Bola vyšetrená adsorpčná účinnosť aktívneho uhlia, Amberlitu XAD-2 a XAD-4 pri hemoperfúzii paracetamolu in vitro. Celkove bolo vykonaných 30 hemoperfúzií paracetamolu in vitro v uzavretom systéme pri použití vyššie uvedených sorbentov. Ako perfúzny roztok sme použili 3000 ml 0,9% NaCl. Koncentrácia paracetamolu v perfúznom roztoku bola 604,3±3,8 mg/l. Jednotlivé hemoperfúzie trvali 5 hodín. Pri hemoperfúzii cez aktívne uhlie v 300. minúte bola koncentrácia paracetamolu v roztoku 1,4±0,1 mg/l. Adsorpčná kapacita hemoperfúznej kapsuly s použitím Amberlitu XAD-2 a XAD-4 bola vyčerpaná už v 180. minúte. Autori na základe získaných výsledkov odporúčajú použitie hemoperfúzie cez aktívne uhlie u chorých po otrave paracetamolom. Okrem toho u 17-ročnej chorej, ktorá sa otrávila v samovražednom úmysle paracetamolom, bola vykonaná 5-hodinová hemoperfúzia cez aktívne uhlie. Táto liečba viedla k rýchlemu uzdraveniu.
The absorption efficacy of active charcoal, Amberlite XAD-2, and XAD-4 was investigated during in vitro paracetamol hemoperfusion. Overall, 30 in vitro paracetamol hemoperfusion procedures were performed in a closed system using the above sorbents. 3000 mL of 0.9% NaCl was used as the perfusion solution. Paracetamol levels in the perfusion solution were 604.3±3.8 mg/L. Individual hemoperfusion procedures took 5 hours to complete. At minute 300 of active charcoal-based hemoperfusion, the paracetamol levels in the solution were 1.4±0.1 mg/L. The adsorption capacity of the hemoperfusion capsules using Amberlite XAD-2 and XAD-4 was used up as early as minute 180. Based on their results, the authors recommend to use active charcoal-based hemoperfusion in patients with paracetamol intoxication. In addition, 5-hour active charcoal-based hemoperfusion was undertaken in a 17- year-old female patient taking paracetamol in a suicidal attempt. This therapy resulted in rapid recovery of the girl.
Byl testován vliv dvou abiotických elicitorů,a to dvou nově syntetizovaných sloučenin (substituovaných anilidů pyrazin-2-karboxylových kyselin)na tvorbu flavonoidů v kalusové kultuře Ononisarvensis L.Testované sloučeniny ovlivňovaly výrazně produkci flavonoidů v kultuře in vitro .Zejména po elicitaci roztokem látky č.I –(4-hydroxyanilid 6-chlor-5-terc.butylpyrazin-2-karboxylová kyselina)v koncentraci (3,32.10 –7 mol.l –1 )a době elicitace 48 hodin došlo ke zvýšení tvorbyflavonoidů o 976 %oproti kontrole.
The effect of two abiotic elicitors –two newly synthetized compounds (substituted anilides ofpyrazine-2-carboxylic acids)–on the formation of flavonoids in the callus culture of Ononis arvensisL.was tested.The compounds markedly influenced the production of flavonoids in an in vitro culture.Particularly after elicitation with a solution of compound No.1 (4-hydroxyanilide 6-chloro-5-terc.butylpyrazine-2-carboxylic acid)in a concentration of 3.32.10 –7 mol.l –1 and within 48 hours of elicitation an increase in flavonoid formation by 976 %versus the control took place.

Bola vyšetrená adsorpčná účinnosť aktívneho uhlia, Amberlitu XAD-2 a XAD-4 pri hemoperf úzii amitriptylínu a nortriptylínu in vitro. Celkove bolo vykonaných 15 hemoperíúzií amitriptylínu a nortriptylínu in vitro v uzavretom systéme pri použití vyššie uvedených sorbentov. Ako perfúzny roztok sme použili 3000 mi 0,9% roztoku NaCI. Koncentrácie amitriptylínu a nortriptylínu v perfúznom roztoku boli 13,1 ±0,5 mg/l, resp. 12,1±0,6mg/l. Jednotlivé hemoperfúzie trvali 5 hodín. Pri hemoperfúzii cez aktívne uhlie v 300. minúte boli koncentrácie amrtnptylínu a nortriptylínu v perfúznom roztoku 2,09±0,21 mg/l, resp. 1,95±0,14. Hemoperfúzia cez Amberlite XAD2 a XAD-4 znížila koncentráciu amitriptylínu a nortriptylínu na nulové hodnoty už v 180. minúte. Extrakcie a klírensy amitriptylínu a nortriptylínu dosiahli maximálne hodnoty (extrakcia: 1,0; klírens: 200 ml/min) v 120. minúte pri hemoperfúzii cez Amberlite XAD-2 a XAD-4. Podľa našich výsledkov získaných zo štúdie in vitro odporúčame pri akútnej otrave tricyklickými antidepresívami (amitriptylínom a nortriptylínom) u ľudí použitie hemoperfúzie cez Amberlite XAD-4 alebo XAD-2 do 6 hodín po akútnej otrave. Okrem toho u 44-ročnej chorej, ktorá sa v samovražednom úmysle otrávila amitriptylínom, sme vykonali dve hemoperfúzie cez Amberlite XAD-4. Táto liečba viedla k uzdraveniu chorej.
The absorption efficacy of charcoal, Amberlite XAD-2 and Amberlite XAD-4, in hemoperfusion using amitriptyline and nortriptylin e in vitro was assessed. An NaCl solution (with a volume of 3,000 ml of 0.9% of NaCl) served as the perfusate. Amitriptyline and nortriptyline levels in the perfusate were 13.1±0.5 mg/l and 12.1±0.6mg/l, respectively. Individual hemoperfusion procedures la sted 5 hours. During charcoal-based hemoperfusion, amitriptyline and nortriptyline levels in perfusate at 300 minutes were 2.09±0.21 m g/l and 1.95±0.14 mg/l, respectively. Hemoperfusion using Amberlite XAD-2 and XAD-4 decreased amitriptyline and nortriptyline level s to zero as early as 180 minutes. Amitriptyline and nortriptyline extraction and clearance rates reached their peak values (extr action: 1.0; clearance: 200 ml/min) at 120 minutes during hemoperfusion with Amberlite XAD-2 and XAD-4. Based on the results of our in vitro study, we recommend to use tricyclic antidepressants for managing acute poisoning with amitriptyline and nortriptyline in man, and hemoperfusion with Amberlite XAD-4 or XAD-2 within 6 hours of acute poisoning. In addition, we performed two hemoperfusion procedures with Amberlite XAD-4 in a 44-year-old woman ingesting amitriptyline in a suicide attempt. The patient recovered succ ess- fully.
