V krátkém přehledu je shrnuta problematika schistosomóz, jejich diagnostika a terapie. Vzhledem k novým trendům vývoje turistiky je totiž možné, že se i v České republice v nejbližších letech dočkáme importovaných případů této u nás zatím jen raritně diagnostikované parazitózy.
Problems of schistosomoses, their diagnosis and therapy is briefly summarized. Regarding new tourism trends it is possible, that new imported cases of such so far rarely diagnosed parasitosis can be expected in Czech Republic in forthcoming years. Mor je dodnes jednou z mála nemocí jejíž výskyt vyvolává iracionální strach a masovou paniku. Vzhledem k tomu, že se dosud vyskytuje v různých oblastech světa, že se zvýšilo cestování a že letecká doprava „zkrátila“ vzdálenosti i čas, je obava z možných zánosů této nákazy do našeho prostředí. Tyto hrozby importace umocňuje dnes strach z možného použití původce moru, Yersinia pestis, jako biologické zbraně. Bioexperti oceňují možnost výroby Y. pestis ve velkých kvantech, její stabilitu při dlouhodobějším skladování i možnost jejího šíření formou umožňující hromadné postižení obyvatelstva. Jsou obavy, že nejen některé vojenské kruhy, ale i některé teroristické skupiny se zabývají plány využití moru jako biozbraně. Nejobávanější hrozbou je rozptyl Y. pestis ve formě aerosolu s úmyslem vyvolat v postiženém obyvatelstvu vysoce letální plicní mor. Ten by se následně šířil interhumáním přenosem daleko za napadenou oblast. Protože bioteroristé neinformují předem o svých aktivitách, biologický útok morovým aerosolem by proběhl pravděpodobně utajeně. Jeho první indikací by bylo náhlé postižení většího počtu dříve zdravých osob vysokou horečkou, bolestmi v hrudníku, dusnosti, kašlem a přítomností krve ve sputu, které by končilo do 2-3 dnů smrtí. Protože se dnešní lékař ve své praxi s plicním morem nesetkal, jsou přehledně uvedeny, kromě historie, klinické, diagnostické a epidemiologické charakteristiky hlavních forem morové nákazy. Mimo standardní kontrolní postupy v případě bioteroristického útoku jsou rovněž uvedena nejdůležitější protiepidemická opatření včetně terapie postižených a postexpoziční profylaxe exponované populace. Jak pro účinnou bioobranu, tak rychlou likvidaci zánosu morové infekce, je rozhodující rychlost prvotní diagnózy, jež závisí především na připravenosti a diagnostických znalostech terénních lékařů.
Even today plague is one of the rare diseases that breed irrational far and mass panic, whenever it crops up. Plague still exists in several region sof the Word – and with large-scale travel and the ,,shrinking“ of distances and time due to air transport, there is some concern that this infection could be brought by variol carriers to our environment. The Great of import sis today enhanced by the possible utilization of the causative agent of plague. Yersinia pestis, as a biological japon. Experts in biology explore the possibihty of producing Y. pestis m large quantities, its stability upon prolonged storage and the posibility of its dissemination in a way allowing a mass infection of the populations. There is some concern that some military circles and some terrorist groups are toying with the idea of using plague as a biological weapon. The most alarming threat is the dissemination of Y. pestis in an aerosol so as to produce in the affected population the highly lethal pulmonic plague, which would then spread by interhuman trnasmission far blond the attacked area. As bioterrorists don’t announce beforehand thein planned activities, an attack with a plague aerosol would probably come as an utter surprise. The first indication would bet he sudden affection of large numbers of previously healthy subjects, presenting with high fever, chest pain, apnoea, coughing with blood in the sputum and ending in death within 2 to 3 days. As today there are practically no physicians who have encounered in their practice pneumonic plague, the autiiors discuss not only the history of the dinase, but also the clinical, diagnostic and epidemiological characteristics of the main forms of plague infections. In addition to standard control measures in the case of a bioterrorist attack, the article discusses the chief anti-epidemic steps, including the treatment of the affected subjects and postexposure prophylaxis of the exposé population. Decisive for an efficacious biodefence and a speedy liquidation of the imported plague infection is a rapid first diagnosis, which in turn hinges on the readiness an diagnostic knowledge of first-line physicians.

The functional significance of having two nuclei in a cell is unknown. Having two stores of genetic material may be advantageous for cell growth. Nuclear protein import is at a critical juncture in the cell to modify cell growth. This study addressed the potential for differential nuclear protein import in two nuclei of the same cell. Isolated adult rat cardiomyocytes were microinjected with an exogenous fluorescent protein conjugated with nuclear localization amino acid sequences to facilitate nuclear import and its detection. Our results demonstrate the rate of nuclear protein import was not significantly different between the two nuclei in the same cell. These data demonstrate that the two nuclei are functionally similar in a binucleated cardiomyocyte, at least as far as nucleocytoplasmic transport is concerned.

The protein Isw1 of Saccharomyces cerevisiae is an imitation-switch chromatin-remodeling factor. We studied the mechanisms of its nuclear import and found that the nuclear localization signal (NLS) mediating the transport of Isw1 into the nucleus is located at the end of the C-terminus of the protein (aa1079-1105). We show that it is an atypical bipartite signal with an unconventional linker of 19 aa (KRIR X(19) KKAK) and the only nuclear targeting signal within the Isw1 molecule. The efficiency of Isw1 nuclear import was found to be modulated by changes to the amino acid composition in the vicinity of the KRIR motif, but not by the linker length. Live-cell imaging of various karyopherin mutants and in vitro binding assays of Isw1NLS to importin-α revealed that the nuclear translocation of Isw1 is mediated by the classical import pathway. Analogous motifs to Isw1NLS are highly conserved in Isw1 homologues of other yeast species, and putative bipartite cNLS were identified in silico at the end of the C-termini of imitation switch (ISWI) proteins from higher eukaryotes. We suggest that the C-termini of the ISWI family proteins play an important role in their nuclear import.
