The potential of mMass software search tool with new compound libraries was demonstrated on metabolomics of Scedosporium prolificans, S. apiospermum and Pseudallescheria boydii sensu stricto. Cyclic peptides pseudacyclins, small molecular weight tyroscherin analogues and various lipids were annotated by public software tool (http://www.mmass.org) utilising accurate matrix-assisted laser desorption/ionisation mass spectral data of intact fungal spores. Electrospray ionisation combined with tandem mass spectrometry was used for monohexosylceramide characterisation in fungal extracts.
Pseudallescheria boydii sensu lato is an emerging fungal pathogen causing fatal infections in both immunocompromised and immunocompetent hosts. In this work, two P. boydii isolates (human and animal origin) have been identified as being producers of cyclic peptides. Five putative nonribosomal peptides with a unique structure, which have been named pseudacyclins, were characterized by nuclear magnetic resonance spectroscopy and mass spectrometry. The most abundant representative of the pseudacyclins was quantified also on fungal spores. The presence of these peptides on inhaled fungal spores creates the possibility for exploitation of pseudacyclins as early indicators of fungal infections caused by Pseudallescheria species.

V práci sa opisuje prípad mykotickej keratitídy s indukovanou uveitídou u mladej zdravej ženy pracujúcej v sklade ovocia a zeleniny, ktorá sa rozvinula v teréne malej erózie na rohovke. Afekcia po opakovane negatívnych kultivačných vyšetreniach nereagovala na liečbu bežnými antibiotikami. Po kultivaci na Sabouraudovom agare bola získaná kultúra vláknitej mikromycéty, identifikovaná ako Scedosporium apiospermum, v našich podmienkach je to prvý prípad tohto pôvodcu mykotického ochorenia oka. Pacientka reagovala úspešne na liečbu miconazolom, nález na rohovke PO zodpovedá vaskularizovanému leukómu, zraková ostrosť 5/50, v bodúcnosti je plánovaná perforujúca kerotoplastika.
Affected is young healthy woman working in a fruit and vegetable store. Affection arised on cornea in the area of previous erosion. Current antibiotics did not have any effect on the affection after repeated negative cultivative examina done on Sabourauďs glucose agar and was identified as Scedosporium apiospermum. This is the first case of mycotic eye infection of this etiology in our conditions. The patient reacted succesfuly on miconazol treatment. The finding on cornea equals vascularized leucoma, visual acuity is 5/50. In the future perforative keratoplastic is planned.
