Brucellosis is a zoonosis caused by Brucella, which poses a great threat to human health and animal husbandry. Pathogen surveillance is an important measure to prevent brucellosis, but the traditional method is time-consuming and not suitable for field applications. In this study, a recombinase polymerase amplification-SYBR Green I (RPAS) assay was developed for the rapid and visualized detection of Brucella in the field by targeting BCSP31 gene, a conserved marker. The method was highly specific without any cross-reactivity with other common bacteria and its detection limit was 2.14 × 104 CFU/mL or g of Brucella at 40 °C for 20 min. It obviates the need for costly instrumentation and exhibits robustness towards background interference in serum, meat, and milk samples. In summary, the RPAS assay is a rapid, visually intuitive, and user-friendly detection that is highly suitable for use in resource-limited settings. Its simplicity and ease of use enable swift on-site detection of Brucella, thereby facilitating timely implementation of preventive measures.

BACKGROUND: Acanthopanax senticosus (Rupr. et Maxim.) is commonly used in Traditional Chinese Medicine. Syringin is a major ingredient of phenolic glycoside in Acanthopanax senticosus. OBJECTIVE: This study was performed to investigate whether Syringin could protect high glucose-induced bone marrow mesenchymal stem cells (BMSCs) injury, cell senescence, and osteoporosis by inhibiting JAK2/STAT3 signaling. METHODS: BMSCs isolated from both the tibia and femur of mice were induced for osteogenesis. The cell senescence was induced using the high glucose medium. The cells were treated with 10 and 100 μmol/l Syringin. Immunohistochemistry staining was performed to determine the β-galactosidase (SA-β-gal) levels in differentially treated BMSCs. MTT assay and flow cytometry analysis were also performed to assess cell viability and cell cycle. The level of ROS in cells with different treatment was measured by using flow cytometry with DCF-DA staining. Calcium deposition and mineralized matrices were detected with alizarin red and ALP staining, respectively. Osteogenesis related genes OCN, ALP, Runx2, and BMP-2 were detected by RT-PCR. Levels of senescence-related proteins including p53 and p21, as well as JAK2, p-JAK2, STAT3, and p-STAT3 were detected by Western blot analysis. RESULTS: Syringin treatment reversed the phenotypes of senescence caused by high glucose in BMSCs, including the arrest of G0/G1 cell cycle, enhanced SA-β-gal activity, and impaired cell growth. Syringin also decreased the elevated ROS production and the levels of p53, p21, and JAK2/STAT3 signaling activation. In addition, Syringin also enhanced the osteogenic potential determined by ARS and ALP staining, as well as increasing OCN, ALP, Runx2, and BMP-2 expressions. CONCLUSION: Syringin protects high glucose-induced BMSC injury, cell senescence, and osteoporosis by inhibiting JAK2/STAT3 signaling.

Pyroptosis is a form of cell death associated with inflammation. In the maintenance of airway homeostasis, pyroptosis goes through activation and assembly of Inflammasome. The pyroptosis pathway is mediated by caspase which activates the pore-forming effect of substrate gasdermin family members. It eventually leads to lysis and release of the cell contents and then induces an inflammatory response. In this process, it participates in airway homeostasis regulation by affecting airway immunity, airway epithelial structure and airway microbiota. Therefore, we discussed the correlation between airway immunity, airway epithelial structure, airway microbiota and the mechanism of pyroptosis to describe the role of pyroptosis in airway homeostasis regulation which is of great significance for understanding the occurrence and treatment of airway inflammatory diseases.

The purpose of the present study was to examine the influence of selected factors (possession duration, game quarter, defensive pressure, shooting distance from the basket, and heart rate level) on shooting efficiency on Under-19 (U19) and senior level of women's basketball competition (second division). The analysis procedures included five U19 and six senior-level games, containing 224 and 252 shooting attempts, respectively. Binary logistic regression identified the opponent's defensive pressure and shooting distance from the basket as significant predictors of shooting efficiency in both categories. When defensive pressure was high, the chance for the missed shot was 3.5 (95% CI; 1.43-8.52) and 3.19 (95% CI; 1.4-7.26) times more likely than it was under the minimum defensive pressure for U19 and senior category, respectively. Shooting efficiency significantly decreased when the horizontal distance from the basket increased. In U19, a chance for a missed shot was 4.63 (95% CI; 2-10.712) and 5.15 (95% CI; 1.91-13.86) times higher for medium and long-distance (respectively), compared to short-range shooting. In the senior category, the chance for the missed shot was 3.9 (95% CI; 1.83-8.31) and 3.27 (95% CI; 1.43-7.52) times higher for medium and long-distance (respectively) when compared to a short distance. The possession duration, game quarter, and heart rate level were identified as insignificant predictors. Therefore, the aforementioned findings suggest that basketball players and coaches may benefit from designing training sessions where the defender puts pressure on the shooting player as in a real game situation and balanced the frequency of shooting from different distances from the basket.

