In the previous article, the author demonstrated the close relationship between bloodletting practices in medieval Europe and acupuncture in Traditional Chinese Medicine. This study aimed to explore how acupuncture-based treatment was applied in medieval Europe. The author hypothesizes that the physical stimulation of acupuncture points associated with bloodletting was one of the main methods of pain management at that time. The study examined the indications for phlebotomy as depicted in the original illustration from Practica Medicinalis written by the 15th-century Archbishop of Prague, Sigismundus Albicus, supplemented by two other European medieval medical manuscripts. A total of 76 distinct symptoms (corresponding to 25 bloodletting acupuncture points) from the Practica Medicinalis illustration were assembled into four groups: 1) Pain and inflammation symptoms; 2) Symptoms commonly associated with pain and inflammation; 3) General symptoms affecting various organs and functions; and 4) Conditions unrelated to pain or inflammation. Among the 76 symptoms and 25 acupuncture points, only nine symptoms and a single bloodletting point were not associated with the treatment of pain or inflammation. This suggests that acupuncture-based therapy was an effective method for managing pain and inflammation in the Middle Ages and that such treatment could still be valuable from a modern clinical perspective.

BACKGROUND: Inflammation-induced testicular damage is a significant contributing factor to the increasing incidence of infertility. Traditional treatments during the inflammatory phase often fail to achieve the desired fertility outcomes, necessitating innovative interventions such as cell therapy. METHODS: We explored the in vivo properties of intravenously administered Sertoli cells (SCs) in an acute lipopolysaccharide (LPS)-induced inflammatory mouse model. Infiltrating and resident myeloid cell phenotypes were assessed using flow cytometry. The impact of SC administration on testis morphology and germ cell quality was evaluated using computer-assisted sperm analysis (CASA) and immunohistochemistry. RESULTS: SCs demonstrated a distinctive migration pattern, importantly they preferentially concentrated in the testes and liver. SC application significantly reduced neutrophil infiltration as well as preserved the resident macrophage subpopulations. SCs upregulated MerTK expression in both interstitial and peritubular macrophages. Applied SC treatment exhibited protective effects on sperm including their motility and kinematic parameters, and maintained the physiological testicular morphology. CONCLUSION: Our study provides compelling evidence of the therapeutic efficacy of SC transplantation in alleviating acute inflammation-induced testicular damage. These findings contribute to the expanding knowledge on the potential applications of cell-based therapies for addressing reproductive health challenges and offer a promising approach for targeted interventions in male infertility.

In the present study, we focused on whether the analgesic effect of Electroacupuncture (EA) is related to the regulation of oxidative stress. We established a chronic inflammatory pain model in male rats by a single injection of complete Freund's adjuvant (CFA) and then treated the animals with daily EA stimulation at the site of "zusanli". The analgesic effect of EA was evaluated by measuring the paw withdrawal threshold (PWT) when rats received mechanical and thermal pain stimulation. The levels of inflammation-related molecules and oxidative stress-related markers in the spinal cord were measured by western blotting or ELISA kits. EA stimulation and antioxidants effectively increased the PWT in CFA rats. Co-treatment of CFA rats with the ROS donor t-butyl hydroperoxide (t-BOOH) further decreased the PWT and weakened the analgesic effect of EA. EA treatment inhibited inflammation and oxidative stress, as shown by decreased levels of tumor necrosis factor-alpha (TNF-alpha), interleukin-1beta (IL-1beta), IL-6, and MDA and increased activity of SOD and catalase. Moreover, EA reduced the expression of p-p38, p-ERK, and p-p65 and simultaneously downregulated the expression of TRPV1 and TRPV4 in CFA rats. In an in vitro study, direct stimulation with t-BOOH to the C6 cells increased the production of TNF-alpha, IL-1beta, IL-6, activated p38, ERK, and p65 and up-regulated the expression of TRPV1 and TRPV4, and these effects could be prevented by the ROS scavenger PBN. Taken together, our data indicate that the inhibition of oxidative stress and the generation of ROS contribute to the analgesic effect of EA in male CFA rats.

