Pri diagnostike niektorých primárnych a metastázujúcich malígnych melanómov môžu vznikať problémy vyvolané cytologickými vlastnosťami alebo „chýbaním" syntézy melanínu. Nakofko expresia „klasickej" kombinácie protilátky proti S-100 proteinu a HMB-45 môže príležitostne zlyhať, odskúšali sme sériu šiestich komerčne dostupných protilátok doteraz neporovnávanú inými autormi v troch najčastejších podtypoch malígneho melanómu. Naša semikvantitativna analýza sa týkala 104 bioptických materiálov z kožného malígneho melanómu (40 nodulámych malígnych melanómov, 46 superficiálne sa šíriacich malígnych melanómov a 18 lentigo maligna melanómov), v ktorých sme určovali počet ©farbených buniek a intenzitu expresie pri použití protilátok proti S-100 proteinu, HMB-45, NKI/C3, NKI/beteb, MARTI (Melan A), KBA 62 a nekomerčnej protilátky ovinuJ^t -^^ testovanú skupinu protilátok považujeme za vhodný doplnok bioptickej alebo cytologickej diagnostiky malígneho melanómu kože, menovite v prípadoch zlyhania tradičnej dvojkombinácie protilátky proti S-100 proteinu a HMB-45. V našich zisteniach sa najúčinnejšou ukázala protilátka proti KBA 62 sledovaná protilátkou proti MARTI (Melan A) a NKI/C3.
In certain primary and metastatic malignant melanomas diagnostic problems may arise due to their cytologic features and/or absence of synthesis of melanin. As the „classic" combination of S-100 protein and HMB-45 may occasionally fail to stain cells of malignant melanoma, we have tested a series of commercially accessible antibodies which were so far not compared by other authors in the three most frequent subtypes of this tumor. In surgical specimens fi'om 104 cutaneous malignant melanomas (40 nodular melanomas, 46 superficiaUy spreading malignant melanomas and 18 lentigo maligna melanomas) the staining intensity and the proportion of neoplastic cells stained with antibodies to S-100 protein, HMB-45, NKI/C3, NKI/beteb, MART 1 (Melan A), KBA 62 and Mitf was semiquantitatively analysed. The use of this group of antibodies against melanoma-associated antigens revealed it to be a favourable supplement for the bioptical or cytological diagnosis of malignant melanoma in case the traditional/conventional combination of S-100 protein and HMB-45 antibody fails. According to the authors' experience the antibody against KBA 62 has shown to be the most effective antibody followed by the antibodies against MARTI (Melan A) and NKI/C3.
Leptin is a circulating pleiotropic hormone that play an important role in appetite control, fat metabolism, regulation of body weight, fetus growth, growth and aging of adults and hematopoiesis. It is expressed abundantly and specifically in the adipose tissue. A liver cell with developed steatosis represents a cell metabolism similar to metabolism of cells of adipose tissue. Analyses of serum leptin and free leptin receptor in the serum of patients with steatosis showed significant variations from reference limits of normal values. However in liver tissue with verified steatosis detection of mRNA gene for leptin was not proven. Such expression of ob gene for leptin was not found even in the liver tissue without steatosis. With respect to the absence of ob gene expression, the direct effect of ob gene expression on other parameters of leptin metabolism could not be evaluated. The RT-PCR method with verified specificity and satisfying sensitivity was developed. The results obtained from analysis of serum leptin and free leptin receptor in the serum are presented and evaluated. The used methods were verified and reference limits for Czech population were defined in dependence on age and other clinical parameters.

The Cathepsin-D proteinase was demonstrated immunohistochemically in cases of intestinal coccidiosis in chickens; its expression concerned almost the cyto-invasive stages of reproductive cycle of several strains of coccidia (merozoites of the II-nd generation and also microgametes). The authors consider Cathepsin-D as a potentional "penetration/spreading factor", being significant for intracellular invasion and spreading of coccidia into host cells. In diagnostic practice, an immunohistochemical demonstration of Cathepsin-D is a relatively simple and sensitive method, which brings information on the extent of parasitic lesions, and on participation of particular evolutive forms of coccidial life cycle; it may also be useful in proving isolated coccidia in the intestinal epithelium, poorly recognized in routine HE preparation. Some Cathepsin-D protease reactivity in intestinal Cryptosporidiosis is also briefly mentioned. The examined cases of cerebral toxoplasmosis and primary acanthamoebic meningoencephalitis (PAME) showed in above mentioned methods negative results.

This review article presents colorectal cancer epidemiology, describing main epidemiologic characteristics and the influence of environmental and life style risk factors. The most important ideas for primary prevention are listed at the end of the article.
