Sledovali sme potlačenie bakteriálneho rastu siedmich kmeňov Acinetohacter spp. po 30 min účinku tobramycínu v suprainhibičných koncentráciách (postantibiotický účinok - PAE) a v supra-subinhibičných koncentráciách (postantibiotický účinok subinhibičných koncentrácií - PA SME) ako aj zmeny v povrchovej hydrofobicite, v produkcii lipázy a histamínu u týchto exponovaných kmeňov. Sledované farmakodynamické parametre (PAE a PA SME) ako aj zmeny v testovaných vlastnostiach Študovaných kmeňov boli závislé na koncentrácii antibiotika a na kmeni. Potlačenie bakteriálneho rastu po účinku tobramycínu v koncentrácii 2x MIC sa pohybovalo v rozsahu od 0,6-4,5 h, vyššia koncentrácia (4x MIC) indukovala dlhšie PAE (1,9-5,4 h). Tobramycín v supra-subinhibičných koncentráciách (2x MIC + 0,2x MIC a 4x MIC + 0,2x MIC) spôsobil úplnú supresiu bakteriálneho rastu. U väčšiny testovaných kmeňov sa po účinku tobramycínu zistilo zvýšenie hydrofobicity hodnotenej adherenciou baktérií na xylén, ako aj zvýšenie lipolytickej aktivity. Tobramycín v testovaných koncentráciách neovplyvnil produkciu histamínu.
Suppression of bacterial growth in seven strains of the Acinetobacter species after 30 min treatment with tobramycin at suprainhibitory concentrations (postantibiotic effect - PAE) and at supra-subinhibitory concentrations (postantibiotic effect of subinhibitory concentrations - PA SME) as well as changes in surface hydrophobicity and in the production of lipase and histamine in the exposed strains were studied. Pharmacodynamic parameters (PAE and PA SME) as well as changes in bacterial characteristics tested were dependent on antibiotic concentration and on the strain used. Suppression of bacterial growth after treatment with tobramycin at 2x MIC was in a range of 0.6-4.5 h, a higher concentration (4x MIC) induced a longer PAE (1.9-5.4 h). Tobramycin at supra-subinhibitory concentratinos (2x MIC + 0.2x MIC and 4x MIC + 0.2x MIC) caused total suppression of bacterial growth. In the majority of the tobramycin-treated strains, an increase in hydrophobicity manifested by adherence of bacteria to xylene as well as an increase in hpolytic activity was observed. Tobramycin at the concentrations tested did not affect the production of histamine.
BACKGROUND: A crucial step in conserving biodiversity is to identify the distributions of threatened species and the factors associated with species threat status. In the biodiversity hotspot of the Himalaya, very little is known about which locations harbour the highest diversity of threatened species and whether diversity of such species is related to area, mid-domain effects (MDE), range size, or human density. In this study, we assessed the drivers of variation in richness of threatened birds, mammals, reptiles, actinopterygii, and amphibians along an elevational gradient in Nepal Himalaya. RESULTS: Although geometric constraints (MDE), species range size, and human population density were significantly related to threatened species richness, the interaction between range size and human population density was of greater importance. Threatened species richness was positively associated with human population density and negatively associated with range size. CONCLUSIONS: In areas with high richness of threatened species, species ranges tend to be small. The preponderance of species at risk of extinction at low elevations in the subtropical biodiversity hotspot could be due to the double impact of smaller range sizes and higher human density.

The Deltaretrovirus genus of retroviruses (family Retroviridae) includes the human T cell leukemia viruses and bovine leukemia virus (BLV). Relatively little is known about the biology and evolution of these viruses, because only a few species have been identified and the genomic 'fossil record' is relatively sparse. Here, we report the discovery of multiple novel endogenous retroviruses (ERVs) derived from ancestral deltaretroviruses. These sequences-two of which contain complete or near complete internal coding regions-reside in genomes of several distinct mammalian orders, including bats, carnivores, cetaceans, and insectivores. We demonstrate that two of these ERVs contain unambiguous homologs of the tax gene, indicating that complex gene regulation has ancient origins within the Deltaretrovirus genus. ERVs demonstrate that the host range of the deltaretrovirus genus is much more extensive than suggested by the relatively small number of exogenous deltaretroviruses described so far, and allow the evolutionary timeline of deltaretrovirus-mammal interaction to be more accurately calibrated.

