Susceptibility of multidrug-resistant isolates of Klebsiella pneumoniae to silver nitrate
Protective effect of the lipopolysaccharide (LPS) antigen of Klebsiella pneumoniae was tested against ascending-mode urinary tract infection in BALB/c and LACA strains of mice. LPS was given by two different routes; LPS was found to be protective (whatever the application route) since colonization with the challenge organism was significantly lower in both cases as compared with unimmunized mice. A maximum decrease in bacterial count in the kidney of LPS-treated animals was observed on challenge after a 4-d treatment.

A silver-resistant mutant of Klebsiella pneumoniae B-5 was produced by passaging in nutrient broth containing graded concentrations of silver nitrate up to 150 ppm. The development of silver resistance in the strain resulted in rough colonies, decrease in cell size, carbohydrate content and change in klebocin pattern. The virulence of the AgR strain as checked by the burn wound model decreased as the mutant could not establish itself in the skin and spleen of the animals and the organism was cleared more efficiently by human lymphocytes than the parent AgS strain.

The virulence of a silver-ion-resistant (AgR) clinical isolate of Pseudomonas aeruginosa and its silver-sensitive mutant (AgS) was compared in the murine burn-wound sepsis model. The AgR strain showed significantly higher colonization of burned skin vis-a-vis the AgS strain. The altered pathogenicity may in part be due to the decrease in capsular content and cell surface hydrophobicity, accompanying the loss of silver ion resistance. The AgR and AgS strains, however, did not exhibit quantitatively significant differences in the production of extracellular hemolysins and proteinases.
