oběhu do mozku. Působí antioxidačně a dokáže odstraňovat některé těžké kovy. Je výchozí látkou pro syntézu žlučových kyselin. V kombinaci s kofeinem a cukrem stimuluje svalové kontrakce. Kreatin vytváří kreatinfosfát, který obnovuje ve svalech hladinu ATP a tím umožňuje zvyšovat jejich energetický výkon – proto je součástí energetických nápojů. Glukuronolakton se přirozeně vyskytuje ve všech pojivových tkáních a do energetických nápojů se přidává proto, že snad napomáhá štěpení glykogenu uvolňováním hormonů a dalších chemických látek a pomáhá při detoxikaci. L-karnitin, kvartérní amoniová sloučenina, jejíž biosyntéza probíhá z aminokyselin lyzinu a methioninu, je nezbytná pro transport mastných kyselin z cytosolu do mitochondrií při odbourávání lipidů pro tvorbu metabolické energie. Mezi nepříliš časté nežádoucí účinky L-karnitinu patří nevolnost, zvracení, bolesti břicha a průjem. B-vitaminy jsou látky, které pomáhají při konverzi potravin na energii, jako např. cukr, který se nachází v energetických nápojích ve značném množství. Energetické nápoje obsahují velké množství cukru, např. glukózy, která je pro organismus preferovaným zdrojem energie. Avšak příjem velkého množství cukru je spojován s diabetem. Ženšen, adaptogenní bylina, o níž je známo, že zvyšuje energii, obsahuje některé protiúnavové látky, které údajně zmírňují stres a zlepšují paměť. Energetickým nápojům a jejich komponentám jsou přičítány četné fyziologické a psychologické účinky. Některé studie ukazují významné zlepšení duševních a kognitivních výkonů, stejně jako zvýšení subjektivní bdělosti. Nadměrná spotřeba těchto nápojů může vyvolat euforii způsobenou stimulačními vlastnostmi kofeinu. Kofein také vyvolává neklid, úzkost, podrážděnost a nespavost.
Energy drinks are soft drinks advertised as boosting energy. Energy drinks usually do not emphasize energy derived from the calories they contain, but rather through a choice of caffeine, taurine, carnitine, vitamins, and different herbal supplements the manufacturer has combined. The central ingredient in most energy drinks is caffeine, the same stimulant found in coffee or tea. Most of these drinks contain between 70 and 200 mg of caffeine, which stimulates the central nervous system giving the body a sense of alertness. It can raise heart rate and blood pressure while dehydrating the body. A lot of people experience side effects above 200 mg, which include sleeplessness, heart palpitations, and headaches. Other common ingredients are for example taurine, L-carnitine, glucuronolactone, vitamins, and various forms of natural anti-oxidants in the form of herbal extracts as guarana, ginseng or Ginkgo biloba. Taurine is an amino acid that helps regulate heartbeat, muscle contractions, and energy levels. Creatine helps with supplying energy to the muscles and is usually found in energy drinks and products that are marketed to body builders. Glucuronolactone occurs naturally in all connective tissue and it is added to energy drinks because is believed to help with glycogen depletion, freeing hormones and other chemicals, and aids in detoxification. L-carnitine, quaternary ammonium compound biosythesized from the amino acids lysine and methionine is required for the transport of fatty acids from the cytosol into the mitochondria during the breakdown of lipids for the generation of metabolic energy. Rare side effects of L-carnitine include nausea, vomiting, abdominal pain, and diarrhea. B-vitamins are essentially the things that help convert food to energy, like sugar which is found in abundance in energy drinks. Energy drinks contain a lot of sugar, for example glucose which is the body’s preferred fuel. However, too much sugar intake has been linked to diabetes. Ginseng, an adaptogenic herb, is known to increase energy, has some anti-fatigue components, supposedly relieves stress, and increases memory. A variety of physiological and psychological effects have been attributed to energy drinks and their ingredients. Some studies reported significant improvements in mental and cognitive performance as well as increased subjective alertness. Excess consumption of these drinks may induce euphoria caused by stimulant properties of caffeine. Caffeine also induces agitation, anxiety, irritability, and insomnia.
