Diclofenac is a drug commonly used in human and veterinary medicine for the treatment of diseases associated with inflammation and pain. Medicinal products enter waste and surface waters on an everyday basis and contaminate the aquatic environment. Fish are therefore permanently exposed to these chemicals dissolved in their aquatic environment. To simulate variable environmental conditions, the aim of our study was to examine adverse effects of diclofenac under different temperatures of cell incubation (18, 21, 24, 27 and 30 °C). Cyto-toxic and -static effects of diclofenac in concentrations of 0.001 mcg/ml, 0.01 microg/ml, 0.1 mcg/ml, 1 mcg/ml, 10 mcg/ml and 100 mcg/ml for the carp (Cyprinuscarpio) cultured leukocytes were quantified using detection of lactate dehydrogenase released from damaged cells. Overall DCF cytotoxicity was relatively low and its impact was pronounced at higher temperature and DCF concentration. Cells growth inhibition is changing more rapidly but it is high mainly at the highest concentration from low temperature. DNA fragmentation was not detected in tested leukocyte cell line. CYP450 increased diclofenac cytotoxicity only at the highest concentration but at incubation temperatures 18 and 27 °C. Leukocyte viability is essential for immune functions and any change can lead to reduction of resistance against pathogens, mainly in cold year seasons, when the immune system is naturally suppressed.

The ability of the DGT technique to predict Hg2+ bioavailability for input tissues (skin, gills, and scales) of common carp in the presence of chloride ions and humic acid (HA) was evaluated. The mercury accumulation by the DGT units and input tissues of carp decreased with an increasing concentration of chloride ions (29-180 mg L-1) and HA (0-5 mg L-1). In the presence of chloride ions and HA, statistically significant correlations (Pearson's correlation coefficients 0.731-0.954) were determined between the rate of mercury accumulation by input tissues of carp and the rate of mercury accumulation by the DGT units. The laboratory experiments suggest the possibility of using the DGT technique for predicting the mercury bioaccumulation in natural aquatic ecosystems instead of commonly used input tissues of fish.

The thermoregulation behavior of Lucilia sericata larvae (Diptera: Calliphoridae), a necrophagous species that feeds on vertebrate cadavers, was investigated. These larvae require high heat incomes to develop, and can elevate temperatures by forming large aggregates. We hypothesized that L. sericata larvae should continue to feed at temperatures up to 38 °C, which can be reached inside larval masses. Thermal regulation behavior such as movement between a hot food spot and colder areas was also postulated. The hypotheses were tested by tracking for 1 h the activity of single, starved third instar larvae in a Petri dish containing 1 food spot (FS) that was heated to a constant temperature of 25 °C, 34 °C or 38 °C with an ambient temperature of 25 °C. The influence of previous conspecific activity in the food on larval behavior was also tested. The crops of larvae were dissected to monitor food content in the digestive systems. Based on relative crop measurements, larvae fed at all food temperatures, but temperature strongly affected larval behavior and kinematics. The total time spent by larvae in FS and the duration of each stay decreased at high FS temperature. Previous activity of conspecifics in the food slightly increased the time spent by larvae in FS and also decreased the average distance to FS. Therefore, necrophagous L. sericata larvae likely thermoregulate during normal feeding activities by adjusting to local fluctuations in temperature, particularly inside maggot masses. By maintaining a steady internal body temperature, larvae likely reduce their development time.

The mercury bioaccumulation by common carp (Cyprinus carpio L.) tissues (gills, skin, eyes, scales, muscle, brain, kidneys, liver, and spleen) and the capability of the diffusive gradient in thin film (DGT) technique to predict bioavailability of mercury for individual carp's tissues were evaluated. Carp and DGT units were exposed to increasing concentrations of mercury (Hg2+: 0 μg L-1, 0.5 μg L-1, 1.5 μg L-1and 3.0 μg L-1) in fish tanks for 14 days. In the uncontaminated fish group, the highest mercury concentration was determined in the muscle tissues and, in fish groups exposed to mercury, the highest mercury concentration was determined in the detoxification (kidneys) and input (gills) organs. A strong and positive correlation between the rate of mercury uptake by the DGT technique and the rate of mercury accumulation by fish tissues (gills, skin, scales, and eyes) was observed.