Introduction of alien species into new areas can have detrimental effects on native ecosystems and impact the native species. The present study aims to identify coccidia infecting native and introduced squirrels in Italy, to gain insight into possible transmission patterns and role of monoxenous coccidia in mediating the competition between alien and native hosts. We collected 540 faecal samples of native red squirrels, Sciurus vulgaris, invasive alien grey squirrels, S. carolinensis, and introduced Pallas's squirrels, Callosciurus erythraeus. Total prevalence of Eimeria spp. was 95.6% in S. vulgaris, 95.7% in S. carolinensis and only 4.1% in C. erythraeus. Morphological examination revealed 3 Eimeria morphotypes. Phylogenetic analyses of Eimeria DNA based on 18S, ITS, cox I markers displayed fairly distinct monophyletic clades in the microscopically indistinguishable E2 morphotype, proving indisputable distinction between the isolates from red and grey squirrels. Grey squirrels successfully introduced E. lancasterensis from their native range, but this species does not spill over to native red squirrels. Similarly, there is no evidence for the transmission of E. sciurorum from red to grey squirrels. The possible transmission and the potential role of monoxenous coccidia in mediating the competition between native and invasive squirrels in Italy were not confirmed.

Paraberrapex atlanticus sp. n. (Cestoda: Lecanicephalidea) is described from the spiral intestine of the angel shark Squatina guggenheim Marini from coastal waters off Buenos Aires Province, Argentina. Paraberrapex atlanticus sp. n. can be distinguished from the only species described in the genus, P. manifestus Jensen, 2001 in having cocoons 5-6 times longer with more eggs per cocoon, the extension of the uterine duct, the distribution of vitelline follicles, and the size and density of microtriches on the bothridial surfaces. The presence of P. atlanticus sp. n. in S. guggenheim confirms the specificity of Paraberrapex Jensen, 2001 for squatiniform sharks.

Two new genera and three new species of paramphistomoid digeneans are described in the family Cladorchiidae Fischoeder, 1901 from doradid, heptapterid, pimelodid and pseudopimelodid fishes in the Amazon River in Peru. Goeldamphistomum amazonum n. g., n. sp. (type-species) from Goeldiella eques (Müller & Troschel) (type-host) and Tenellus trimaculatus (Boulenger), and Goeldamphistomum peruanum n. g., n. sp. from Duopalatinus peruanus Eigenmann & Allen (type-host), Calophysus macropterus (Lichtenstein) and Microglanis sp. are placed in the Dadayiinae Fukui, 1929. Both species have an accessory acetabular sucker, which distinguishes the genus from all taxa previously reported from South American freshwater fishes. They differ from each other primarily in that G. amazonum has a prebifurcal genital pore and oblique, separated testes, the levels of which rarely overlap longitudinally, whereas G. peruanum has a postbifurcal genital pore and testes directly to obliquely tandem. Iquitostrema papillatum n. g., n. sp. (Kalitrematinae Travassos, 1933) from the intestine of Hassar orestis (Steindachner) differs from other members of the subfamily in the combination of a massive acetabulum with a papillate luminal surface and symmetrical testes which overlie the caeca close to the caecal arch. These are the first records of paramphistomes from the five host species studied here.

Survival at high latitude requires the capability to cope with seasonally imposed stress, such as low winter temperatures or large temperature fluctuations. The Colorado potato beetle, Leptinotarsa decemlineata, is an invasive pest of potato that has rapidly spread from low latitudes to higher latitudes. During the last 30 years, a decrease in range expansion speed is apparent in Europe. We use a comparative approach to assess whether this could be due to an inability of L. decemlineata to cope with the harsher winters encountered at high latitude, when compared to two native northern chrysomelid beetles with similar overwintering ecology. We investigated several cold-tolerance-related physiological traits at different time points during winter. Cold tolerance followed a latitudinal pattern; the northern species were more tolerant to short-term subzero temperatures than the invasive L. decemlineata. The other northern species, the knotgrass leaf beetle, Chrysolina polita, was found to tolerate internal freezing. Interestingly, the pattern for overwinter survival at 5°C was the opposite and higher in L. decemlineata than the northern species and could be related to behavioral differences between species in overwintering location selection and a potential physiological trade-off between tolerance to cold shock and to chronic cold exposure. Furthermore, while the northern species accumulated large amounts of different sugars and polyols with probable cryoprotectant functions, none were detected in L. decemlineata at high concentrations. This lack of cryoprotectant accumulation could explain the difference in cold tolerance between the species and also suggests that a lack of physiological capacity to tolerate low temperatures could slow further latitudinal range expansion of L. decemlineata.

