The proposed project is primarily aimed at studying of the mechanism of neoadjuvant chemotherapy-induced cell death and studying of the biomarkers in predicting pathologic response to the treatment in patient with breast carcinoma. The presumption of effectivity of targeted therapy is exact identification of target structure on the molecular level. Await results of the proposed project will have a contribution in field of discovery of new molecular prognostic markers of breast carcinoma in correlation with findings of routine histopathology and with clinical outcome. Moreover, understanding of the mechanism of neoadjuvant chemotherapy-induced cell death might have impact on identification of tumor specific genes which might be used as new pharmacological targets or their detection might be used as significant fast diagnostic routine test. Breast cancer is a clinically heterogeneous disease, and existing histological classifications do not fully capture the varied clinical course of this disease.
Navrhovaný projekt se primárně zaměřuje na studium mechanismu neoadjuvantní chemoterapií indukované buněčné smrti či rezistence na chemoterapii u karcinomu prsu, s předpokladem účinnosti cílené molekulárně-biologické léčby ?targeted therapy?, na základěpřesné identifikace terčové struktury na molekulární úrovni. Druhotně se pak projekt zaměřuje na studium biomárkerů využitelných pro predikci odpovědi na terapii, jež bude v budoucnu podkladem pro odhad biologického chování onemocnění a cílený výběr léčby. Cílem navrhovaného projektu je též nalezení nových poznatků v oblasti molekulární diagnostiky a na jejich základě formulovat nové, efektivnější postupy v diagnostice nádorů prsu. Klasifikace nádorů založená na histopatologickém obraze není pro potřebymoderní léčebné koncepce dostačujcí neboť jen částečně odráží klinickou diverzitu tohoto onemocnění. Charakteristika každéhoho nádoru na molekulární úrovni se postupně stává základem diagnosticko-terapeutického procesu.
