Cytochrome P450 2A13 is an omitted brother of CYP2A6 that has an important role in the drug metabolism of liver. Due to extrahepatic expression, it has gained less attention than CYP2A6, despite the fact that it plays a significant role in toxicant-induced pulmonary lesions and, therefore, lung cancer. The purpose of this mini-review is to summarize the basic knowledge about this enzyme in relation to the substrates, inhibitors, genetic polymorphisms, and transcriptional regulation that are known so far (September 2021).

Dihydrofuranocoumarin, chalepin (1) and furanocoumarin, chalepensin (2) are 3-prenylated bioactive coumarins, first isolated from the well-known medicinal plant Ruta chalepensis L. (Fam: Rutaceae) but also distributed in various species of the genera Boenminghausenia, Clausena and Ruta. The distribution of these compounds appears to be restricted to the plants of the family Rutaceae. To date, there have been a considerable number of bioactivity studies performed on coumarins 1 and 2, which include their anticancer, antidiabetic, antifertility, antimicrobial, antiplatelet aggregation, antiprotozoal, antiviral and calcium antagonistic properties. This review article presents a critical appraisal of publications on bioactivity of these 3-prenylated coumarins in the light of their feasibility as novel therapeutic agents and investigate their natural distribution in the plant kingdom, as well as a plausible biosynthetic route.

Coumarin belongs to a class of lactones that are fundamentally comprised of a benzene ring fused to an α-pyrone ring; these lactones are known as benzopyrones. Similarly, coumarin has a conjugated electron-rich framework and good charge-transport properties. Plants produce coumarin as a chemical response to protect themselves from predation. Coumarins are used in different products, such as cosmetics, additives, perfumes, aroma enhancers in various tobaccos and some alcoholic drinks, and they play a relevant role in natural products and in organic and medicinal chemistry. In addition, as candidate drugs, many coumarin compounds have strong pharmacological activity, low toxicity, high bioavailability and better curative effects and have been used to treat various types of diseases. Various endeavors were made to create coumarin-based anticoagulant, antimicrobial, antioxidant, anticancer, antidiabetic, antineurodegenerative, analgesic and anti-inflammatory agents. A class of chemical compounds called furocoumarins has phototoxic properties and is naturally synthesized via the fusion of coumarin to a furan ring in different plant species. Psoralens belong to the furocoumarin class and occur naturally in various plants, e.g., lemons, limes, and parsnips. Angelicin is an isomer of psoralens, and most furocoumarins, e.g., xanthotoxin, bergapten, and nodekenetin, are derivatives of psoralens or angelicin. The present work demonstrated that psoralen molecules exhibit anti-tumoral activity against breast cancer and influence different intracellular signals to maintain the high survival of breast cancer cells. Psoralens perform different functions, e.g., antagonize metabolic pathways, protease enzymes, and cell cycle progression and even interfere in the crosslinking between receptors and growth factor mitogenic signaling.

Psoriázu v oblasti vlasové pokožky, nehtů, dlaní a plosek řadíme mezi obtížně léčitelné formy psoriázy. Frekvence výskytu těchto forem psoriázy (kromě postižení vlasové pokožky) nebývá tak častá jako plakovitá forma v typické predilekční lokalizaci, avšak svým umístěním bývá pro pacienty velmi znevýhodňující. Představuje velkou psychosociální zátěž vzhledem k viditelnosti projevů a postižené omezuje při práci i v běžném životě. Možnosti terapie jsou v těchto lokalizacích limitované. Vzhledem k horší prostupnosti extern přes nehtovou ploténku, vlasovou bariéru a silnou rohovou vrstvu dlaní a plosek bývá lokální léčba často neúspěšná a je nutné volit celkovou terapii s možným výskytem nežádoucích účinků. V článku zmíníme přehled nejčastějších terapeutických postupů u jednotlivých lokalizací obtížně léčitelné formy psoriázy a ilustrativní kazuistiku pacientky s nehtovou formou psoriázy.
Psoriasis of scalp, nails, palms and soles belong to group called difficult to treat psoriasis. Incidence of these types of psoriasis is not as frequent as typical plaque psoriasis, but it means a big psychosocial impact on the patient. For patient it presents limitation at work and daily routine. Possibilities of treatment are mostly limited by its localization (barrier of hair, nail plate and strong hyperkeratosis of palms and soles), it is often resistant to therapy and shows frequent relapse. Systemic treatments are necessary in case the topical therapy is not successful considering the higher psychological impact on the patient. In this article we mention review of the most frequent treatment approaches in each localization and an ilustrative case report of patient with nail psoriasis.

