The migratory properties of leukemic cells are commonly associated with their pathological potential and can significantly affect the disease progression. While the research in immunopathology mostly employed powerful indirect methods such as flow cytometry, these cells were rarely observed directly using live imaging microscopy. This is especially true for the malignant cells of the B-cell lineage, such as those originating from chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL). In this study, we employed open-source image analysis tools to automatically and quantitatively describe the amoeboid migration of four B-cell leukemic and lymphoma cell lines and primary CLL cells. To avoid the effect of the shear stress of the medium on these usually non-adherent cells, we have confined the cells using a modified under-agarose assay. Surprisingly, the behavior of tested cell lines differed substantially in terms of basal motility or response to chemokines and VCAM1 stimulation. Since casein kinase 1 (CK1) was reported as a regulator of B-cell migration and a promoter of CLL, we looked at the effects of CK1 inhibition in more detail. Migration analysis revealed that CK1 inhibition induced rapid negative effects on the migratory polarity of these cells, which was quantitatively and morphologically distinct from the effect of ROCK inhibition. We have set up an assay that visualizes endocytic vesicles in the uropod and facilitates morphological analysis. This assay hints that the effect of CK1 inhibition might be connected to defects in polarized intracellular transport. In summary, 1) we introduce and validate a pipeline for the imaging and quantitative assessment of the amoeboid migration of CLL/MCL cells, 2) we provide evidence that the assay is sensitive enough to mechanistically study migration defects identified by the transwell assay, and 3) we describe the polarity defects induced by inhibition or deletion of CK1ε.

We determined two characteristic morphological patterns typical for the organization of microtubule systems of murine peritoneal macrophages and lymphocytes. A small number of both cell types formed uropods with an axial bundle of microtubules. This number remains practically unchanged during 48 h of cultivation in non-antigenic environment but grows rapidly in the presence of an antigen.

The palaemonid shrimp Periclimenaeus rastrifer is here reported from Taiwan based on three specimens. It bears a feeble but distinct anterodorsal lobe on the first pleonite (not reported before). The mouthparts, not previously described, are also remarkable by a multidentate apex of the mandibular incisor, asymmetric distal endite of the second maxilla, and expanded dorsal lobe on the second maxilliped merus. The species is thus redescribed here. One of the examined specimens possesses an unusual rostrum, and malformed telson and uropodal spinulation; the latter specimen is described and illustrated in detail, including the mouthparts. Previously, the only species in the genus published in faunistic repors from Taiwan were P. wolffi Bruce 1993 and P. gorgonidarum (Balss, 1913) recorded from the Taiwan Strait, and the Green Island, respectively; P. rastrifer is reported here as the first of the genus from Taiwan mainland.

Madeirasquilla tuerkayi is described as a new genus and species of the nannosquillid mantis shrimps (Stomatopoda) based on a single specimen collected from Madeira, eastern Atlantic. That specimen is remarkable by a combination of the following morphological characters: rostral plate with three sharp anterior projections; antennal protopod with two mesial and one ventral papillae; cornea subglobular; raptorial claw dactylus with 11 or 12 teeth, and with acute proximal tooth on outer margin; pleonite 6 with strong posterolateral spine and two posteriorly directed sternal spines; telson bearing smooth shield-like dorsal prominence with acute median spine posteriorly; four pairs of fixed primary teeth posteriorly on the telson; outer primary spine of uropodal protopod longer than inner primary spine. The separate position of the new genus is supported also by molecular comparison. A key to the genera of the family Nannosquillidae is proposed.

A very small adult specimen of symbiotic palaemonid shrimp collected from Kavieng Lagoon, N.W. New Ireland Island, Papua New Guinea, is described as a new genus and species. Due to the slender subcylindrical body, reduced rostrum, elongated eyes and distolaterally produced uropodal exopods, the new species is somewhat similar to shrimps of the endosymbiotic sponge genus Nippontonia. Both second pereiopods are lost, but the specimen is unique by its bidentate scaphocerites, an apical corona of spiniform teeth on the mandibular molar process, five posterior telson cuspidate setae from which the median and intermediate ones are long, hooked, and the broadly spoon-like dactyli of the first pereiopods chelae with marginal pectination. Based on those characters, the new genus is without parallel among all symbiotic palaemonid shrimps. Its close phylogenetic relationship to the spongobiotic genera Nippontonia, Onycocaridella, and Thaumastocaris, is also confirmed by molecular comparison.

Crayfish (Astacus fluviatilis Fabricius, 1793) were treated three times with a bath of diluted ammonia water and proflavine for four and six minutes at a temperature of 18 degrees C to get rid of the ectoparasitic worms of Branchiobdella parasita Henle, 1835; the hatching ability of the Branchiobdella parasita eggs was stopped after the treatment. No lethal effect of the bath on the crayfish of 5 to 14 cm was observed, the lethal effect on the young crayfish borne on the uropods of the female crayfish cannot be eliminated: during the baths the young crayfish died (the death rate was approximately 6% of the average fertility of crayfish); the young crayfish may have been damaged during the treatment operations with the crayfish. The bath of diluted ammonia water and proflavine may be recommended for the treatment of branchiobdellosis in crayfish to be planted, or after several-hour storage of crayfish supplied to the market. One replication of the above cycle is sufficient according to the results obtained. The ammonia content in the ammonia water depends also on the duration of storage and on the storage temperature--it can drop during the storage. Therefore the ammonia water should be used as soon as possible after the despatchment from the place of production; the ammonia content should be determined by means of titration before preparing the bath, and according to this content the dilution ratio should be adjusted.
