Cannabidiol (CBD), a non-psychotropic cannabinoid produced by the genus Cannabis, is a phytoceutical that activates the endocannabinoid system (ECS) through binding to CB1 and CB2 receptors. The ECS is involved in cellular homeostasis and regulates metabolic processes in virtually all mammalian tissues. Published studies on CBD focus, inter alia, on its use in prophylaxis and as an anti-inflammatory agent. Here the authors present a critical assessment of the effects of CBD on inflammatory periodontal diseases caused by bacterial virulence factors, and evaluate critically the possible benefits and drawbacks of CBD use in dentistry. Particular attention is paid to the interaction of CBD with microbially colonized oral tissues, the inflammatory response in relation to the immune response, and the destruction/regeneration of hard and soft tissues of the periodontium.

Karnozin (β-alanyl-L-histidin) je dipeptid, tvořený aminokyselinou β-alaninem a L-histidinem. Karnozin a obě aminokyseliny se vyskytují v mozku a kosterním svalstvu všech obratlovců. Karnozin v buňkách mozkové tkáně chrání intracelulární bílkoviny před jejich poškozením kyslíkovými radikály, glykací a tvorbou příčných vazeb mezi jednotlivými řetězci bílkovin. V kosterním svalstvu karnozin snižuje svalovou únavu způsobenou zvýšenou koncentrací kyseliny mléčné, která vede k poklesu pH. S rostoucím biologickým věkem klesá koncentrace karnozinu jak v buňkách mozkové tkáně, tak v bílých svalových vláknech kosterního svalstva, čímž dochází k zhoršování senzorických a kognitivních funkcí a úbytku svaloviny. Tyto degenerativní procesy lze zpomalit konzumací potravin bohatých na karnozin nebo doplňováním diety β-alaninem. β-alanin určuje rychlost tvorby karnozinu a jeho koncentraci v buňkách mozkové a svalové tkáně. Článek shrnuje současné názory medicíny na roli karnozinu v procesu stárnutí lidského organismu a významu obohacování diety β-alaninem.
Carnosine (β-alanyl-L-histidine) is a dipeptide composed of the amino acid β-alanine and the L-histidine. Carnosine and both amino acids are found in the brain and skeletal muscle of all vertebrates. The physiological function of carnosine in brain tissue is to protect intracellular proteins from damage by oxygen radicals, modification by glycation and formation of cross-links between protein chains. In skeletal muscle, carnosine reduces muscle fatigue caused by increased lactic acid levels and decreased tissue pH. With increasing biological age, carnosine concentration in both brain tissue and white skeletal muscle fibres decreases. There is a loss of muscle mass (sarcopenia) and deterioration of sensory and cognitive functions. These degenerative processes can be delayed by consuming carnosine-rich foods or diet supplemented with pure β-alanine. This amino acid determines the rate of endogenic carnosine synthesis and thus its concentration in brain and muscle tissues. This article is an overview of the present knowledge on the role of carnosine in the process of aging and the importance of the diet enrichment with β-alanine or carnosine.

Black salves are escharotic skin cancer therapies in clinical use since the mid 19th century. Sanguinaria canadensis, a major ingredient of black salve formulations, contains a number of bioactive phytochemicals including the alkaloid sanguinarine. Despite its prolonged history of clinical use, conflicting experimental results have prevented the carcinogenic potential of sanguinarine from being definitively determined. Sanguinarine has a molecular structure similar to known polyaromatic hydrocarbon carcinogens and is a DNA intercalator. Sanguinarine also generates oxidative and endoplasmic reticulum stress resulting in the unfolded protein response and the formation of 8-hydroxyguanine genetic lesions. Sanguinarine has been the subject of contradictory in vitro and in vivo genotoxicity and murine carcinogenesis test results that have delayed its carcinogenic classification. Despite this, epidemiological studies have linked mouthwash that contains sanguinarine with the development of oral leukoplakia. Sanguinarine is also proposed as an aetiological agent in gallbladder carcinoma. This literature review investigates the carcinogenic potential of sanguinarine. Reasons for contradictory genotoxicity and carcinogenesis results are explored, knowledge gaps identified and a strategy for determining the carcinogenic potential of sanguinarine especialy relating to black salve are discussed. As patients continue to apply black salve, especially to skin regions suffering from field cancerization and skin malignancies, an understanding of the genotoxic and carcinogenic potential of sanguinarine is of urgent clinical relevance.

U pacientů s roztroušenou sklerózou (RS) se neurologický deficit projevuje širokým spektrem klinických potíží, včetně mikčních dysfunkcí. Infekce močových cest (IMC) zhoršují kvalitu života pacientů a mohou významně ovlivnit průběh onemocnění. Pilotní, randomizovaná a placebem kontrolovaná studie ověřila, že šestiměsíční užívání 1,5 g (odpovídá 75 g čerstvého plodu) klikvy velkoplodé denně mělo vliv na kvalitu života a na možné snížení rizika onemocnění infekcí močových cest pacientů s RS. Průkaz profylaxe IMC plodem V. macrocarpon však vyžaduje rozsáhlejší klinickou studii.
In patients with multiple sclerosis (MS), neurological deficit is manifested by a wide range of clinical complaints, including voiding dysfunction. Urinary tract infections (UTIs) impair the quality of life of patients and may significantly affect the course of the disease. A pilot, randomized and placebo-controlled trial validated that a six-month course of 1.5 g (an equivalent of 75 g of fresh fruit) of the large cranberry daily had an effect on the quality of life and a possible reduction in the risk of urinary tract infections in patients with MS. However, a more extensive clinical trial is required to demonstrate the prophylaxis of UTIs with V. macrocarpon.
