A high VO(2)max in middle-age is related to high metabolic flexibility and lowered risk of metabolic diseases. However, the influence of a high VO(2)max induced by years of regular training in middle-age on protein expression related to muscle metabolism is not well studied. This study measures key proteins involved in mitochondrial oxidation, glucose and lipid metabolism in skeletal muscle of trained and untrained middle-aged men. 16 middle-aged men, matched for lean body mass, were recruited into an endurance trained (TR, n=8) or an untrained (CON, n=8) group based on their VO(2)max. A muscle biopsy was obtained from m. vastus lateralis and protein levels were analyzed by Western blotting. The TR had higher protein levels of mitochondrial complex III-V, endothelial lipase (EL) and perilipin 5 compared to the CON. Glycogen synthase (P=0.05), perilipin 3 (P=0.09) and ATGL (P=0.09) tended to be higher in TR than CON, but there was no difference in AKT I/II, HKII, GLUT4 and LPL protein expression. Lastly, there was a positive correlation between plasma HDL and EL (R(2)=0.53, P<0.01). In conclusion, a high VO(2)max in middle-aged men was as expected is reflected in higher muscle oxidative capacity, but also in higher endothelial lipase and perilipin 5 expression and a borderline higher glycogen synthase protein expression, which may contribute to a higher metabolic flexibility.

Poly-unsaturated fatty acids (PUFAs) are considered to be healthier than saturated fatty acids (SFAs), but others postulate that especially the ratio of omega-6 to omega-3 PUFAs (n6/n3 ratio) determines health. Health can be determined with biomarkers, but functional health status is likely better reflected by challenge tests that assess metabolic flexibility. The aim of this study was to determine the effect of high-fat diets with different fatty acid compositions, but similar n6/n3 ratio, on metabolic flexibility. Therefore, adult male mice received isocaloric high-fat diets with either predominantly PUFAs (HFpu diet) or predominantly SFAs (HFs diet) but similar n6/n3 ratio for six months, during and after which several biomarkers for health were measured. Metabolic flexibility was assessed by the response to an oral glucose tolerance test, a fasting and re-feeding test and an oxygen restriction test (OxR; normobaric hypoxia). The latter two are non-invasive, indirect calorimetry-based tests that measure the adaptive capacity of the body as a whole. We found that the HFs diet, compared to the HFpu diet, increased mean adipocyte size, liver damage, and ectopic lipid storage in liver and muscle; although, we did not find differences in body weight, total adiposity, adipose tissue health, serum adipokines, whole body energy balance, or circadian rhythm between HFs and HFpu mice. HFs mice were, furthermore, less flexible in their response to both fasting- re-feeding and OxR, while glucose tolerance was indistinguishable. To conclude, the HFs versus the HFpu diet increased ectopic fat storage, liver damage, and mean adipocyte size and reduced metabolic flexibility in male mice. This study underscores the physiological relevance of indirect calorimetry-based challenge tests.

Using skeletal muscle homogenates for respirometry has many advantages, but the main challenge is avoiding the damage to outer mitochondrial membrane (OMM) and complex I. By optimising the amount of muscle and careful titration of substrates and inhibitors we developed a new protocol and compared it to isolated mitochondria. We found acceptable damage to OMM (~10-15% increment of oxygen flux after addition of cytochrome c) and to complex I (~70% of electron flux). Homogenate retained ~90% of phosphorylation capacity of isolated mitochondria. The use of fresh homogenate was crucial as mitochondrial function declined rapidly after 2-3h of cold storage.

This study was a comparison between Ru-catalysts and similar, previously investigated, Pt-catalysts. In this paper, ruthenium catalysts for catalytic wet air oxidation are prepared, characterized and tested. Both catalysts were supported on commercial CeO2 as well as mixed oxide Zr(0.1)(Ce(0.75)Pr(0.25))(0.9)O2. The catalysts were characterized by measuring the oxygen storage capacities (OSC), BET, XRD, FTIR and chemisorption of hydrogen. In addition, the effect of sintering (treatments under H2) was compared with both of the catalysts. The comparison of the results showed that initial intrinsic activity of ruthenium is not significantly influenced by the type of the support, which is contrast to platinum. Furthermore, the particle size of Ru had an important effect on CWAO activity: the higher the particle size, the better the activity. This was different with Pt-catalysts, where the optimal particle size was smaller, having about 15% of metal dispersion.

Over a period of several decades multiple leaks of large volumes from storage facilities located near Hnevice (Czech Republic) have caused the underlying Quaternary aquifer to be severely contaminated with nonaqueous phase liquid (NAPL) petroleum hydrocarbons. Beginning in the late 1980's the NAPL plume started to shrink as a consequence of NAPL dissolution exceeding replenishment and due to active remediation. The subsurface was classified geochemically into four different zones, (i) a contaminant-free zone never occupied by NAPL or dissolved contaminants, (ii) a re-oxidation zone formerly occupied by NAPL, (iii) a zone currently occupied by NAPL, and (iv) a lower fringe zone between the overlying NAPL and the deeper underlying contaminant-free zone. The study investigated the spatial and temporal variability of the redox zonation at the Hnevice site and quantified the influence of iron-cycling on the overall electron balance. As a first step inverse geochemical modelling was carried out to identify possible reaction models and mass transfer processes. In a subsequent step, two-dimensional (forward) multi-component reactive transport modelling was performed to evaluate and quantify the major processes that control the geochemical evolution at the site. The study explains the observed enrichment of the lower fringe zone with ferrihydrite as a result of the re-oxidation of ferrous iron. It suggests that once the NAPL zone started to shrink the dissolution of previously formed siderite and FeS by oxygen and nitrate consumed a significant part of the oxidation capacity for a considerable time period and therefore limited the penetration of electron acceptors into the NAPL contaminated zone.
