Současné uplatnění rituximabu ve farmakoterapii

Rituximab je monoklonální protilátka, která byla jako první registrována k léčbě hematologických malignit. Váže se na povrch B-lymfocytů, které mají na povrchu antigen CD20. V roce 1997 byl schválen FDA (Food and Drug Administration) k použití v léčbě pro relabovaný nebo refrakterní folikulární lymfom. Výsledky dalšího zkoumání vedly později k rozšíření indikace na oblast revmatologie a nefrologie. Zavedení do praxe přineslo obrat v léčbě a změnilo osud nemocných, zejména minimalizací toxicity a vyšší specifitou účinku. Cílená léčba monoklonálními protilátkami se dnes stala (standardní) součástí léčebných postupů u řady onemocnění.
Rituximab is the first registered monoclonal antibody in the treatment of hematologic malignancies. It binds to the surface of B-lymphocytes that have CD20 antigen on the surface. It was approved by the FDA (Food and Drug Administration) in 1997 for the treatment of relapsing or refractory follicular lymphoma. The results of further investigations extended its indication to other specialities such as rheumatology and nephrology. Its introduction in the practice represented a turning point in the treatment and changed the patients’ fate, particularly due to its minimal toxicity and higher effect specificity. Targeted monoclonal antibody therapy has now become a regular (standard) part of medical practice in a variety of diseases.
