The objectives of this study were to evaluate the relationships among urea and acetone content in cows' cervical mucus (CM), its crystallization type (CT) and sperm survival (SS) after timed AI. Samples of CM were collected from 192 Holstein cows treated by Ovsynch(®) protocol. Analysis of the urea and acetone content for identification of the metabolic status, the arborization test for evaluation of insemination timing and the short-term heat test of SS for assessment of its suitability as a biological matrix were performed. The data set was analyzed by the GLM procedure using SAS(®). The results documented the existence of substantial differences in individual response to the Ovsynch(®) protocol causing insemination of 55.2% cows at an inappropriate time. The urea content was found as a possible indicator of a cow's metabolism and/or of insemination timing, concentrations of less than 500 mg/L corresponded (P<0.05-0.01) to the cows' expected response to timed AI. The greater the urea content, the greater the proportion of cows inseminated at an inappropriate time. Effects (P<0.05-0.01) of CT, urea and acetone content on SS were determined. The greatest values of SS were detected in cows with an expected response to precisely timed oestrus documented by the corresponding CT. Greater values of urea (>260 mg/L) and acetone (>5mg/L) negatively affected SS as well (P<0.05-0.01). The results confirmed that the accuracy of insemination timing can be affected by the metabolism intensity, just as CM quality directly influences sperm survival.

Methotrexate (MTX) remains a mainstay in the treatment of children with hematological malignancies. The availability of an antidote/rescue agent, leucovorin (LV) has allowed escalation of MTX doses to achieve enormous plasma concentrations, compared with plasma folate. However, a recent review of more than 40 trials for children with ALL concluded that the addition of high dose MTX (HDMTX) in many different doses and schedules did not improve CNS therapy and made only minor improvements in systemic therapy for children with ALL [11]. Some assessment suggested that by HDMTX benefits only limited amount of children with ALL. Recent treatment schedules vary markedly in terms of timing, dosing and scheduling of MTX and/or leukovorin, which may leave us uncertain with ideas such as "how should we best use HDMTX and LV?" or "why are we still using such by industry recommended doses of MTX?" The answer of how best to incorporate HDMTX and/or LV into ALL treatment plans is still not known and further clinical and pharmacological studies dealing with still controversial systemic MTX issue are actual even now, after more than 5 decades of clinical experiences with the MTX in pediatric oncology.

A report on three manuscripts written in German by Doc. Dr. J. Franta (1899-1945) in prison after his death verdict (May 7, 1943) for his activity in the resistance movement. The first manuscript--a contribution on the genesis and medicamentous treatment of cataract--was a conspectus (16 pp.). After a positive review by prof. Dr. H. Rieger it served as the basis for five chapters which formed the second manuscript (52 pp.). The working hypothesis on the influence of infrared radiation on the intraocular metabolism and the suggested detailed local and general, clinical and experimental examination of subjects with a transparent and opaque lens was important. In the third manuscript (12 pp.) the author outlined his plan of biochemical examinations in keratoplastic operations, in particular as regards material from donors. Even after reading Franta's letters from prison addressed to his family, the circumstances under which the manuscripts were written are not quite clear--permission to do research given to a person sentenced to death is quite exceptional.