Skin explants are a suitable model which can replace dermatological experiments on animals or human volunteers. In this study, we searched for a fast, cheap and reproducible method for screening skin explant viability after treatment with UVA radiation or/and chemical agents. We compared frequently used methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), neutral red (NR) and lactate dehydrogenase (LDH) activity assay with a rarely used 2,3,5-triphenyl-2H-tetrazolium chloride (TTC) assay for the evaluation of UVA radiation and/or chlorpromazine and 8-methoxypsoralen effect as model agents. Histological analysis of skin explants was also performed by a simple haematoxylin-eosin method. Only the TTC assay was able to show the toxicity of model agents in a dose- and concentration-dependent manner. LDH assay was partially able to demonstrate results comparable to the TTC method, however, the agents' effect was less pronounced. The MTT and NR assays completely failed in the evaluation. Haematoxylin-eosin staining showed discrete structural changes in samples treated with UVA alone and CPZ+UVA, but only after 48h. Therefore, the method is not useful for screening of toxic or phototoxic effects either. In conclusion, the TTC assay was the most suitable for the evaluation of toxicity or phototoxicity in ex vivo skin.

Fytofotodermatitida představuje různě se manifestující onemocnění, které se vyskytuje zejména u dětí a dále také u osob s potenciálním rizikem profesionální expozice. Základem pro rozvoj tohoto onemocnění je kontakt s tzv. fotosenzitizujícími látkami, které se označují jako furanokumariny a následné expozice slunečnímu záření. K rozvoji kožních projevů dochází typicky v odstupu 12–36 hodin. Kožní projevy mohou být vyjádřeny jen velmi diskrétně (erytém), ale mohou se také prezentovat jako hluboké kožní defekty s rozvojem nekrózy a bez možnosti spontánní reepitelizace. V kazuistice prezentujeme případ rozvoje tohoto syndromu u mladého chlapce.
Phytophotodermatitis is a condition with varying presentations that mainly occurs in children as well as in those with a potentialrisk of occupational exposure. The disease results from contact with photosensitizing compounds, referred to as furanocoumarins,and subsequent exposure to sunlight. Skin manifestations typically develop within 12 to 36 hours. Skin manifestations canbe very subtle (erythema), but they can also present as deep skin defects with the development of necrosis and no possibility ofspontaneous re-epithelialization. The case report presents a young boy who developed this type of syndrome.

Efficacies of the Ammi visnaga seeds extract and a majority of substances on larval Culex quinquefasciatus mortality in various development stages including pupae were studied. The effect of exposure time on larval mortality was also studied. The effect of sublethal concentrations or short exposure times on further larval development and subsequent fecundity in adults were studied as well. Lethal doses of the extract were estimated for the 2nd, 3rd and 4th instar of C. quinquefasciatus (LC50 for 18, 23 and 180 mg L(-1), respectively). The majority of furanochromenes, khellin and visnagin, were identified by analysing the extract. Khellin was significantly more effective compared to visnagin, whose LC50 was estimated at 8, 10 and 41 mg L(-1) for the 2nd, 3rd and 4th instar larvae. Khellin showed very fast efficacy on mortality for the 3rd instar larvae in a concentration of 100 mg L(-1). Fifty percent mortality was determined 30 min after application, a time which was considerably shorter compared to the extract (113 min) or visnagin (169 min). The effect of the application of lethal concentrations on C. quinquefasciatus larval mortality was studied. The least number of adults were hatched after application of the extract and khellin (41.8% and 37.9%, respectively), less than after visnagin application (46.7%) or in the control (94.2%). LC50 application caused lower fecundity in the hatched adults, lower hatchability of the eggs, and also very low natality, more than 77% lower for khellin compared to the control. A short exposure, corresponding to our estimated LT30, caused no significant acute toxicity in the larvae (until 24 h) for the extract or visnagin (4.3% and 11.5%, respectively); however, 18 min of action from khellin caused a 54.3% mortality rate of the larvae within 24 h.

Autoři předkládají současné poznatky o patogenezi fytofotodermatitid a uvádí přehled rostlin vyvolávajících tuto reakci. Fytofotodermatitidy jsou akutní zánětlivá kožní onemocnění vyvíjející se po kontaktu s rostlinami a expozici UV záření. Mohou být způsobeny řadou rostlin a rostlinných produktů, mívají bizarní a rozmanité klinické obrazy. Většinou se jedná o neimunologickou – toxickou – reakci s typickým dlouhodobým přetrváváním sytých pigmentací.
The authors present the current knowledge of the pathogenesis of phytophotodermatitis and list of plants causing this reaction. Phytophotodermatitis is an acute inflammatory skin disease resulting from a contact with plants and UV light exposure. It can be caused by a large amounts of plants and plant products. It usually has a bizarre and divergent clinical picture. In most cases it is a non-immunologic toxic reaction with a typically long lasting deep pigmentation.
