OBJECTIVE: Evaluation of clinical relevance of histopathological examination on the basis of macroscopic and microscopic analyses of changes in placentas in chosen clinical cases. DESIGN: Summarizing article. SETTING: Department of Obstetrics and Gynecology and Department of Pathology, City Hospital in Ostrava. METHODS: In this summarizing article we are presenting histopathological changes in placental tissues in connection with chosen clinical cases of obstetrics studied in the Department of Gynecology and Obstetrics in City Hospital in Ostrava from January to August 2015.After processing, placentas were examined via light microscope in the Department of Pathology. Over the period mentioned above, more than 80 human placentas were examined and compared. From found histopathological changes and their comparison with clinical cases we can deduce their straight connection to premature birth. CONCLUSION: The clinical relevance of histopathological examination of placenta was proven from the view of obstetrician, specifically in cases of pathological pregnancy or birth and preterm birth.

Samples of paravertebral muscles were obtained from the lesion sites of forty patients operated on for recurrent lumboischiadic syndrome of discogenic etiology. None of the specimens was physiological. All of them were pathologically altered in various grades of involvement. The histopathological changes were highly varied. The findings included muscle fibre atrophy of a nonspecific type, neurogenic atrophy of typical fascicular distribution. Apart from atrophic fibres there were hypertrophic ones either at the same time or in connection with another histopathological process. Changes of myopathic nature were also present with rounded up muscle fibres and shift of nuclei from the subsarcolemnic spaces into the centre of the muscle fibre, multiplication of the connective tissue and vicarious growth of adipose tissue. The visible neuromuscular spindles contained thickened connective tissue capsules and atrophied intrafusal fibres. The present authors' conclusion of their interpretation of these histopathological changes is that they are not produced by one but by a whole set of factors. What results is a cyclic nature of the changes, when it is hard to decide whether we are facing a primary cause or its sequela. The regular company of discopathies are repeated microtraumas during recurrent paravertebral contractures and spinal blocks. A not negligible part can be played also by primary muscle diseases affecting, among others, back muscles, e.g. progressive muscular dystrophy of Duchenne's and Becker's type, various myopathies, nonspecific myositis and paravertebral polymyositis, and other rheumatological involvement. These muscular diseases with disturbed proprioception causing unphysiological posture and loading the spine can share in the origin degenerative processes on intervertebral disks.

This study aimed to characterize the mechanical properties of native human ligamentum flavum (LF) and correlate them with histopathological changes. Mechanical property gradients across the cranial, medial, and caudal regions of LF were mapped and compared with histological sections. We also compared lumbar spinal stenosis (LSS) samples with disc herniation (DH) samples as reference material to identify differences in mechanical properties and histopathological features. Our results revealed significant heterogeneity in LF mechanical properties, with local variations correlating with specific histopathological changes such as chondroid metaplasia and loss of elastic fibers. These findings underscore the importance of considering LF heterogeneity in mechanical characterization and provide insights into its behavior under pathological conditions.

The asexual reproduction of Sarcocystis dispersa was studied in the liver of the house mouse. Histopathological examinations revealed infection of parenchymal cells with the parasite and diffuse cellular infiltrations with the presence of polymorphonuclear cells, monocytes and eosinophilic leukocytes. The volume and number of cellular infiltrates increased gradually from day 3 till day 10 p.i. Then appeared necrotic foci and both phenomena reached the maximum on days 10-12 p.i. On days 21-34 p.i., no changes occurred in the tissue and the liver healed without scars. The infection of common vole with Sarcocystis cernae sporocysts caused cellular infiltrations in the liver around the portobiliary veins. The parasite was present from day 3 till day 7 p.i. The histopathological picture of liver changes was similar to that in the house mouse.

The aim of the study was to evaluate relationship between infertility and the histological structure of the testes following the subcutaneous administration of different doses of sodium fluoride (5, 10, 20 and 50 mg/kg/day), for 100 days, to groups of six male albino rabbits; the six control animals were given 1 cc distilled water/kg b.w./day for the same length of time. Deficient maturation and differentiation of the spermatocytes and an increase in the amount of interstitial tissue were found in the experimental animals. In the higher dosage groups, spermatogenesis stopped and the seminiferous tubules became necrotic. The study thus established the existence of a definite relationship between fluorosis and testicular damage.

