Předmětem řešení projektu je studium vzájemných interakcí tumorsupresorového proteinu p53 a prozánětlivého cytokinu interleukinu-1alfa včetně charakterizace vlivu histonacetyltransferáz na tyto interakce. Součástí projektu je i výzkum možnosti ovlivněnífunkčních interakcí p53 s IL-1alfa pomocí specifických nízkomolekulárních inhibitorů. Projekt předpokládá využití sofistikovaných metod molekulární a buněčné biologie včetně imunoprecipitace, konfokální fluorescenční mikroskopie, RNA interference, qRT-PCR a systematického prohledávání kombinatorické knihovny chemických látek potenciálně využitelných v protinádorové terapii.; This project aims to study the interaction between the tumour supressor protein p53 and the proinflammatory cytokine IL-1alpha including the characterization of the influence of histone acetyltransferases on this interaction. Part of the project involvesalso testing the possibility of influencing the functional interaction of p53 and IL-1alpha with specific low-molecular-weight inhibitors. The project supposes the application of sophisticated molecular and cell biology techniques including immunoprecipitation, confocal fluorescence microscopy, RNA interference, qRT-PCR and systematically searching the combinatorial library of drug-like chemical compounds suitable for use in the anti-tumour therapy.