There are limited studies examining the impacts of perfectionism and achievement motivation on collegiate athletes' extra training and academic achievement in a Chinese context. This study aimed to examine the association of perfectionism (five facets) with extra training and academic performance among Chinese collegiate athletes and identify the mediating role of achievement motivation (two attributes) in the relationship between perfectionism and extra training and academic performance. With a prospective study design, 243 eligible participants completed two-wave surveys from September to December 2021. Measures included demographics, perfectionism (concern over mistake, CM; doubts about action, DA; personal standard, PS; organization; parental expectation, PE), achievement motivation (motive for success, MS; motive for avoiding failure, MF), extra-training (minutes/week), and academic performance (GPA). Results showed that CM, DA, PS, and MS were associated with extra training among Chinese collegiate athletes, while the associations of DA and PS with extra training were mediated by MS. In addition, DA, PS, organization, and MS were associated with participants' GPA, while MS was a salient mediator for the contributions of DA and PS on participants GPA. Research findings give new insights to the psychological mechanisms of perfectionism and achievement motivation on collegiate athletes' extra training and academic performance, contributing to future studies in relevant domains.

Rye is a valuable food and forage crop, an important genetic resource for wheat and triticale improvement and an indispensable material for efficient comparative genomic studies in grasses. Here, we sequenced the genome of Weining rye, an elite Chinese rye variety. The assembled contigs (7.74 Gb) accounted for 98.47% of the estimated genome size (7.86 Gb), with 93.67% of the contigs (7.25 Gb) assigned to seven chromosomes. Repetitive elements constituted 90.31% of the assembled genome. Compared to previously sequenced Triticeae genomes, Daniela, Sumaya and Sumana retrotransposons showed strong expansion in rye. Further analyses of the Weining assembly shed new light on genome-wide gene duplications and their impact on starch biosynthesis genes, physical organization of complex prolamin loci, gene expression features underlying early heading trait and putative domestication-associated chromosomal regions and loci in rye. This genome sequence promises to accelerate genomic and breeding studies in rye and related cereal crops.

Basketball shooting is one of the most important offensive skills in basketball. Winning or losing a game mostly depends on the shooting effectiveness. The study aims to compare the selected kinematic variables of 2-point (2-pt) and 3-point (3-pt) jump shots (after making a cut and receiving the ball) and ascertain the differences between elite male under 16 and 18 (U16M, U18M) and female under 16 and 18 (U16F, U18F) basketball players. Overall, forty-eight young male and female basketball players participated in the study. 3D motion analysis using an inertial suit with the addition of utilizing a smart ball was performed for assessing the 2-pt and 3-pt shooting techniques. Players in male categories shot for 2-pt with a higher center of mass difference in the vertical direction (U16M 5.7 cm, U18M 3.9 cm vs. U16F 1.4 cm, U18F 0.6 cm), with higher release shoulder angle (U16M 110.9, U18M 113.8 vs. U16F 103, U18F 105), and with a higher entry angle of the ball (U16M 34, U18M 32 vs. U16F 30, U18F 30) when compared to female categories (p < 0.001). In the 3-pt shooting, there were differences between male and female categories in the shoulder angle when releasing the ball (p < 0.001). In the players shooting speed, there were differences between U16M vs. U18F (0.95 ± 0.1 vs. 0.88 ± 0.1; p = 0.03) and U16F vs. U18F (0.96 ± 0.06 vs. 0.88 ± 0.1; p = 0.02) players. Male categories shot 3-pt shots with a smaller center of mass difference in the horizontal direction when compared to 2-pt shots (p < 0.001). The entry angle was higher in successful shooting attempts compared to unsuccessful shooting attempts when shooting for 3-pt (p = 0.02). Player shooting speed was higher in all categories (except U18F) when shooting for 3-pt (p < 0.001). It appears that performers show difference in kinematic variables based on distance from the basket. Basketball coaches and players should work to minimize the kinematic differences between 2-pt and 3-pt shooting and to optimize the shooting technique.

