Hemodynamické parametry hrají významnou roli ve vývoji intrakraniálních aneurysmat a jejich změny v průběhu dlouhodobého sledování mohou vést ke zvýšení rizika ruptury či k samotné ruptuře aneurysmat. Definování těchto změn by významně přispělo k pochopení vývoje a ruptury aneurysmatu. V této jedinečné práci jsou analyzována data čtyř incidentálních neprasklých intrakraniálních aneurysmat u čtyřech pacientů, kteří byli sledování v průměru po dobu 5 let až do chvíle ruptury aneurysmatu. Bylo provedeno matematické modelování hemodynamiky z dostupných dvou až tří angiografických vyšetření od každého pacienta, získaných během sledování, a změny hodnot hemodynamických parametrů byly analyzovány. Až na jedno, původně fusiformní, disekující aneurysma, velikost tří aneurysmat byla popsána na vyšetření MRA jako stacionární. Hodnoty minimálního smykového napětí se významně snížily a velikost oblasti s minimálním smykovým napětím ve vaku aneurysmat se významně zvýšily v průběhu času a výsledky naznačily podíl snižujících se hodnot smykového tření a rostoucí oblasti nízkého smykového napětí v průběhu času na zvyšování rizika ruptury aneurysmatu. K přesnému posouzení rizika ruptury aneurysmatu pomocí matematického modelování hemodynamiky, je nutné analyzovat více než dva modely aneurysmat a zaměřit se na signifikantní změny v hodnotách hemodynamických veličin.
Hemodynamic parameters play a significant role in the development of intracranial aneurysms and their time-depending changes during a prolonged follow-up period could lead to an increasing risk of rupture or to aneurysm rupture. The characteristics of these changes could bring significant information about the development of aneurysms and their rupture. In this unique study, we analyzed four incidental unruptured intracranial aneurysms in four patients who were followed-up for a mean period of 5 years until their rupture. We performed computational fluid dynamic simulations with the data from two or three follow-up angiographic examinations from each patient and analyzed the results with regard to time-dependent changes in terms of the values of hemodynamic parameters. Except one aneurysm of a fusiform dissecting origin, three aneurysms on MRA were described as non-growing. In the aneurysm domes, the minimal wall shear stress decreased and the low wall shear stress area increased significantly over time; the results indicated that the time-dependent changes such as decreasing values of minimal wall shear stress and increasing values of low wall shear stress area could lead to an increased risk of rupture. For accurate evaluation of the rupture risk using computational fluid dynamic simulation, it is important to analyze more than two models of aneurysm during a follow-up period and focus on significant changes in the values of hemodynamic parameters.

Noise exposure during the critical period of postnatal development in rats results in anomalous processing of acoustic stimuli in the adult auditory system. In the present study, the behavioral consequences of an acute acoustic trauma in the critical period are assessed in adult rats using the acoustic startle reflex (ASR) and prepulse inhibition (PPI) of ASR. Rat pups (strain Long-Evans) were exposed to broad-band noise of 125 dB SPL for 8 min on postnatal day 14; at the age of 3-5 months, ASR and PPI of ASR were examined and compared with those obtained in age-matched controls. In addition, hearing thresholds were measured in all animals by means of auditory brainstem responses. The results show that although the hearing thresholds in both groups of animals were not different, a reduced strength of the startle reflex was observed in exposed rats compared with controls. The efficacy of PPI in exposed and control rats was also markedly different. In contrast to control rats, in which an increase in prepulse intensity was accompanied by a consistent increase in the efficacy of PPI, the PPI function in the exposed animals was characterized by a steep increase in inhibitory efficacy at low prepulse intensities of 20-30 dB SPL. A further increase of prepulse intensity up to 60-70 dB SPL caused only a small and insignificant change of PPI. Our findings demonstrate that brief noise exposure in rat pups results in altered behavioral responses to sounds in adulthood, indicating anomalies in intensity coding and loudness perception.

The development of neurogenic pulmonary edema (NPE) can be elicited by an immediate epidural balloon compression of the thoracic spinal cord. To evaluate whether a slower balloon inflation could prevent NPE development, we examined the extent of NPE in animals lesioned with a rapid (5 microl - 5 microl - 5 microl) or slow rate (3 microl - 2 microl - 2 microl - 2 microl - 2 microl - 2 microl - 2 microl) of balloon inflation. These groups were compared with the NPE model (immediate inflation to 15 microl) and with healthy controls. Slow balloon inflation prevented NPE development, whereas the pulmonary index and histology revealed a massive pulmonary edema in the group with a rapid rate of balloon inflation. Pulmonary edema was preceded by a considerable decrease in heart rate during the inflation procedure. Moreover, rapid inflation of balloon in spinal channel to either 5 microl or 10 microl did not cause NPE. Thus, a slow rate of balloon inflation in the thoracic epidural space prevents the development of neurogenic pulmonary edema, most likely due to the better adaptation of the organism to acute circulatory changes (rapid elevation of systemic blood pressure accompanied by profound heart rate reduction) during the longer balloon inflation period. It should be noted that spinal cord transection at the same level did not cause neurogenic pulmonary edema.

The pathological potential of glial cells was recognized already by Rudolf Virchow, Santiago Ramon y Cajal and Pio Del Rio-Ortega. Many functions and roles performed by astroglia in the healthy brain determine their involvement in brain diseases; as indeed any kind of brain insult does affect astrocytes, and their performance in pathological conditions, to a very large extent, determines the survival of the brain parenchyma, the degree of damage and neurological defect. Astrocytes being in general responsible for overall brain homeostasis are involved in virtually every form of brain pathology. Here we provide an overview of recent developments in identifying the role and mechanisms of the pathological potential of astroglia.

Spinal cord injury results in a permanent neurological deficit due to tissue damage. Such a lesion is a barrier for "communication" between the brain and peripheral tissues, effectors as well as receptors. One of the primary goals of tissue engineering is to bridge the spinal cord injury and re-establish the damaged connections. Hydrogels are biocompatible implants used in spinal cord injury repair. They can create a permissive environment and bridge the lesion cavities by providing a scaffold for the regeneration of neurons and their axons, glia and other tissue elements. The advantage of using artificial materials is the possibility to modify their physical and chemical properties in order to develop the best implant suitable for spinal cord injury repair. As a result, several types of hydrogels have been tested in experimental studies so far. We review our work that has been done during the last 5 years with various types of hydrogels and their applications in experimental spinal cord injury repair.
