The purpose of the present study was to investigate the intra- and inter-individual variability in arm-leg coordination during the underwater phase of the turn segment in 200 m breaststroke. Thirteen male swimmers were recruited and performed a 200 m breaststroke in a pre-calibrated 25 m pool. Sub-phases during the underwater segment were obtained using a notational analysis, and the mean velocity, displacement and duration during each sub-phase were obtained. A hierarchical cluster analysis (HCA) was performed using the analysed variables in all phases to identify inter-individual variability and random intra-individual variability. In addition, a linear mixed model (LMM: lap as a fixed effect and the participant as a random effect) was conducted to investigate systematic intra-individual variability. HCA identified three coordination patterns that were distinguished by the timing of the dolphin kick relative to the arm pull-out and the duration of the glide with arms at the side. All swimmers except one performed the arm pull-out after the dolphin kick. Nine swimmers maintained one coordination pattern, but other swimmers switched their coordination during the trial, particularly by shortening the duration of the glide with arms at the side. LMM showed a linear decrease (from the first to the last turn) in the time gap between the end of the dolphin kick and the start of the arm pull-out (a glide with the streamlined body position; F = 9.64, p = 0.034) and the glide duration with the arms at the side (F = 11.66, p = 0.015). In conclusion, both inter- and intra-individual variabilities during the underwater phase were evident in 200 m breaststroke turns, which were categorised into three patterns based on the timing of the dolphin kick and the duration of glides.

In breaststroke races, the dolphin kick could finish before, at the same time, or during the arm pull-out, but it is unclear how swimmers perform this technique. The aim of this study was to investigate whether swimmers glide between the dolphin kick and arm pull-out, favour continuity or even overlap those two phases, as it would impact the active underwater sequence. Fourteen international and national male swimmers performed 100-m breaststroke with all-out effort in a pre-calibrated 25 m swimming pool. A multi-camera system tracked the head of the swimmers. Key points of the active underwater sequence were obtained from notational analysis. A hierarchical cluster analysis identified three coordination profiles. All swimmers started their dolphin kick before the arm pull-out. However, one swimmer started the arm pull-out before the end of the dolphin kick, seven swimmers started the arm pull-out after the end of the dolphin kick, and four swimmers synchronised the beginning of the arm pull-out and the end of the dolphin kick, while two other swimmers mixed two coordination profiles among the start and the three turns. Those different profiles allow achieving similar performance outcome, suggesting individual training regarding the underwater phase.

Introduction: The objective of the study was to examine the relationship between the values of selected parameters of physical function, body composition, body mass index (BMI) and biochemical markers of metabolic health with the total testosterone (TT) levels in adult males. We aimed to analyse the correlation between these values and variations in the TT levels. Methods: A total of 17 subjects (age = 50.2 ± 8.1 years, TT = 11.4 ± 3.8 nmol/l) were included in the study. Subjects were tested on physical function (1RM on leg press, bench-press, handgrip, VO2max), body composition (DXA), biochemical parameters (morning fasting blood samples). Results: TT was inversely correlated with abdominal circumference (AC) (p < 0.01) and with overall body fat, measured in kg (p < 0.01). On a biochemical level, significant correlations were found between TT and insulin (p < 0.01), and TT and homeostasis model assessment of insulin resistance (HOMA-IR) (p < 0.01). Physical function, muscle strength or lean mass were not significantly correlated with TT. Conclusion: The main finding of this study was that testosterone levels had a strong inverse correlation with abdominal circumference and total body fat mass. On metabolic level, strong inverse correlation was also found between TT with insulin and TT with HOMA-IR. However, we did not find statistically significant correlation between total testosterone levels and lean mass, muscle strength or physical function in middle aged males.

