The ever-increasing number of Lyme borreliosis patients led us to consider more effective procedures for disease prevention. The aim of our study was to monitor the annual activity and infectivity of Ixodes ricinus ticks in the Pisárky region, City of Brno, CR, and to test the responses of the locally-captured ticks to selected repellents. The result of regular one-hour-per-week monitoring in 2011 was the collection of ticks that directly reflected the highest number of Lyme disease patients (4,835) detected throughout the period of recording in the Czech Republic. The ticks were examined for spirochaetes by dark field microscopy. The positive samples were identified by PCR analysis, confirming that 76% of these were infected with Borrelia burgdorferi sensu lato. Ticks were most abundant in May and June, with August having the highest risk for spirochaetal infection. Tick activity was statistically correlated with temperature. The moving-object-bioassay was used to study repellent efficiency on the Ixodes ricinus nymphs captured in the above-mentioned suburban park. Five selected commercial repellents based on DEET (N, N-diethyl-3-thylbenzamide) showed statistically different effects on the non-repellent control group.

Cíl práce: Cílem této publikace je prezentace výsledků monitoringu prevalence spirochét Borrelia burgdorferi s. l. v klíštěti Ixodes ricinus ve dvou různých lokalitách v Brně. Současně jsou sledovány faktory, které by mohly ovlivnit aktivitu klíšťat v těchto lokalitách. Záměrem této studie je předložit aktuální stav monitoringu prováděného první tři jarní měsíce letošního roku (2011) a porovnat tento stav s minulým rokem 2010. Materiál a metody: V letech 2010–2011 byla v pravidelných intervalech sbírána klíšťata ve dvou lokalitách v Brně (Pisárky a Zamilovaný hájek) metodou vlajkování. Klíšťata byla vyšetřována metodou zástinové mikroskopie (DFM) na přítomnost patogenních spirochét. Pozitivní vzorky na DFM z roku 2010 byly vyšetřeny metodou PCR pro identifikaci Borrelia burgdorferi sensu lato (Bbsl). Výsledky: V prvních třech měsících (březen–květen) 2011 bylo odchyceno 234 klíšťat v lokalitě Pisárky a 211 klíšťat v lokalitě Zamilovaný hájek (ZH). Pozitivita spirochét zjištěná metodou DFM činila 5,56 % v Pisárkách a 25,12 % v ZH. V 75 % bylo konfirmováno metodou PCR, že se jedná o Bbsl. Statistická analýza potvrdila závislost aktivity klíšťat na teplotě ovzduší na obou lokalitách. Závěr: Obě lokality vykazují promořenost na patogenní Bbsl. Ta je však v rámci jednoho města na různých lokalitách rozdílná. Zvláště lokalita ZH vykazuje vysokou hodnotu promořenosti (25,12 %), která by mohla být způsobena ohniskovým šířením nákazy z několika zdrojů: klíšťat i rezervoárů, např. hlodavců. Odrazem této vysoké promořenosti by mohla být nepříznivá epidemiologická situace v Brně (ústní konzultace s MUDr. R. Vaverkovou – KHS). Na druhé straně lokalita Pisárky se promořeností neodchyluje od hodnoty zjištěné již dříve v několikaleté studii.
Objective: The aim of the article is to present the results of monitoring the prevalence of the spirochete Borrelia burgdorferi sensu lato (Bbsl) in Ixodes ricinus ticks in two different locations in the city of Brno, Pisárky and Zamilovaný hájek (ZH). At the same time, factors potentially affecting the activity of ticks in these locations were studied. Results of the monitoring carried out in the first three spring months of the year 2011 are compared with 2010 results. Material and Methods: In 2010–2011, Ixodes ricinus ticks were periodically sampled in two locations in Brno by flagging. Ticks were investigated by dark field microscopy (DFM) for the presence of spirochetes. DFM-positive samples from 2010 were tested by PCR to identify Bbsl. Results: In the first three spring months (March–May) of 2011, a total of 234 ticks were collected in Pisárky and 211 ticks in ZH. Positive samples detected by DFM accounted for 5.56 % in Pisárky and 25.12 % in ZH. PCR confirmed Bbsl in at least 75 %. Statistical analysis confirmed the dependence of tick activity on the air temperature in both locations. Conclusion: Within the same city, seroprevalence was found to be different in two locations. High seroprevalence in ZH (25.12 %) could be attributed to focal spread of the disease from several sources, both ticks and reservoirs such as rodents. This may results in unfavourable epidemiological situation in Brno. On the other hand, the seroprevalence rates in Pisárky are consistent with findings from an earlier long-term study.
