Biological effects of manumycin-related compounds will be tested using a tissue culture models with human monocyte/macrophages (THP-1) and epithelial cell lines. For a screening of potential effect of tested compounds on pro-inflammatory cytokines and chemokines production, multiplex qRT-PCR will be used followed by measurement of proinflammatory cytokine concentrations, immunophenotypying and functional tests. Genetic screening will be used for identification of streptomycete strains producing manumycine-related compounds with consequent isolation and characterization of new compounds, construction of cosmid libraries, and genetic manipulations with the upper polyketide chain of manumycins (length, branching, saturation) including gene shuffling between producers.
Biologické účinky látek manumycinového typu budou testovány pomocí modelu tkáňových kultivací s lidskými liniemi monocytů/makrofágů (THP-1) a epiteliálních linií. Pro screening potenciálního efektu testovaných látek na tvorbu prozánětlivých cytokinů a chemokinů bude využita multiplexová kvantitativní RT-PCR s následným měřením koncentrací cytokinů, imunofenotypizací a funkčními testy. Kmeny streptomycet produkující manumycinové látky budou identifikovány pomocí genového screeningu, bude provedena izolace a charakterizace nových látek, konstrukce kosmidových knihoven a genetická manipulace horního polyketidového řetězce manumycinů (délka, štěpení, saturace) včetně genového "shufflingu" mezi producenty.