Wildlife-associated Cryptosporidium are an emerging cause of cryptosporidiosis in humans. The present study was undertaken to determine the extent to which North American tree squirrels and ground squirrels host zoonotic Cryptosporidium species and genotypes. Fragments of the Cryptosporidium 18S rRNA and actin genes were amplified and sequenced from fecal samples obtained from three tree squirrel and three ground squirrel species. In tree squirrels, Cryptosporidium was identified in 40.5% (17/42) of American red squirrels (Tamiasciurus hudsonicus), 40.4% (55/136) of eastern gray squirrels (Sciurus carolinensis), and 28.6% (2/7) of fox squirrels (Sciurus niger). Human-pathogenic Cryptosporidium ubiquitum and Cryptosporidium skunk genotype were the most prevalent species/genotypes in tree squirrels. Because tree squirrels live in close proximity to humans and are frequently infected with potentially zoonotic Cryptosporidium species/genotypes, they may be a significant reservoir of infection in humans. In ground squirrels, Cryptosporidium was detected in 70.2% (33/47) of 13-lined ground squirrels (Ictidomys tridecemlineatus), 35.1% (27/77) of black-tailed prairie dogs (Cynomys ludovicianus), and the only golden-mantled ground squirrel (Callospermophilus lateralis) that was sampled. Cryptosporidium rubeyi and ground squirrel genotypes I, II, and III were identified in isolates from these ground squirrel species. In contrast to the Cryptosporidium infecting tree squirrels, these species/genotypes appear to be specific for ground squirrels and are not associated with human disease.

Successful co-introduction of a parasite and its host relies not only on presence of the parasite on host individuals in the founder population but also on the ability of both host and parasite to persist in the new area. Gyrodactylus proterorhini (Monogenea) has been successfully co-introduced with its Ponto-Caspian goby hosts (Babka gymnotrachelus, Neogobius fluviatilis, Neogobius melanostomus, Ponticola kessleri, Proterorhinus semilunaris) to many freshwater systems in Europe and is now widely distributed over four large European river basins (Danube, Rhine, Scheldt and Vistula). Within Europe, higher infection levels are documented in sites further from the native host range. In North America, however, G. proterorhini appears to be absent. Host specificity of G. proterorhini tested under natural conditions showed accidental host-switching onto local fish species (native Perca fluviatilis and non-native Perccottus glenii) in the river Vistula. Further examination of host-switching under experimental conditions, however, showed that G. proterorhini were unable to survive on non-gobiid hosts longer than 24 h. Our results indicate extremely low potential for host-switching of introduced G. proterorhini to non-gobiid hosts, at least in the freshwater systems of Central and Western Europe.

The spontaneous host-range mutants 812F1 and K1/420 are derived from polyvalent phage 812 that is almost identical to phage K, belonging to family Myoviridae and genus Kayvirus. Phage K1/420 is used for the phage therapy of staphylococcal infections. Endolysin of these mutants designated LysF1, consisting of an N-terminal cysteine-histidine-dependent aminohydrolase/peptidase (CHAP) domain and C-terminal SH3b cell wall-binding domain, has deleted middle amidase domain compared to wild-type endolysin. In this work, LysF1 and both its domains were prepared as recombinant proteins and their function was analyzed. LysF1 had an antimicrobial effect on 31 Staphylococcus species of the 43 tested. SH3b domain influenced antimicrobial activity of LysF1, since the lytic activity of the truncated variant containing the CHAP domain alone was decreased. The results of a co-sedimentation assay of SH3b domain showed that it was able to bind to three types of purified staphylococcal peptidoglycan 11.2, 11.3, and 11.8 that differ in their peptide bridge, but also to the peptidoglycan type 11.5 of Streptococcus uberis, and this capability was verified in vivo using the fusion protein with GFP and fluorescence microscopy. Using several different approaches, including NMR, we have not confirmed the previously proposed interaction of the SH3b domain with the pentaglycine bridge in the bacterial cell wall. The new naturally raised deletion mutant endolysin LysF1 is smaller than LysK, has a broad lytic spectrum, and therefore is an appropriate enzyme for practical use. The binding spectrum of SH3b domain covering all known staphylococcal peptidoglycan types is a promising feature for creating new chimeolysins by combining it with more effective catalytic domains.
