Objevení struktury vazopresinu du Vigneaudem a jeho spolupracovníky pøed padesáti léty je pøíležitostí ke zpìtnému pohledu na tento významný objev. Na vasokonstrikèní aktivitu kyselých extraktù hypofýzy upozornili poprvé londýnští fyziologové Oliver a Schäfer již v roce 1895. Další aktivity tìchto extraktù byly popsány v prùbìhu prvých dvou desetiletí dvacátého století. Extrakty byly pak èistìny soudobými biochemickými postupy; pro výskyt nejménì dvou biologicky aktivních komponent, oxytocické a vazopresorické, byla poèátkem tøicátých let snesena øada experimentálních dùkazù. Pøesto trval spor o poètu pøítomných hormonù a jejich charakteru až do poèátku padesátých let. Dík novým purifikaèním a analytickým postupùm (protiproudné vytøepáváni, sloupcová chromatografie, nové metody sekvence bílkovin a peptidù) dospìla du Vigneaudova laboratoø na Cornell University v Ithace (New York) v roce 1953 ke struktuøe oxytocinu a o rok pozdìji vazopresinu. Obì struktury byly potvrzeny totální syntézou. Za tyto objevy byl Vincent du Vigneaud odmìnìn v roce 1955 Nobelovou cenou za chemii.
Clarification of the vasopressin structure by du Vigneaud and his coworkers fifty years ago offers an opportunity to look back on the history and pre-history of this crucial discovery. First indications for active principles in the pituitary gland go back to 1895 when Oliver and Schäfer in London recognized strong vasopressor activities of acid-extracts of the gland. Further activities were described in the course of the first two decades of 20th century. Later on, the extracts were purified by the current biochemical methods used in that time, and it was finally shown that at least two components, an oxytocic and a vasopressor, were present. The disputes concerning their number and nature lasted actually until the final synthesis by du Vigneaud in 1954. The group of du Vigneaud at the Cornell University in Ithaca, NY, having used newly developed purification and analytical methods (countercurrent distribution, column chromatography, new procedures for sequencing of proteins and peptides) eventually reached in 1953 the structure of oxytocin, and one year later the one of vasopressin; both were verified by a total synthesis. Vincent du Vigneaud was awarded by the Nobel Prize for Chemistry in 1955 for these discoveries.
