Despite the obvious benefits of using ureteral stents to drain the ureters, there is also a risk of complications from 80-90%. The presence of a foreign body in the human body causes disturbances in its proper functioning. It can lead to biofilm formation on the stent surface, which may favor the development of urinary tract infections or the formation of encrustation, as well as stent fragmentation, complicating its subsequent removal. In this work, the effect of the polymeric coating containing the active substance-papaverine hydrochloride on the functional properties of ureteral stents significant for clinical practice were assessed. Methods: The most commonly clinically used polyurethane ureteral Double-J stent was selected for the study. Using the dip-coating method, the surface of the stent was coated with a poly(D,L-lactide-glycolide) (PLGA) coating containing the papaverine hydrochloride (PAP). In particular, strength properties, retention strength of the stent ends, dynamic frictional force, and the fluoroscopic visibility of the stent during X-ray imaging were determined. Results: The analysis of the test results indicates the usefulness of a biodegradable polymer coating containing the active substance for the modification of the surface of polyurethane ureteral stents. The stents coated with PLGA+PAP coating compared to polyurethane stents are characterized by more favorable strength properties, the smaller value of the dynamic frictional force, without reducing the fluoroscopic visibility.

The study was aimed to compare the effects of pentoxyphylline, aminophylline, choline theophyllinate and ethymizol on guinea pig and dog trachea with those of theophylline, papaverine and ephedrine. The effects of these drugs on the basal tension, on dose-response curves for muscle contraction produced by histamine and on cAMP level were investigated in guinea pig trachea, together with their influence on the resting and histamine-evoked mechanical and membrane activities of dog trachea. Like papaverine, pentoxyphylline did not alter the resting membrane potential, although it relaxed both tracheal preparations, and it antagonised the effects histamine and raised the cAMP level of the smooth muscle. The effects of ethymizol were similar to those of theophylline and its water soluble derivatives (aminophylline and choline theophyllinate). Whereas, ephedrine although it decreased the basal tension and inhibited histamine-evoked responses, also elicited substantial hyperpolarization of the smooth muscle membrane with no effect on the cAMP level. These findings are consistent with the hypothesis that cAMP has an important role in the action of some bronchodilator drugs; however, it is concluded that the possibility of contributing of their action on membrane potential to their action needs to be considered. The similarity of the potencies of ethymizol and pentoxyphylline to that of classical bronchodilators in inhibiting contraction of guinea pig and dog tracheal smooth muscle suggests that they may have a therapeutic value.

The development of rat myocardial lesion induced by isoprenaline was studied under the influence of Faustan, morphine and Spasmo-eunalgit. A protective effect diminishing the myocardial lesion was limited to morphine.

The ramus interventricularis ventralis (RIV) of the dog heart was perfused with donor blood from the femoral artery. The inflow and outflow pressure, the blood flow and the diameter of the RIV were recorded. An increase in the blood flow from 11.3 +/- 0.3 to 81.4 +/- 6.2 ml/min induced, in 24.4 +/- 1.7 s a gradual increase in RIV diameter which became stabilized in 90-120 s and amounted to 80.87 +/- 11.68 microns, i.e. to 3.37 +/- 0.70% of the resting diameter. Since the mean pressure at the site where the diameter was recorded was maintained constant, relaxation of RIV smooth muscle has been assumed to have occurred. Dilation could not be evoked after RIV had been dilated by papaverine. Any interference by changes in pressure amplitude accompanying the increase in blood flow was precluded by using non-pulsating perfusion of the RIV. An increase in the non-pulsating blood flow from 12.11 +/- 2.20 to 90.33 +/- 11.30 ml/min likewise--in 19.50 +/- 1.83 s--produced an increase in RIV diameter of 1.8%. Blood flow is being suggested as one factor regulating the tone of the smooth muscle of the major coronary artery. This mechanism is supposed to counteract sympathetic constriction of the major coronary artery.

Electrical and mechanical activity of smooth muscle of the guinea-pig caecum was recorded by means of the sucrose-gap technique. The responses of longitudinal and circular smooth muscle to acetycholine were differently affected by changes of extracellular calcium concentration (0.8, 2.5 and 7.5 mM). The contractions of both preparations were depressed at high Ca++ concentrations, whereas at low Ca++ concentrations only contractions of the circular smooth muscle were augmented. The stimulatory effect of acetylcholine was decreased by papaverine in both preparations at all three concentrations of Ca++. This inhibition was the greater the lower the concentration of extracellular Ca++ and this process was more pronounced in the circular muscle. The ability of papaverine to counteract the effect of lowered concentrations of extracellular Ca++ on membrane excitability may well explain its inhibitory effect upon intestinal smooth muscle.
