Development of a chromatographic method in bioanalysis is a challenging and complex procedure with many pitfalls and often unexpected reversals that can require several months to accomplish. Even an experienced analytical team must contend many limitations mainly in connection with the strict requirements imposed on current clinical research. These restrictions typically persist throughout the whole development process, from clinical trial assignment, across optimization of extraction of biological materials and chromatographic separation, to validation and data interpretation. This paper describes questions and their possible answers raised during the pre-analytical phase such as use of modern sample preparation techniques in clinical methods, application of internal standards, as well as selection of stationary phases and detection techniques in the analytical phase. Validation problems and interpretation of results are demonstrated with three typical examples of characteristics to be considered, i.e. recovery, matrix effect, and limit of detection vs. lower limit of quantification.

In our work, we introduced a novel concept of the lab-in-a-syringe tests. We solved the problem of detection in already published LIS tests by putting all the reaction and detection pads directly into the syringe barrel. We also used more layers to make the results visible for users. Two detection layouts: (i) with using rounded pads-based detection, and (ii) with using rectangular detection pads, were studied. As the proof of concept, we studied the determination of Ni(II) using dimethylglyoxime as the reagent and blocking of the interference of Fe(II). The calibrations for Ni(II) at the optimal conditions has excellent R2 of 0.998 with production costs of 0.2 USD per one test.

Previously reported DNA aptamers developed against surface proteins extracted from Campylobacter jejuni were further characterized by aptamer-based Western blotting and shown to bind epitopes on proteins weighing ~16 and 60 kD from reduced C. jejuni and Campylobacter coli lysates. Proteins of these approximate weights have also been identified in traditional antibody-based Western blots of Campylobacter spp. Specificity of the capture and reporter aptamers from the previous report was further validated by aptamer-based ELISA-like (ELASA) colorimetric microplate assay. Finally, the limit of detection of the previously reported plastic-adherent aptamer-magnetic bead and aptamer-quantum dot sandwich assay (PASA) was validated by an independent food safety testing laboratory to lie between 5 and 10 C. jejuni cells per milliliter in phosphate buffered saline and repeatedly frozen and thawed chicken rinsate. Such ultrasensitive and rapid (30 min) aptamer-based assays could provide alternative or additional screening tools to enhance food safety testing for Campylobacter and other foodborne pathogens.

Fluorescence resonance energy transfer (FRET) in combination with quantum dots (QDs) and their superior properties has enabled designing of the new and improved sensors. In this review, the latest novelties in development and application of FRET nanosensors employing QDs are presented. QDs offer several advantages over organic dyes - broad excitation spectra, narrow defined tunable emission peak, longer fluorescence lifetime, resistance to photobleaching and 10-100 times higher molar extinction coefficient. These properties of QDs allow multicolor QDs to be excited from one source by common fluorescent dyes without emission signal overlap and results in brighter probes comparing to conventional fluorophores. Due to these benefits, QD-FRET-based nanosensors gained a wide spread popularity in a variety of scientific areas. These sensors are most frequently applied in the domain of the nucleic acid and enzyme activity detection. Other applications are detection of peptides and low-molecular compounds, environmental pollutants, viruses, microorganisms and their toxins, QD-FRET-based immunoassays, and pH sensors.

Při podezření na expozici ionizujícím zářením se u osob provádí vyšetření pomocí biodozimetrických metod. Změny v chromozomech umožňují detekovat míru expozice, předvídat stupeň poškození a rozhodnout o léčení. Kromě toho byla prokázána korelace mezi zvýšenou hladinou chromozomálních aberací po expozici genotoxickým faktorům a zvýšeným rizikem rakoviny. Biodozimetrických metod existuje celá řada, ale záleží na množství ozářených osob, laboratorních podmínkách, senzitivitě a časovém faktoru, který test je nejvhodnější. Mikrojaderný test je jedním z metod volby, protože výsledky mohou být známy do 48 až 72 hodin po odběru krve a je dostatečně citlivým ukazatelem dávky ionizujícího záření. Mikrojaderný test periferních lymfocytů je standardní biodozimetrická metoda, kterou lze použít jak v případě radiačních nehod, tak i monitoringu profesního ozáření osob. Byl vyvinut Fenechem a Morleym v roce 1985 a jeho principem je detekce malých acentrických chromozomálních fragmentů, které zůstanou mimo jádro během buněčného dělení. Tato mikrojádra jsou v cytoplazmě mimo hlavní dceřiná jádra. Mikrojádra mohou obsahovat jak celé chromozomy, tak i pouze jejich části.
Examinations based on biodosimetric methods are implemented in the case of suspect exposure of persons to ionizing radiation. Changes in chromosomes make it possible to detect the level of the exposure, predict the degree of the damage and make decisions on the treatment. In addition, a correlation was demonstrated between enhanced levels of chromosomal aberrations after the exposure to genotoxic factors and increased risk of cancer disease. There are a number of biodosimetric methods, but their choice depends on the number of persons exposed, laboratory conditions, sensitivity and time factor. The test of micronuclei is one of the methods of choice, since the results can be known as soon as within 48 to 72 hours after sampling the blood and the results are a sufficiently sensitive parameter indicating the ionizing radiation dose. The text of peripheral lymphocyte micronuclei is a standard biodosimetric method, which may be employed in the case of radiation accidents as well as in the monitoring of the occupational exposure of persons. It was developed by Fenech and Morley in 1985 and is based on the detection of small acentric chromosomal fragments, which remain beyond the nucleus in the course of the cell division. These micronuclei are present in the cytoplasm beyond the main daughter nuclei. The micronuclei can contain whole chromosomes as well as their parts only
