Drying river networks include non-perennial reaches that cease to flow or dry, and drying is becoming more prevalent with ongoing climate change. Biodiversity responses to drying have been explored mostly at local scales in a few regions, such as Europe and North America, limiting our ability to predict future global scenarios of freshwater biodiversity. Locally, drying acts as a strong environmental filter that selects for species with adaptations promoting resistance or resilience to desiccation, thus reducing aquatic α-diversity. At the river network scale, drying generates complex mosaics of dry and wet habitats, shaping metacommunities driven by both environmental and dispersal processes. By repeatedly resetting community succession, drying can enhance β-diversity in space and time. To investigate the transferability of these concepts across continents, we compiled and analyzed a unique dataset of 43 aquatic invertebrate metacommunities from drying river networks in Europe and South America. In Europe, α-diversity was consistently lower in non-perennial than perennial reaches, whereas this pattern was not evident in South America. Concomitantly, β-diversity was higher in non-perennial reaches than in perennial ones in Europe but not in South America. In general, β-diversity was predominantly driven by turnover rather than nestedness. Dispersal was the main driver of metacommunity dynamics, challenging prevailing views in river science that environmental filtering is the primary process shaping aquatic metacommunities. Lastly, α-diversity decreased as drying duration increased, but this was not consistent across Europe. Overall, drying had continent-specific effects, suggesting limited transferability of knowledge accumulated from North America and Europe to other biogeographic regions. As climate change intensifies, river drying is increasing, and our results underscore the importance of studying its effects across different regions. The importance of dispersal also suggests that management efforts should seek to enhance connectivity between reaches to effectively monitor, restore and conserve freshwater biodiversity.

The aim of this cross-sectional study was to review the incidence of the dry eye syndrome in rheumatoid arthritis (RA) patients, evaluate the association among the incidence of the dry eye syndrome, presence of positive rheumatoid factor (RF), the RA stage, and the duration of the disease. The group consisted of altogether 100 patients, 16 men and 84 women; the average age was 58.9 years (SD 14.6). The average duration of RA was 12.3 years, SD 11.0. In each patient, the Schirmer test I was performed, the presence of the LIPCOF (Lid Parallel Conjunctival Folds) on the slit lamp was assessed, the BUT (Tear Break-Up Time) was measured and vital fluorescein staining was performed. In each patient the data of the presence or absence of the RF in the serum, RA severity according to the X-ray examination, and the disease duration were recorded. The Pearson's association test for nominal variables was used for statistical evaluation of the association between the rheumatoid arthritis presence and the dry eye syndrome. In our group of 100 patients, the Schirmer test I was positive in 67% of patients. Positive BUT was marked in 84 % of patients. The conjunctival folds were present in 45 % of patients only. The pathological findings after cornea fluorescein staining appeared in 18 % of patients. The dry eye syndrome incidence was marked in 74% of patients with RA. Subjective difficulties were declared by 38.3% of patients only. The local treatment was already established in 23.0% of patients only. We did not find statistically significant correlation between the RF positive rheumatoid arthritis appearance and dry eye syndrome, nor between the stage of the rheumatoid arthritis and presence of the dry eye syndrome. We proved statistical connection between the presence of dry eye syndrome and the duration of rheumatoid arthritis longer than 10 years. Keratoconjunctivitis sicca is the most common ocular complication in rheumatoid arthritis patients. We proved the connection between the dry eye syndrome presence and duration of the RA longer than 10 years; we did not find the dependence among the RF presence and stage of the rheumatoid arthritis and the appearance of the dry eye syndrome. The early diagnosis of the dry eye syndrome and the effective local therapy may prevent very serious corneal complications, which are difficult to treat.

The reconstruction of the settlement´s hinterland and acquisition of plant resources is one of the crucial questions in the field of environmental archaeology. Our study is focused on the reconstruction of the settlement's structure and character of the environment from which the site drew resources. These research questions were addressed by the interpretation of plant macroremains, charcoals, and the results of the spatial model. We have focused on the maximum size of the settlement that the surrounding countryside was able to withstand. Our results clearly demonstrated significant deforestation and intensive land use in the vicinity of the Late Bronze Age study site. As the weed taxa showed, a wide range of crops was grown in rather dry or less often in damp fields. Based on our archaeobotanical results, we were able to reconstruct several types of grasslands: dry pastures and fallow fields on plateaus and slopes, wet pastures or meadows in the floodplain. Acidophilous oak forests, alluvial forests, and shrubs were reconstructed as the most common forest habitats in the vicinity of the study site. Based on the archaeological knowledge of the region, we assume relatively low population density during the Late Bronze Age, and thus only a small part of the more or less forested landscape was significantly affected by human activities.

