To explore the mechanism whereby cGAS-STING pathway regulates the pyroptosis of cryptorchidism cells, with a view to finding a new strategy for clinically treating cryptorchidism-induced infertility. Spermatogonial GC-1 cells were heat stimulated to simulate the heat hurt microenvironment of cryptorchidism. The cell viability was assayed by CCK-8, and cellular DNA damage was detected by gamma-H2AX immunofluo-rescence assay. Flow cytometry was employed to assess pyroptosis index, while western blot, ELISA and PCR were used to examine the expressions of pyroptosis-related proteins (Caspase-1, IL-1beta, NLRP3) and cGAS-STING pathway proteins (cGAS, STING). After STING silencing by siRNA, the expressions of pyroptosis-related proteins were determined. Pyroptosis occurred after heat stimulation of cells. Morphological detection found cell swelling and karyopyknosis. According to the gamma-H2AX immunofluorescence (IFA) assay, the endonuclear green fluorescence was significantly enhanced, the gamma-H2AX content markedly increased, and the endonuclear DNA was damaged. Flow cytometry revealed a significant increase in pyroptosis index. Western blot and PCR assays showed that the expressions of intracellular pyrogenic proteins like Caspase-1, NLRP3 and GSDMD were elevated. The increased STING protein and gene expressions in cGAS-STING pathway suggested that the pathway was intracellularly activated. Silencing STING protein in cGAS-STING pathway led to significantly inhibited pyroptosis. These results indicate that cGAS-STING pathway plays an important role in heat stress-induced pyroptosis of spermatogonial cells. After heat stimulation of spermatogonial GC-1 cells, pyroptosis was induced and cGAS-STING pathway was activated. This study can further enrich and improve the molecular mechanism of cryptorchidism.

Muscle cooling before and in the middle of a session: there are benefits on subsequent localized endurance performance in a warm environment. J Strength Cond Res 38(3): 533-539, 2024-Localized cold-water immersion (CWI) has been shown to facilitate recovery in the middle of a session of exhaustive repeated forearm contractions. However, it has been suggested that these benefits may be attributed to "precooling" the muscle before an activity, as opposed to cooling a previously overheated muscle. Therefore, this study aimed to determine how precooling and mid-cooling affects localized repeated muscular endurance performance in a warm environment. Nineteen subjects completed a familiarization session and 3 laboratory visits, each including 2 exhaustive climbing trials separated by 20 minutes of recovery: PRE CWI (CWI, trial 1; passive sitting [PAS], trial 2); MID CWI (PAS, trial 1; CWI, trial 2); and CONTROL (PAS, trial 1; PAS, trial 2). Climbing trial 1 in PRE CWI was 32 seconds longer than in CONTROL ( p = 0.013; d = 0.46) and 47 seconds longer than in MID CWI ( p = 0.001; d = 0.81). The time of climbing trial 2 after PAS (PRE CWI and CONTROL) was very similar (312 vs. 319 seconds) irrespective of the first trial condition. However, the time of the second trial in MID CWI was 43 seconds longer than in PRE CWI ( p < 0.001; d = 0.63) and 50 seconds longer than in CONTROL ( p < 0.001; d = 0.69). In warm environments, muscle precooling and mid-cooling can prolong localized endurance performance during climbing. However, the effectiveness of mid-cooling may not be as a "recovery strategy" but as a "precooling" strategy to decrease muscle temperature before subsequent performance, delaying the onset of localized heat-induced neuromuscular fatigue.

PURPOSE: To compare different thermal tissue models for head and neck hyperthermia treatment planning, and to assess the results using predicted and measured applied power data from clinical treatments. METHODS: Three commonly used temperature models from literature were analysed: "constant baseline", "constant thermal stress" and "temperature dependent". Power and phase data of 93 treatments of 20 head and neck patients treated with the HYPERcollar3D applicator were used. The impact on predicted median temperature T50 inside the target region was analysed with maximum allowed temperature of 44 °C in healthy tissue. The robustness of predicted T50 for the three models against the influence of blood perfusion, thermal conductivity and the assumed hotspot temperature level was analysed. RESULTS: We found an average predicted T50 of 41.0 ± 1.3 °C (constant baseline model), 39.9 ± 1.1 °C (constant thermal stress model) and 41.7 ± 1.1 °C (temperature dependent model). The constant thermal stress model resulted in the best agreement between the predicted power (P = 132.7 ± 45.9 W) and the average power measured during the hyperthermia treatments (P = 129.1 ± 83.0 W). CONCLUSION: The temperature dependent model predicts an unrealistically high T50. The power values for the constant thermal stress model, after scaling simulated maximum temperatures to 44 °C, matched best to the average measured powers. We consider this model to be the most appropriate for temperature predictions using the HYPERcollar3D applicator, however further studies are necessary for developing of robust temperature model for tissues during heat stress.

