Metabolic syndrome (MetS) belongs to the serious health complications expanding in cardiovascular diseases, obesity, insulin resistance, and hyperglycemia. In this study, hypertriacylglycerolemic rats fed a high-fat-fructose diet (HFFD) were used as an experimental model of MetS to explore the effect of tested compounds. Effects of a new prospective pyridoindole derivative coded SMe1EC2 and the natural polyphenol rutin were tested. Endothelial nitric oxide synthase (NOS3) and nuclear factor kappa B (NF-?B) expression were assessed in the left ventricle immunohistochemically and left ventricle activity was monitored in isolated perfused rat hearts. NOS3 activity in the left ventricle decreased markedly as a result of a HFFD. NOS3 expression was upregulated by both substances. NF-?B expression was increased in the MetS group in comparison to control rats and the expression further increased in the SMe1EC2 treatment. This compound significantly improved the coronary flow in comparison to the control group during reperfusion of the heart followed after ischemia. Further, it tended to increase left ventricular systolic pressure, heart product, rate of maximal contraction and relaxation, and coronary flow during baseline assessment. Moreover, the compound SMe1EC2 decreased the sensitivity of hearts to electrically induced ventricular fibrillation. Contrary to this rutin decreased coronary flow in reperfusion. Present results suggest that despite upregulation of NOS3 by both substances tested, pyridoindole SMe1EC2 rather than rutin could be suitable in treatment strategies of cardiovascular disorders in MetS-like conditions.

BACKGROUND: Quercetin-3-O-β-D-glucopyranoside (isoquercitrin) and quercetin-3-O-rutinoside (rutin) are common components of a normal human diet and are increasingly used in food supplements. Here their effect on mutagenesis and immunity is shown. RESULTS: The in vitro (anti)mutagenic potential was compared with that of quercetin using the Ames test in Salmonella typhimurium His(-) strains TA100, TA98 and TA102. Isoquercitrin only slightly increased the number of revertants, while rutin was totally non-mutagenic. On the other hand, all compounds displayed dose-dependent protective activity against H2O2 - and tert-butyl hydroperoxide-induced oxidative damage to the TA102 strain and at 75 µmol L(-1) inhibited H2O2/Fe(2+)-induced formation of the open circular and linear forms of the DNA plasmid pBSIISK(-). In mice, none of the flavonols (0.86 µmol day(-1), 34 days) induced harmful effects. In immunized animals, all compounds enhanced ex vivo B cell proliferation; quercetin stimulated lymphocyte basal proliferation and increased the number of IgM-producing lymphocytes. Rutin promoted NK cytotoxic activity, supported T cells and enhanced gut epithelium renewal. No effect on IgG-forming cells was found. CONCLUSION: Isoquercitrin displayed negligible and rutin no mutagenicity, but both showed significant antimutagenic and DNA-protective effects against oxidative damage. In vivo, they supported the readiness of the immune system for specific humoral immune response.

Článek shrnuje možnosti využití léků pro systémovou enzymoterapii při řešení nejrůznějších poruch pohybového aparátu. Hlavními účinnými látkami těchto léků jsou proteolytické enzymy živočišného a rostlinného původu s rutinem. Nejznámější jsou jejich účinky protiotokové a protizánětlivé, které se velmi dobře uplatňují u stavů po úrazech a operacích. Lze je ale využít i při konzervativní léčbě degenerativních chorob pohybového aparátu a nejrůznějších bolestivých syndromů zahrnovaných pod pojem revmatismus měkkých tkání. Zde se mimo jiné velmi dobře uplatňuje také jejich analgetický efekt. Jsou prezentovány hlavně novější studie a práce českých autorů. Upozorňujeme také na některé experimentální práce, které přispívají k objasnění podstaty účinků jednotlivých proteáz i jejich směsí obsažených v lécích pro systémovou enzymoterapii.
The paper summarizes possibilities of the use of systemic enzyme therapy for locomotor apparatus disorders. The main active components of these medicaments are proteolytic enzymes of animal and plant origin with rutin. Their antiedematous and antiinflammatory effects are well known and are usable in the treatment of conditions after an injury and a surgery. Medicaments for systemic enzyme therapy are as well suitable for conservative treatment of degenerative disorders of locomotor apparatus and various painful syndromes classified as soft tissue rheumatism. Analgetic effect of systemic enzyme therapy is very useful in these disorders. The newer clinical trials and experience of Czech author are mainly presented in this paper.

Preparation of coated pellets intended for rutin colon delivery, their evaluation in vitro and in vivo in experimental colitis in rats was the purpose of this study. Pellets were obtained using extrusion/spheronization and coated with three types of coatings (caffeic acid/hypromellose/alginic acid; sodium alginate/hypromellose/zinc acetate; sodium alginate/chitosan). Dissolution using buffers of pH values, β-glucosidase and times corresponding to gastrointestinal tract (GIT) was provided. Pellets coated with alginate/chitosan showed low rutin dissolution (12-14%) in upper GIT conditions and fast release (87-89%) under colon conditions; that is a good presumption of intended rutin release. After colitis induction and development, the rats were treated with pellets and rutin solution administered orally, solution also rectally. Colon/body weight ratio, myeloperoxidase activity and histological evaluation were performed. Rutin was able to promote colonic healing at the dose of 10mg/kg: colon/body weight ratio decreased and myeloperoxidase activity was significantly suppressed. Pellets coated with alginate/chitosan applied orally and rutin solution administered rectally showed the best efficacy. The combination of rutin as natural product, mucoadhesive chitosan degraded in the colon and sodium alginate as the main coating substance in the form of pellets create a promising preparation for therapy of this severe illness.