BACKGROUND: There is a continuing interest in finding effective methods for scar treatment. Dry needling is gaining popularity in physiotherapy and is defined by Western medicine as a type of acupuncture. The terms acupuncture and dry needling have been used interchangeably so we have focused on the efficacy of dry needling or acupuncture in scar treatment. OBJECTIVE: The aim of this systematic review was to determine the usefulness of dry needling or local acupuncture for scar treatment. In our search process, we used the terms 'acupuncture,' 'needling,' or 'dry needling' to identify all relevant scientific papers. We have focused on the practical aspects of local management of different scar types with dry needling or acupuncture. SEARCH STRATEGY: The search strategy included different combinations of the following keywords: 'scar', 'keloid', 'dry needling', 'needling', 'acupuncture', 'treatment', 'physical therapy'. This systematic review was conducted in accordance with PRISMA guidelines. MEDLINE (PubMed, EBSCOHost and Ovid), EMBASE (Elsevier), and Web of Science databases were searched for relevant publications from inception through October 2023. INCLUSION CRITERIA: The studies that investigated the effectiveness of dry needling or acupuncture for scar treatment were included. DATA EXTRACTION AND ANALYSIS: The main extraction data items were: the needling technique; needle: diameter, length; needling locations; manual needling manipulation; number of sessions; settings; outcomes and results. RESULTS: As a result of a comprehensive search, 11 manuscripts were included in the systematic review, of which eight were case reports, two were randomized trials and one study concerned case series. Two case reports scored 2-4 out of 8 points on the JBI checklist, five studies scored 5-7, and one study scored 8 points. The methodological quality of the two clinical trials was rated as good or fair on the PEDro scale. The case series study scored 7 of 10 points on the JBI checklist. A meta-analysis was not possible as only two randomized trials, eight case reports, and one case series were eligible for review; also, scar assessment scales and pain severity scales were highly heterogeneous. CONCLUSIONS: The studies differed regarding the delivery of dry needling or local acupuncture for scar treatment. Differences included treatment frequency, duration, number of treatments, selection of needle insertion sites, number of needles used, angle of needle placement, and use of manual needling manipulation. BACKGROUND: Females can significantly improve their fitness by utilizing a range of maternal effects. Embryos of annual killifish survive the dry season in ephemeral pools encased in dry substrate for several months. Here, we experimentally test the association between energetic provisioning and maternally controlled duration of embryonic development in the African annual killifish Nothobranchius furzeri (Cyprinodontiformes). RESULTS: We found that embryonic energetic reserves do not limit duration of development. However, differences in energetic reserves affect the size at which embryos hatched, with larger yolk size resulting in larger hatchling size. CONCLUSIONS: These findings suggest uncoupling of the two traits examined (i.e., embryonic energetic reserves and development duration) and emphasize the strong buffering role of diapause in the energetic balance of embryonic development in the annual killifish. Hydrogen cyanide (HCN), a key molecule of significant importance in contemporary perspectives on prebiotic chemistry, originates in planetary atmospheres from various processes, such as photochemistry, thermochemistry, and impact chemistry, as well as from delivery by impacts. The resilience of HCN during periods of heavy bombardment, a phenomenon caused by an influx of material on unstable trajectories after accretion, remains relatively understudied. This study extensively investigates the stability of HCN under impact conditions simulated using a laboratory Nd:YAG laser in the ELISE experimental setup. High-resolution infrared spectroscopy was employed to monitor the gas phase composition during these simulations. Impact chemistry was simulated in bulk nitrogen atmospheres with varying mixing ratios of HCN and water vapor. The probed range of compositions spans from ∼0 to 1.8% of HCN and 0 to 2.7% of H2O in a ∼1 bar nitrogen atmosphere. The primary decomposition products of HCN are CO and CO2 in the presence of water and unidentified solid phase products in dry conditions. Our experiments revealed a range of initial HCN decomposition rates between 2.43 × 1015 and 5.17 × 1017 molec J-1 of input energy depending on the initial composition. Notably, it is shown that the decomposition process induced by the laser spark simulating the impact plasma is nonlinear, with the duration of the irradiation markedly affecting the decomposition rate. These findings underscore the necessity for careful consideration and allowance for margins when applying these rates to chemical models of molecular synthesis and decomposition in planetary atmospheres.

The widespread use of multipotent mesenchymal stromal cell-derived secretome (MSC-sec) requires optimal preservation methods. Lyophilization offers benefits like concentrating the secretome, reducing the storage volume, and making storage conditions more flexible. This study evaluated the influence of storage duration and temperature on lyophilized MSC-sec. The conditioned medium from Wharton's jelly MSCs was stored at - 80 °C or lyophilized with or without trehalose. Lyophilized formulations were kept at - 80 °C, - 20 °C, 4 °C, or room temperature (RT) for 3 and 30 months. After storage and reconstitution, the levels of growth factors and cytokines were assessed using multiplex assay. The storage of lyophilized MSC-sec at - 80 °C ensured biomolecule preservation for 3 and 30 months. Following 3 month storage at 4 °C and RT, a notable decrease occurred in BDNF, bNGF, and sVCAM-1 levels. Prolonged 30 month storage at the same temperatures significantly reduced BDNF, bNGF, VEGF-A, IL-6, and sVCAM-1, while storage at - 20 °C decreased BDNF, bNGF, and VEGF- A levels. Trehalose supplementation of MSC-sec improved the outcome during storage at 4 °C and RT. Proper storage conditions were crucial for the preservation of lyophilized MSC-sec composition. Short-term storage at various temperatures maintained over 60% of the studied growth factors and cytokines; long-term preservation was only adequate at -80 °C.