This study consists of nine case studies addressing thermal comfort in the public areas of city centres, with particular emphasis on the measurable effects of blue and green infrastructure on thermal exposure. Daytime on-site measurements were taken in summer in the paved areas of squares, in the proximity of water fountains, and in the shade of trees in order to evaluate levels of heat stress based on the universal thermal climate index (UTCI). The differences in UTCI values between the research points confirm substantial cooling associated with high vegetation (trees induced differences up to 10.5 °C in UTCI), while the measurable cooling effect of low vegetation was negligible (not more than 2.3 °C UTCI). It was also quite low around water fountains, spray fountains, and misting systems. It follows that municipal authorities should consider the differences in cooling effect potential of individual types of blue and green infrastructure when incorporating climate adaptation measures into urban planning.

Elevated summer temperature is reported to be the leading cause of stress in dairy and beef cows, which negatively affects various reproductive functions. Follicular cells respond to heat stress (HS) by activating the expression of heat shock family proteins (HSPs) and other antioxidants. HS is reported to negatively affect the bi-directional communication between the follicular cells and the oocyte, which is partly mediated by follicular fluid extracellular vesicles (EVs) released from surrounding cells. As carriers of bioactive molecules (DNA, RNA, protein, and lipids), the involvement of EVs in mediating the stress response in follicular cells is not fully understood. Here we used an in vitro model to decipher the cellular and EV-coupled miRNAs of bovine granulosa cells in response to HS. Moreover, the protective role of stress-related EVs against subsequent HS was assessed. For this, bovine granulosa cells from smaller follicles were cultured in vitro and after sub-confluency, cells were either kept at 37 °C or subjected to HS (42 °C). Results showed that granulosa cells exposed to HS increased the accumulation of ROS, total oxidized protein, apoptosis, and the expression of HSPs and antioxidants, while the viability of cells was reduced. Moreover, 14 and 6 miRNAs were differentially expressed in heat-stressed granulosa cells and the corresponding EVs, respectively. Supplementation of stress-related EVs in cultured granulosa cells has induced adaptive response to subsequent HS. However, this potential was not pronounced when the cells were kept under 37 °C. Taking together, EVs generated from granulosa cells exposed to HS has the potential to shuttle bioactive molecules to recipient cells and make them robust to subsequent HS.

Purpose: Thermal dose-effect relations have demonstrated that clinical effectiveness of hyperthermia would benefit from more controlled heating of the tumor. Hyperthermia treatment planning (HTP) is a potent tool to study strategies enabling target conformal heating, but its accuracy is affected by patient modeling approximations. Homogeneous phantoms models are being used that do not match the body shape of patients in treatment position and often have unrealistic target volumes. As a consequence, simulation accuracy is affected, and performance comparisons are difficult. The aim of this study is to provide the first step toward standardization of HTP simulation studies in terms of patient modeling by introducing the Erasmus Virtual Patient Repository (EVPR): a virtual patient model database.Methods: Four patients with a tumor in the head and neck or the pelvis region were selected, and corresponding models were created using a clinical segmentation procedure. Using the Erasmus University Medical Center standard procedure, HTP was applied to these models and compared to HTP for commonly used surrogate models.Results: Although this study was aimed at presenting the EVPR database, our study illustrates that there is a non-negligible difference in the predicted SAR patterns between patient models and homogeneous phantom-based surrogate models. We further demonstrate the difference between actual and simplified target volumes being used today.Conclusion: Our study describes the EVPR for the research community as a first step toward standardization of hyperthermia simulation studies.