Background: The level of pro-inflammatory Cluster of Differentiation 68 (CD 68) could be beneficial for examination as a biomarker for identifying cartilage or knee tissue degradation in joint problems. Because Cluster of Differentiation 68 appears to be linked to cell damage in the injury area, its measurement may be an effective and sensitive tool for detecting the early development of knee osteoarthritis (KOA) in people at risk for knee osteoarthritis.Aim of the study: The study aimed to evaluate which type of PRP (Pure-PRP and leukocyte-PRP) are suitable for patients with KOA via assessing the levels of serum CD 68 concentration.Materials and Methods: Serum Cluster of Differentiation 68 level was computed using ELISA kits. The experimental study comprises 21 pure-platelet-rich plasma (P-PRP) injections,11 leukocytes platelet-rich plasma (L-PRP) injections, and 16 control groups. Ranged from 35- 75 years old. All patients with diabetes mellitus, autoimmune disease, and severe knee osteoarthritis were excluded from this study. The period of the study was between November 2021 to June 2022. This study assessed other factors such as age, sex, family inheritance, and body mass index (BMI). The level of CD 68 was measured in the serum before and after the injection for six weeks.Results: The level of the study showed CD 68 elevated before injection in patients with knee osteoarthritis. A significant decrease of CD 68 (P< 0.01-P<0.001) in the serum concentration after injection as compared to before injection. However, the concentration was significantly higher than the control.Conclusions: In conclusion, both P-PRP and L-PRP demonstrated anti-inflammatory properties. Patients in both groups experienced a significant decrease in CD 68 serum levels, however, the P-PRP was more effective than the L-PRP.

Rheumatoid arthritis (RA) is a chronic disorder manifested by joint damage due to inflammation and autoimmunity which in turn has significant impacts on a patient's lifestyle. During disease, inflammation occurs due to activation of different immune cells, including macrophages and lymphocytes. Currently, the mainstay of treatment for RA is drug therapy. However, drug options are limited due to their negative effects and the possibility of reducing therapeutic benefits over time. Therefore, an effective and tolerable alternative therapy is needed. Several natural products have been found to have anti-inflammatory and anti-oxidant effects by affecting multiple molecular targets such as transcription factors and cytokines. Cinnamon, an aromatic plant, is a popular spice used for cooking and in traditional medicine all over the world. Cinnamon is composed mostly of essential oils and various components, such as cinnamaldehyde and eugenol. This review demonstrates the anti-inflammatory activity of cinnamon components in various preclinical and clinical studies illustrating their potential role in the treatment of RA.
Introduction: Osteoid osteoma is a benign bone tumor with the classical characteristic of pain that subsides significantly with the use of nonsteroidal anti-inflammatory drugs. When conservative therapy fails, a surgical approach is then recommended. Radiofrequency ablation (RFA) has become more widely used compared to open resection due to fewer serious postoperative complications. But it is still important that the complications of RFA be recognized and addressed. Case report: We present a case of a 22-year-old man with acute pain on his left shin, accompanied by signs of localized inflammation. The clinical findings and radiology support the diagnosis of osteoid osteoma. A surgical intervention with percutaneous radiofrequency ablation was performed. However, post-operatively, the patient complains of prolonged fluid discharge from the surgical site. Following the biopsy and debridement surgery, both specimen culture and histopathology results revealed sterile inflammation with no specific process. Conclusion: RFA has become the most popular treatment of choice for osteoid osteoma, but it still comes with complications, most commonly involving subcutaneous bones such as the tibia. In conclusion, extra caution is needed when treating subcutaneously located bones with RFA.

Compelling evidence supports the health benefits of physical exercise on the immune system, possibly through the molecules secreted by the skeletal muscles known as myokines. Herein, we assessed the impact of exercise interventions on plasma Heat shock protein 90 (Hsp90) levels in 27 patients with idiopathic inflammatory myopathies (IIM) compared with 23 IIM patients treated with standard-of-care immunosuppressive therapy only, and in 18 healthy subjects undergoing strenuous eccentric exercise, and their associations with the traditional serum markers of muscle damage and inflammation. In contrast to IIM patients treated with pharmacotherapy only, in whom we demonstrated a significant decrease in Hsp90 over 24 weeks, the 24-week exercise program resulted in a stabilization of Hsp90 levels. These changes in Hsp90 levels were associated with changes in several inflammatory cytokines/chemokines involved in the pathogenesis of IIM or muscle regeneration in general. Strenuous eccentric exercise in healthy volunteers induced a brief increase in Hsp90 levels with a subsequent return to baseline levels at 14 days after the exercise, with less pronounced correlations to systemic inflammation. In this study, we identified Hsp90 as a potential myokine and mediator for exercise-induced immune response and as a potential biomarker predicting improvement after physiotherapy in muscle endurance in IIM.