Seizure prediction is feasible, but greater accuracy is needed to make seizure prediction clinically viable across a large group of patients. Recent work crowdsourced state-of-the-art prediction algorithms in a worldwide competition, yielding improvements in seizure prediction performance for patients whose seizures were previously found hard to anticipate. The aim of the current analysis was to explore potential performance improvements using an ensemble of the top competition algorithms. The results suggest that minor increments in performance may be possible; however, the outcomes of statistical testing limit the confidence in these increments. Our results suggest that for the specific algorithms, evaluation framework, and data considered here, incremental improvements are achievable but there may be upper bounds on machine learning-based seizure prediction performance for some patients whose seizures are challenging to predict. Other more tailored approaches that, for example, take into account a deeper understanding of preictal mechanisms, patient-specific sleep-wake rhythms, or novel measurement approaches, may still offer further gains for these types of patients.

Rationale: Several common and rare genetic variants have been associated with idiopathic pulmonary fibrosis, a progressive fibrotic condition that is localized to the lung. Objectives: To develop an integrated understanding of the rare and common variants located in multiple loci that have been reported to contribute to the risk of disease. Methods: We performed deep targeted resequencing (3.69 Mb of DNA) in cases (n = 3,624) and control subjects (n = 4,442) across genes and regions previously associated with disease. We tested for associations between disease and 1) individual common variants via logistic regression and 2) groups of rare variants via sequence kernel association tests. Measurements and Main Results: Statistically significant common variant association signals occurred in all 10 of the regions chosen based on genome-wide association studies. The strongest risk variant is the MUC5B promoter variant rs35705950, with an odds ratio of 5.45 (95% confidence interval, 4.91-6.06) for one copy of the risk allele and 18.68 (95% confidence interval, 13.34-26.17) for two copies of the risk allele (P = 9.60 × 10-295). In addition to identifying for the first time that rare variation in FAM13A is associated with disease, we confirmed the role of rare variation in the TERT and RTEL1 gene regions in the risk of IPF, and found that the FAM13A and TERT regions have independent common and rare variant signals. Conclusions: A limited number of common and rare variants contribute to the risk of idiopathic pulmonary fibrosis in each of the resequencing regions, and these genetic variants focus on biological mechanisms of host defense and cell senescence.

Accurate seizure prediction will transform epilepsy management by offering warnings to patients or triggering interventions. However, state-of-the-art algorithm design relies on accessing adequate long-term data. Crowd-sourcing ecosystems leverage quality data to enable cost-effective, rapid development of predictive algorithms. A crowd-sourcing ecosystem for seizure prediction is presented involving an international competition, a follow-up held-out data evaluation, and an online platform, Epilepsyecosystem.org, for yielding further improvements in prediction performance. Crowd-sourced algorithms were obtained via the 'Melbourne-University AES-MathWorks-NIH Seizure Prediction Challenge' conducted at kaggle.com. Long-term continuous intracranial electroencephalography (iEEG) data (442 days of recordings and 211 lead seizures per patient) from prediction-resistant patients who had the lowest seizure prediction performances from the NeuroVista Seizure Advisory System clinical trial were analysed. Contestants (646 individuals in 478 teams) from around the world developed algorithms to distinguish between 10-min inter-seizure versus pre-seizure data clips. Over 10 000 algorithms were submitted. The top algorithms as determined by using the contest data were evaluated on a much larger held-out dataset. The data and top algorithms are available online for further investigation and development. The top performing contest entry scored 0.81 area under the classification curve. The performance reduced by only 6.7% on held-out data. Many other teams also showed high prediction reproducibility. Pseudo-prospective evaluation demonstrated that many algorithms, when used alone or weighted by circadian information, performed better than the benchmarks, including an average increase in sensitivity of 1.9 times the original clinical trial sensitivity for matched time in warning. These results indicate that clinically-relevant seizure prediction is possible in a wider range of patients than previously thought possible. Moreover, different algorithms performed best for different patients, supporting the use of patient-specific algorithms and long-term monitoring. The crowd-sourcing ecosystem for seizure prediction will enable further worldwide community study of the data to yield greater improvements in prediction performance by way of competition, collaboration and synergism.10.1093/brain/awy210_video1awy210media15817489051001.
