The aim of this study was to determine particle size distributions (PSDs) generated by six pocket inhalers (2 × MDIs – metered dose inhalers and 4 × DPIs – dry powder inhalers) used in the treatment of asthma and chronic obstructive pulmonary disease (COPD). Two methods were compared – cascade impactor and time-of-flight online spectrometer. The results showed that the main modes positions of mass PSDs obtained by either of the methods were comparable. During the measurements of number PSDs by using of APS spectrometer, we measured bimodal distributions for MDI inhalers and monomodal ones for DPI inhalers. The differences of PSDs measured under ambient and respiratory tract conditions were minimal and did not influence the deposition area of inhalants. The results showed that the determination of the PSDs by online spectrometer is a suitable alternative to the traditional methods of measuring the particle size distributions of inhalation aerosol.
BACKGROUND: Although keratin hydrolysates have become established as standard components in hair and nail cosmetics, studies on the moisturizing effects of keratin hydrolysates do not appear among contemporary literature. OBJECTIVES: To test if adding keratin hydrolysate into an ointment base increases hydration of the skin and improves skin barrier function, or diminishes trans-epidermal water loss. METHODS: Formulations were prepared containing 2%, 4%, and 6% keratin hydrolysates (based on weight of the ointment base). The moisturizing properties of keratin hydrolysates were tested by measuring skin hydration, trans-epidermal water loss and skin pH; measurements were carried out at intervals of 1, 2, 3, 4, 24, and 48 h. Testing was conducted on 10 women. RESULTS: As regards hydration, adding 2% keratin hydrolysate to the ointment base is optimal, as an increase of 14%-23% occurs in hydration of the stratum corneum. For trans-epidermal water loss, adding 4% KH to the ointment base is preferential, as this triggers a 26%-46% decrease in trans-epidermal water loss. CONCLUSIONS: Keratin hydrolysate acts as a humectant (it binds water from lower layers of the epidermis to the stratum corneum) as well as an occlusive (it reduces trans-epidermal water loss). The highly favorable properties of keratin hydrolysates are attributed to the wide distribution of keratin hydrolysates molecular weights; low-molecular weight fractions easily penetrate the SC, while high-molecular weight fractions form a protective film on the epidermis. Adding keratin hydrolysates to the ointment base did not cause phase separation even after 6 mo storage.

V práci sa študuje vplyv pomocných látok na permeáciu kofeínu z dermálnych polotuhých liekov v in vitro podmienkach. Vďaka výhodným vlastnostiam nachádza kofeín využitie v dermálnych hydrogéloch. Dôležitý je tiež výber pomocných látok zo skupiny humektantov a katiónových tenzidov. Ich cieľom je zabezpečiť dlhodobú stabilitu hydrogélov a čo najlepšiu permeáciu použitého liečiva z gélov. Predmetom výskumu je štúdium vplyvov rôznych pomocných látok na permeáciu kofeínu z gélov a taktiež ich vplyv na reologické vlastnosti hydrogélov. Na základe získaných výsledkov sa určilo nasledovné optimálne zloženie pre kofeínový gél: 2,5% chitosan + 0,1% kofeín + 10% propylénglykol + 0,1% benzyldimetylhexadecylamóniumchlorid.
The subject of this paper is a study of the influence of excipients on caffeine permeation from dermal semisolids drugs. Due to its advantageous properties, caffeine can be successfully used in dermal hydrogels. The choice of excipients from the group of humectants and cationic surfactants also plays an important role. The excipients should ensure long-term stability of hydrogels and the best permeation of the drug from gels. In particular, the influence of various excipients on caffeine permeation from gels and at the same time their influence on rheological properties of hydrogels were investigated. Based on the obtained results, the following optimal composition of caffeine gel was determined: 2.5 % chitosan + 0.1 % caffeine + 10 % propylenglycol + 0.1 % benzyldimethylhexadecylammonium chloride. Promašťují, hydratují a zvláčňují pokožku a tím se stávají důležitou součástí léčby chronických kožních onemocnění a uplatňují se i v prevenci rozvoje kožních onemocnění.