In this paper, we present the ImmunoDisk, a fully automated sample-to-answer centrifugal microfluidic cartridge, integrating a heterogeneous, wash-free, magnetic- and fluorescent bead-based immunoassay (bound-free phase detection immunoassay/BFPD-IA). The BFPD-IA allows the implementation of a simple fluidic structure, where the assay incubation, bead separation and detection are performed in the same chamber. The system was characterized using a C-reactive protein (CRP) competitive immunoassay. A parametric investigation on air drying of protein-coupled beads for pre-storage at room temperature is presented. The key parameters were buffer composition, drying temperature and duration. A protocol for drying two different types of protein-coupled beads with the same temperature and duration using different drying buffers is presented. The sample-to-answer workflow was demonstrated measuring CRP in 5 µL of human serum, without prior dilution, utilizing only one incubation step, in 20 min turnaround time, in the clinically relevant concentration range of 15-115 mg/L. A reproducibility assessment over three disk batches revealed an average signal coefficient of variation (CV) of 5.8 ± 1.3%. A CRP certified reference material was used for method verification with a concentration CV of 8.6%. Our results encourage future testing of the CRP-ImmunoDisk in clinical studies and its point-of-care implementation in many diagnostic applications.

A digestate with amendments provides plants with available nutrients and improves the microbiological properties of treated soil. Modification of a digestate through the addition of a biochar and sulphur source is less well-known. This pot experiment aimed at comparing the short- and long-time fertilization effects of a digestate enriched with biochar, with elemental sulphur, or with a combination of both on soil health and plant biomass. The experiment was carried out with maize, cultivated twice (1st-12th week = pre-cultivation; re-sowing after shoot harvest, 13th-24th = main cultivation) in soil amended with prepared digestate. The digestate used in pre-cultivation was incubated untreated (D) and was then treated with biochar (D + B), with elemental sulphur at a low (LS) and high (HS) dose, or with a combination of both (D + B + LS and D + B + HS). An additional unamended digestate (D) was added to each soil variant before the main cultivation. The application of digestate with a high dose of elemental sulphur and biochar mediated the most significant differences in the soil. The increase (compared to the unamended soil) was of short-term type (+11% and +6% increased total nitrogen and carbon after 12 weeks), then of long-term type (+54% and +30% increased sulphur and arylsulfatase activity after 24 weeks), and later emerged in the 13th to the 24th week of the experiment (+57% and +32% non-inhibited urease, increased N-acetyl-β-D-glucosaminidase and phosphatase). No significant differences in the effect of the applied amendments on dry aboveground plant biomass were observed.

Climate change and human pressures are changing the global distribution and the extent of intermittent rivers and ephemeral streams (IRES), which comprise half of the global river network area. IRES are characterized by periods of flow cessation, during which channel substrates accumulate and undergo physico-chemical changes (preconditioning), and periods of flow resumption, when these substrates are rewetted and release pulses of dissolved nutrients and organic matter (OM). However, there are no estimates of the amounts and quality of leached substances, nor is there information on the underlying environmental constraints operating at the global scale. We experimentally simulated, under standard laboratory conditions, rewetting of leaves, riverbed sediments, and epilithic biofilms collected during the dry phase across 205 IRES from five major climate zones. We determined the amounts and qualitative characteristics of the leached nutrients and OM, and estimated their areal fluxes from riverbeds. In addition, we evaluated the variance in leachate characteristics in relation to selected environmental variables and substrate characteristics. We found that sediments, due to their large quantities within riverbeds, contribute most to the overall flux of dissolved substances during rewetting events (56%-98%), and that flux rates distinctly differ among climate zones. Dissolved organic carbon, phenolics, and nitrate contributed most to the areal fluxes. The largest amounts of leached substances were found in the continental climate zone, coinciding with the lowest potential bioavailability of the leached OM. The opposite pattern was found in the arid zone. Environmental variables expected to be modified under climate change (i.e. potential evapotranspiration, aridity, dry period duration, land use) were correlated with the amount of leached substances, with the strongest relationship found for sediments. These results show that the role of IRES should be accounted for in global biogeochemical cycles, especially because prevalence of IRES will increase due to increasing severity of drying events.
