Acanthamoebae success as human pathogens is largely due to the highly resistant cysts which represent a crucial problem in treatment of Acanthamoeba infections. Hence, the study of cyst wall composition and encystment play an important role in finding new therapeutic strategies. For the first time, we detected high activity of cytoskeletal elements - microtubular networks and filamentous actin, in late phases of encystment. Cellulose fibrils - the main components of endocyst were demonstrated in inter-cystic space, and finally in the ectocyst, hereby proving the presence of cellulose in both layers of the cyst wall. We detected clustering of intramembranous particles (IMPs) and their density alterations in cytoplasmic membrane during encystment. We propose a hypothesis that in the phase of endocyst formation, the IMP clusters represent cellulose microfibril terminal complexes involved in cellulose synthesis that after cyst wall completion are reduced. Cyst wall impermeability, due largely to a complex polysaccharide (glycans, mainly cellulose) has been shown to be responsible for Acanthamoeba biocide resistance and cellulose biosynthesis pathway is suggested to be a potential target in treatment of Acanthamoeba infections. Disruption of this pathway would affect the synthesis of cyst wall and reduce considerably the resistance to chemotherapeutic agents.

Meňavky rodu Acanthamoeba Volkonsky, 1931 sú ubikvistické organizmy, s amfizoickým spôsobom života a kozmopolitným výskytom. Patogénne kmene sú pôvodcami ťažko liečiteľného ochorenia – granulomatóznej meňavkovej encefalitídy (GAE) a kožných infekcií u imunosuprimovaných pacientov a bolestivého ochorenia rohovky – akantamébovej keratitídy (AK) u imunokompetentných jedincov. Predpokladanou vstupnou bránou infekcie je nazofaryngeálna sliznica, pľúcny parenchým, infekcia z kožných lézií (GAE, kožné infekcie), nedostatočná hygiena pri používaní kontaktných šošoviek alebo pri korneálnej traume (AK). Symptómy jednotlivých ochorení sú nešpecifické a často veľmi variabilné, čo spolu s nedostatočnou informovanosťou odbornej verejnosti výrazne sťažuje včasné určenie správnej diagnózy. Kým v prípade GAE a kožných infekcií sú možnosti terapie obmedzené a málo úspešné, v prípade AK bolo úspešne použitých viacero typov antimykotík a antibiotík, avšak terapia je často veľmi komplikovaná a zdĺhavá.
Amoebae of the genus Acanthamoeba Volkonsky, 1931 are ubiquitous, amphizoic organisms with a cosmopolitan distribution. Pathogenic strains are the causative agents of a difficult to treat disease, granulomatous amoebic encephalitis (GAE), and skin infections in immunocompromised individuals, and of a painful corneal disease – amoebic keratitis (AK) in immunocompetent individuals. The major portals of entry are the nasopharyngeal mucosa, pulmonary parenchyma, skin lesions (GAE, skin infections), eyes in contact lenses wearers with a history of improper contact lens wear and care, or corneal trauma (AK). Symptoms of the diseases are non-specific and variable which alongside with the lack of awareness among health care professionals often hamper early diagnosis. While treatment options for GAE and skin infections are limited and poorly effective, various antifungals and antimicrobials have proved beneficial in AK, although the therapy is often complicated and long.