Many workers are exposed to heat stress that can be exacerbated by the type of clothing they wear. The resulted heat strain can lead to short or long-term heat-related disorders. This study aimed to measure clothing properties of sugarcane field workers and evaluate the heat strain by an international standard, predicted heat strain model (PHS). The clothing thermal insulation and evaporative resistance values of sugarcane cutter and chemical sprayer outfits were acquired for the whole body, body regions and specific body parts via thermal manikin measurements. The detailed clothing insulation values of body parts can be utilized in advanced thermo-physiological models, while in this study, the values for the whole body together with weather data were used in PHS. Estimated duration limited exposure times (DLE) for an hour-by-hour prediction over a workday and for a range of high humidity scenarios were calculated. Such evaluation tools can be used for risk assessment and management to support organizational measures and prepare equipment and materials in the case of hot weather events in order to avoid dehydration and other heat-related disorders.

We present here a fatal case of heatstroke, involving olanzapine and levomepromazine medications. A male in his sixties was found dead in his storage room in the middle of August, with a high rectal temperature. Autopsy revealed congestion of the lungs without any specific findings. Quantitative toxicological analysis demonstrated concentrations of olanzapine, levomepromazine, 7-aminonitrazepam, and 7-aminoflunitrazepam in a femoral blood sample of 0.433 µg/mL, 0.177 µg/mL, 0.604 µg/mL, and 0.041 µg/mL, respectively. The concentration of olanzapine exceeded toxic levels; however, levomepromazine level was within the therapeutic range. Due to the blocking mechanism of both olanzapine and levomepromazine against muscarinic receptors, they might depress sweating and impair heat dissipation. Based on autopsy findings, results of toxicological examination, and investigation by the authorities, we concluded that the cause of death was heatstroke under the influence of olanzapine and levomepromazine.

The objective of this current work was to determinate the effect of high temperatures on milk production of dairy cows in southern Slovakia in the year 2015. The hypotheses that milk production is influenced by the altitude and cooling were tested. Production data included 227,500 test-day records belonging to 34 Holstein breed herds situated in lowlands, 115 to 150 m above sea level (ASL) and kept in free-stall housing. Dairy farms were classified into groups based on cooling system. The first group of cows (19 herds) was cooled evaporative (foggers) and forced ventilation, and the second group (15 herds) was using cooled only forced ventilation (automatically controlled fans in housing and feeding areas). During the period from May to September, 36 summer and 22 tropical days were recorded, 37 days had a mean thermal humidity index value above 72.0, and on 34 days we recorded mean values above 78.0. The highest milk yields were recorded at the altitude 1 (115 m ASL) (9219.0 kg year-1; 10327.0 kg year-1) and the lowest at the altitude 2 (126 m ASL) (7598.7 kg year-1; 8470.21 kg year-1) (P < 0.001). Dairy cows cooled evaporative milked significantly more milk than cows cooled only with forced air flow (9650.4 kg vs. 8528.0 kg; P < 0.001). Fat and protein production differed also significantly (364.0 kg vs. 329.5 kg, P < 0.001; 312.2 kg vs. 279.7 kg, P < 0.001). It can be concluded that not only heat stress but also location farm above sea level can affect milk production. Evaporative cooling associated with increased air velocity is the appropriate protection against high temperatures.

We compared selected thermal indices in their ability to predict heat-related mortality in Prague, Czech Republic, during the extraordinary summer 2015. Relatively, novel thermal indices-Universal Thermal Climate Index and Excess Heat Factor (EHF)-were compared with more traditional ones (apparent temperature, simplified wet-bulb globe temperature (WBGT), and physiologically equivalent temperature). The relationships between thermal indices and all-cause relative mortality deviations from the baseline (excess mortality) were estimated by generalized additive models for the extended summer season (May-September) during 1994-2014. The resulting models were applied to predict excess mortality in 2015 based on observed meteorology, and the mortality estimates by different indices were compared. Although all predictors showed a clear association between thermal conditions and excess mortality, we found important variability in their performance. The EHF formula performed best in estimating the intensity of heat waves and magnitude of heat-impacts on excess mortality on the most extreme days. Afternoon WBGT, on the other hand, was most precise in the selection of heat-alert days during the extended summer season, mainly due to a relatively small number of "false alerts" compared to other predictors. Since the main purpose of heat warning systems is identification of days with an increased risk of heat-related death rather than prediction of exact magnitude of the excess mortality, WBGT seemed to be a slightly favorable predictor for such a system.