The aim of this study was to investigate effects of subchronic exposure to sublethal levels of diclofenac on growth, oxidative stress, and histopathological changes in Danio rerio. The juvenile growth tests were performed on Danio rerio according to OECD method number 215. Fish at the age of 20 days were exposed to the diclofenac environmental concentration commonly detected in the Czech rivers (0.02 mg L(-1)) and the range of sublethal concentrations of diclofenac (5, 15, 30, and 60 mg L(-1)) for 28 days. A significant decrease (P < 0.01) in the fish growth caused by diclofenac was observed in the concentrations of 30 and 60 mg L(-1). The identified value of LOEC (lowest observed effect concentration) was 15 mg L(-1) of diclofenac and NOEC (no observed effect concentration) value was 5 mg L(-1) of diclofenac. We did not find histopathological changes and changes of selected parameters of oxidative stress (glutathione S-transferase, glutathione reductase) in tested fish. The environmental concentration of diclofenac in Czech rivers did not have any effect on growth, selected oxidative stress parameters (glutathione S-transferase, glutathione reductase), or histopathological changes in Danio rerio but it could have an influence on lipid peroxidation.

OBJECTIVES: Patients with a bicuspid aortic valve (BAV) often present with a dilated ascending aorta. However, the underlying pathogenesis for the observed changes in the aortic wall and the resulting aneurysmal dilation remains a subject of debate. This study aims to compare the histological abnormalities of the ascending aorta in BAV and tricuspid aortic valve (TAV) patients and their correlation with aortic diameter and patient age. METHODS: A total of 376 patients from our institution's clinical database were included in the retrospective analysis. These patients underwent either elective surgery for ascending aorta dilation or emergency surgery for aortic dissection, either isolated or with a structurally diseased aortic valve. After excision, the ascending aorta samples were analysed by a pathologist. RESULTS: On histological examination, a higher degree of elastic fibre fragmentation and loss and mucoid extracellular matrix accumulation was present in the samples from TAV patients when compared with that from BAV patients (P < 0.001). However, correlation was poor for all variables when considering aortic diameter and histological abnormalities or age and histological abnormalities in both BAV and TAV patients. CONCLUSIONS: Our study demonstrates a greater incidence of severe histological abnormalities in TAV patients when compared with BAV patients.

Bradymyces oncorhynchi is a poorly known melanised fungal species that has been isolated only from a hyperaemic focus near the enlarged spleen of a rainbow trout. Although the pathogenicity of this species to fish is suspected, it has not been fully confirmed. Four laboratory experiments were conducted to test the effect of the fungus on the health of rainbow trout fingerlings. Mycelia were cultivated under different conditions to increase inoculum variability and to test the impact of the conditions on the pathogenicity. The inoculum was subsequently administered by the intraperitoneal route. The clinical manifestations, gross pathological lesions and histopathological changes were identical in all experimental groups, i.e. lethargy, inappetence, anorexia, weight reduction, pale gills, full-thickness ulceration of the abdominal wall, muscle atrophy, haemorrhagic or pale liver, fat-altered pyloric region with haemorrhages, enlarged spleen, haemorrhagic ascites, systemic granulomatous lesions with the presence of melanised hyphae (phaeohyphomycosis), multifocal granulomatous hepatitis, perivascular lymphocytic infiltration, vacuolation of hepatocytes and vacuolation of the kidney tubule epithelium. No statistically significant differences were found in the survival of the fish with respect to experimental settings. The survival period ranged between 3 and 135 days.
Deoxynivalenol (DON), produced by the Fusarium genus, is a major contaminant of cereal grains used in the production of fish feed. The effect of mycotoxin deoxynivalenol on rainbow trout (Oncorhynchus mykiss) was studied using a commercial feed with the addition of DON in a dose of 2 mg/kg feed. The fish (n=40) were exposed to the mycotoxin for 23 days. The trout were divided into two groups, control and experimental groups. Control groups were fed a commercial feed naturally contaminated with a low concentration of DON (225 μg/kg feed); experimental groups were fed a commercial feed with the addition of DON (1964 μg/kg feed). Plasma biochemical and haematological indices, biometric parameters, and histopathological changes were assessed at the end of the experiment. The experimental groups showed significantly lower values in MCH (P<0.05). In biochemical indices, after 23-day exposure, a significant decrease in glucose, cholesterol (P<0.05), and ammonia (P<0.01) was recorded in the experimental group compared to the control group. Our assessment showed no significant changes in biometric parameters. The histopathological examination revealed disorders in the caudal kidney of the exposed fish. The obtained data show the sensitivity of rainbow trout (O. mykiss) to deoxynivalenol.