AIM: Chitinase-3-like protein 1 (CHI3L1) is involved in tissue remodelling and inflammatory processes. Plasma levels are elevated in patients with insulin resistance and T2DM. We recently showed that CHI3L1 and its receptor protease-activated receptor 2 (PAR-2) are expressed in skeletal muscle. Activation of PAR-2 by CHI3L1 protects against TNF-α-induced inflammation and insulin resistance. However, the effect of exercise on CHI3L1 and PAR-2 signalling remains unknown. The aim of this work was to study the impact of exercise on CHI3L1 production and the effect of CHI3L1/PAR-2 signalling on skeletal muscle growth and repair. METHODS: Three human exercise studies were used to measure CHI3L1 plasma levels (n = 32). In addition, muscle and adipose tissue CHI3L1 mRNA expression was measured in response to acute and long-term exercise (n = 24). Primary human skeletal muscle cells were differentiated in vitro, and electrical pulse stimulation was applied. In addition, myoblasts were incubated with CHI3L1 protein and activation of MAP kinase signalling as well as proliferation was measured. RESULTS: Circulating CHI3L1 levels and muscle CHI3L1 mRNA were increased after acute exercise. In addition, CHI3L1 mRNA expression as well as CHI3L1 secretion was enhanced in electrically stimulated cultured myotubes. Incubation of cultured human myoblasts with CHI3L1 protein leads to a strong activation of p44/42, p38 MAPK and Akt as well as enhanced myoblast proliferation. CONCLUSION: Our findings suggest that CHI3L1 is induced by acute exercise and that CHI3L1/PAR-2 signalling activates myocyte proliferation, which is important for restructuring of skeletal muscle in the response to exercise training.

BACKGROUND: The marine n-3 fatty acids, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) exert numerous beneficial effects on health, but their potency to improve treatment of type 2 diabetic (T2D) patients remains poorly characterized. We aimed to evaluate the effect of a combination intervention using EPA + DHA and the insulin-sensitizing drug pioglitazone in overweight/obese T2D patients already treated with metformin. METHODS: In a parallel-group, four-arm, randomized trial, 69 patients (66 % men) were assigned to 24-week-intervention using: (i) corn oil (5 g/day; Placebo), (ii) pioglitazone (15 mg/day; Pio), (iii) EPA + DHA concentrate (5 g/day, containing ~2.8 g EPA + DHA; Omega-3), or (iv) pioglitazone and EPA + DHA concentrate (Pio& Omega-3). Data from 60 patients were used for the final evaluation. At baseline and after intervention, various metabolic markers, adiponectin and cytokines were evaluated in serum using standard procedures, EPA + DHA content in serum phospholipids was evaluated using shotgun lipidomics and mass spectrometry, and hyperinsulinemic-euglycemic clamp and meal test were also performed. Indirect calorimetry was conducted after the intervention. Primary endpoints were changes from baseline in insulin sensitivity evaluated using hyperinsulinemic-euglycemic clamp and in serum triacylglycerol concentrations in fasting state. Secondary endpoints included changes in fasting glycemia and glycated hemoglobin (HbA1c), changes in postprandial glucose, free fatty acid and triacylglycerol concentrations, metabolic flexibility assessed by indirect calorimetry, and inflammatory markers. RESULTS: Omega-3 and Pio& Omega-3 increased EPA + DHA content in serum phospholipids. Pio and Pio& Omega-3 increased body weight and adiponectin levels. Both fasting glycemia and HbA1c were increased by Omega-3, but were unchanged by Pio& Omega-3. Insulin sensitivity was not affected by Omega-3, while it was improved by Pio& Omega-3. Fasting triacylglycerol concentrations and inflammatory markers were not significantly affected by any of the interventions. Lipid metabolism in the meal test and metabolic flexibility were additively improved by Pio& Omega-3. CONCLUSION: Besides preventing a modest negative effect of n-3 fatty acids on glycemic control, the combination of pioglitazone and EPA + DHA can be used to improve lipid metabolism in T2D patients on stable metformin therapy. The health-promoting effects of regular exercise are well known, and myokines may mediate some of these effects. The small leucine-rich proteoglycan decorin has been described as a myokine for some time. However, its regulation and impact on skeletal muscle has not been investigated in detail. In this study, we report decorin to be differentially expressed and released in response to muscle contraction using different approaches. Decorin is released from contracting human myotubes, and circulating decorin levels are increased in response to acute resistance exercise in humans. Moreover, decorin expression in skeletal muscle is increased in humans and mice after chronic training. Because decorin directly binds myostatin, a potent inhibitor of muscle growth, we investigated a potential function of decorin in the regulation of skeletal muscle growth. In vivo overexpression of decorin in murine skeletal muscle promoted expression of the pro-myogenic factor Mighty, which is negatively regulated by myostatin. We also found Myod1 and follistatin to be increased in response to decorin overexpression. Moreover, muscle-specific ubiquitin ligases atrogin1 and MuRF1, which are involved in atrophic pathways, were reduced by decorin overexpression. In summary, our findings suggest that decorin secreted from myotubes in response to exercise is involved in the regulation of muscle hypertrophy and hence could play a role in exercise-related restructuring processes of skeletal muscle.