The natural flavonoid quercetin is a low affinity ligand of the aryl hydrocarbon receptor (AhR), a transcription factor regulating the expression of cytochrome P450 (CYP) 1A enzymes. This study examined the ability of quercetin, isoquercitrin (quercetin-3-O-glucoside), rutin (quercetin-3-O-rutinoside) and taxifolin (dihydroquercetin) to activate AhR and to induce CYP1A1 expression in human hepatoma HepG2 cells. Gene reporter assays showed that quercetin significantly activated AhR and triggered CYP1A1 transcription after 24 h exposure. These effects were, however, much lower than those of 2,3,7,8-tetrachlorodibenzo-p-dioxin, a prototypical AhR ligand. Quercetin also induced a significant increase in CYP1A1 mRNA levels together with a moderate increase in the level of CYP1A1 activity. In contrast, isoquercitrin and rutin had negligible effects on AhR activity and CYP1A1 expression. Taxifolin at the highest concentration tested (50 µm) produced a mild non-significant increase in AhR activity and CYP1A1 transcription. Taxifolin also significantly increased CYP1A1 mRNA expression, but this effect was approximately 15 times weaker than that of quercetin and was not accompanied by induction of CYP1A1 activity. It is concluded that quercetin, but not its 3-O-glycosides isoquercitrin and rutin, induces AhR activation and CYP1A1 expression in HepG2 cells and that the CYP1A1-inducing activity of taxifolin has a low toxicological potential.

Podávání venoaktivních látek může být použito u symptomatických pacientů s chronickými žilními onemocněními. Nejvyšší úroveň evidence účinnosti venofarmak existuje pro diosmin-hesperidin, hydroxyetylrutosidy a kalcium-dobesilát. Aplikace diosminuhesperidinu a pentoxifylinu může v kombinaci s lokální a kompresní léčbou zlepšit průběh hojení bércových vředů.
Submission of venoactive substances may be used in symptomatic patients with chronic venous diseases. The highest level of effectiveness evidence exists for the following venopharmacs: diosmin-hesperidin, hydroxyetylrutosides and calcium dobesilate. Administration of diosmin-hesperidin and pentoxifylline in combination with local and compressive treatment may improve healing of varicose ulcers.

Cílem přehledného článku bylo dosáhnout hlubšího pochopení ve vědeckých časopisech publikovaných účinků flavonolů, zejména rutinu, kvercitrinu a jejich aglykonu kvercetinu, s důrazem na jejich potenciální příznivé působení u nespecifických střevních zánětů. Výzkum v oblasti flavonoidů v několika posledních létech významně vzrostl a objevila se řada nových skutečností týkajících se jejich absorpce, metabolismu a možných mechanismů účinků. Článek uvádí nedávno publikované výsledky získané v pokusech in vitro a experimentech in vivo na zvířecích modelech. Další výzkum zaměřený na klinické účinky rutinu může být důležitý pro vývoj nové léčivé látky k terapii nespecifických střevních zánětů v humánní medicíně.
The aim of this review article was to obtain a deeper understanding of the reported effects of flavonols, in particular rutin, quercitrin and their aglycone quercetin, with respect to their potential beneficial action on inflammatory bowel disease. Research in the field of flavonoids has increased significantly in recent few years and many new investigations have been performed concerning their absorption, metabolism, and probable mechanisms of action. Recently published results obtained in in vitro approaches and in vivo experiments on animal models are reported. Further investigation aimed at the clinical effects of rutin may be important for the development of a new therapeutic agent for the treatment of inflammatory bowel disease in humans.
High levels of catecholamines are cardiotoxic and may trigger acute myocardial infarction (AMI). Similarly, the synthetic catecholamine isoprenaline (ISO) evokes a pathological state similar to AMI. During AMI there is a marked increase of free iron and copper which are crucial catalysts of reactive oxygen species formation. Rutin, a natural flavonoid glycoside possessing free radical scavenging and iron/copper chelating activity, may therefore be potentially useful in reduction of catecholamine cardiotoxicity as was previously demonstrated after its long-term peroral administration. Male Wistar:Han rats received rutin (46 or 11.5 mg kg(-1) i.v.) alone or with necrogenic dose of ISO (100 mg kg(-1) s.c.). Haemodynamic parameters were measured 24h after drug application together with analysis of blood, myocardial content of elements and histological examination. Results were confirmed by cytotoxicity studies using cardiomyoblast cell line H9c2. Rutin in a dose of 46 mg kg(-1) aggravated ISO-cardiotoxicity while the dose of 11 mg kg(-1) had no effect. These unexpected results were in agreement with in vitro experiments, where co-incubation with larger concentrations of rutin significantly augmented ISO cytotoxicity. Our results, in contrast to previous studies in the literature, suggest that the reported positive effects of peroral administration of rutin were unlikely to have been mediated by rutin per se but probably by its metabolite(s) or by some other, at this moment, unknown adaptive mechanism(s), which merit further investigation.
